AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

The cognitive effects of creatine supplementation in older adults and preclinical models of dementia: A scoping review.

BACKGROUND & OBJECTIVES: Dementia represents a growing global health crisis, yet disease-modifying therapies remain limited. Impaired brain energy metabolism, neuroinflammation, and synaptic dysfunction are some shared pathological features across dementia subtypes that may be amenable to metabolic intervention. Creatine supplementation, through its role in the phosphocreatine/ATP buffering system and proposed neuroprotective properties, has attracted growing scientific and public interest as a candidate intervention for dementia. This scoping review maps the existing evidence on creatine supp

Developmental variation in second-to-fourth digit ratio (2D:4D) in children with cerebral palsy: an exploratory morphometric analysis.

OBJECTIVE: Cerebral palsy (CP) is an etiologically heterogeneous neurodevelopmental condition arising from disturbances of the developing brain during prenatal, perinatal, or early postnatal life. The second-to-fourth digit ratio (2D:4D) has been proposed as an imperfect indirect correlate of first-trimester prenatal sex-steroid exposure. This study aimed to compare 2D:4D ratios between children with CP and healthy controls and to examine whether digit ratio was associated with functional and clinical severity in children with CP. METHODS: This case-control, cross-sectional study included 75

Beyond attendance: participation and relational work in planned group recreational activities in a long-term care village setting.

BACKGROUND AND OBJECTIVES: Person-centered care (PCC) has shaped dementia care reforms and informed innovative long-term care models such as care villages. Recreational activities are often seen as a site where PCC is enacted, yet little empirical work has examined how participation in structured activities is enabled, mediated, and constrained through everyday social, relational, and organizational dynamics. This study examines how participation in structured group activities was produced in a care village. RESEARCH DESIGN AND METHODS: An ethnographic study was conducted in Canada's first pu

Parkinsonism and psychopathology severity in real-world outpatients with schizophrenia: A cross-sectional PRISM study.

Extrapyramidal symptoms (EPS) have been described as intrinsic to schizophrenia spectrum disorders (SSD), beyond medication-induced effects. Among EPS, parkinsonism is the most prevalent subtype and has been linked to poorer clinical and functional outcomes. We aimed to examine the relationships between parkinsonism and psychopathology and social functioning, as well as its demographic and pharmacological correlates, in a real-world sample of outpatients with schizophrenia receiving stable antipsychotic medication. Data were drawn from the PRISM project, an EU-funded, multicentre, cross-sectio

Posttherapy [ 18 F]FDOPA PET Findings in an Anaplastic Pleomorphic Xanthoastrocytoma With a BRAF V600E Mutation.

Anaplastic pleomorphic xanthoastrocytoma (APXA) is a rare pediatric high-grade glioma (pHGG). We report a 10-year-old boy with a left temporal APXA harboring a BRAF V600E mutation, who presented with generalized tonic-clonic seizures 3-4 times weekly, treated with combined BRAF and MEK inhibition. This case illustrates the utility of [ 18 F]FDOPA PET in monitoring targeted therapy, providing metabolic information that preceded and complemented conventional imaging.

Use of Pointwise Encoding Time Reduction with Radial Acquisition Magnetic Resonance Angiography for Coil Embolization.

INTRODUCTION: It is imperative to perform imaging evaluation following endovascular coil treatment for cerebral aneurysms. Noninvasive vascular assessment using Magnetic Resonance Angiography (MRA), specifically Time-of-Flight (TOF) MRA, is commonly performed. Thus, this study assessed the diagnostic performance of Pointwise Encoding Time Reduction with Radial Acquisition (PETRA) MRA in patients who underwent simple coil embolization. METHODS: Occlusion status, overall image quality, and treated-site image quality were compared across Digital Subtraction Angiography (DSA), TOF MRA, and PETRA

Improved stability and interpretability of motor modules computed with an autoencoder.

Motor module analysis is an important tool in the study of movement, particularly in people with impaired neural control. The most common method for computing motor modules is non-negative matrix factorization (NMF), which identifies a matrix of motor modules and their corresponding time-series activity from electromyography data. NMF has several limitations, including dependence of the muscle weightings on the number of modules selected. Approaches for selecting the number of modules vary between studies, making it difficult to compare and reproduce results. Some metrics of motor control comp

Counterintuitive: reducing regression-to-the-mean worsens efficiency in simulated clinical trials.

OBJECTIVE: Our lab previously showed in simulation that excluding the formal baseline from the eligibility window would reduce regression-to-the-mean (RTM), lower apparent placebo response, and improve antiseizure medication (ASM) trial efficiency. A recent software re-review revealed a reversal on the final point: excluding baseline worsened trial efficiency. We investigated this paradoxical finding further by comparing two otherwise identical designs that differed only in whether baseline contributed to eligibility. METHODS: The calibrated seizure-diary simulator CHOCOLATES generated a shar

Advancement in therapeutic application of quantum dots in amyotrophic lateral sclerosis: current opportunities and challenges.

Despite advancements in healthcare settings in developed countries, the early detection and higher mortality rate associated with amyotrophic lateral sclerosis (ALS), a fatal motor neuronal disorder, remain challenging. Recently, quantum dots (QDs) have emerged as a promising nanocarrier in the prognosis and treatment of ALS owing to their unique multifunctional properties. QDs, through their photoluminescence properties upon excitation, can facilitate the identification and real-time monitoring of disease biomarkers. They also act as a nanocarrier for the targeted delivery of therapeutics, av

Valosin-containing protein counteracts ATP-driven dissolution of FUS condensates through its ATPase activity in vitro.

Fused in sarcoma (FUS) forms phase-separated condensates implicated in amyotrophic lateral sclerosis (ALS). Although millimolar ATP concentrations paradoxically dissolve FUS condensates through hydrotropic activity, condensates nevertheless persist in cells, suggesting active regulatory mechanisms. Here, using a reconstituted system, we show that the AAA+ATPase valosin-containing protein (VCP) counteracts ATP-driven dissolution of FUS condensates. VCP preserved both wild-type and ALS-linked P525L condensates under high ATP conditions, and this protection required catalytic ATPase activity rath

The Theoretical Evolution of Social Memory and the Core Contribution of the Medial Prefrontal Cortex.

Social interaction is a process in which two or more conspecific individual information and engage with one another, mutually influencing each other's behavior. Within this framework, social cognition and social memory constitute fundamental components that enable adaptive social behaviors. Deficits in social cognition and memory have been implicated in various psychiatric disorders since social memory itself represents a core expression of episodic memory. In this review, we summarize the theoretical evolution of understanding the social cognitive memory from the grandmother cell hypothesis t

How robust are secondary dose algorithms for Patient-Specific quality assurance in modulated stereotactic Radiosurgery?

PURPOSE: Accurate dose verification is essential in stereotactic radiosurgery (SRS), where small fields and steep dose gradients may amplify uncertainties, especially with complex techniques like multitarget single-isocenter VMAT. This technical note evaluated the robustness of two secondary dose calculation algorithms for SRS patient-specific quality assurance (PSQA). METHODS: RadCalc Collapsed Cone Convolution Superposition (RC-CCCS) and Monte Carlo (RC-MC) were commissioned on beam-matched VersaHD linacs. Work frequency and subsequent health and well-being among older adults in Japan: An outcome-wide study.

BACKGROUND: Working in later life has been linked to health and well-being, yet how work frequency relates to these outcomes remains uncertain. We examined associations between work frequency and subsequent multiple domains of health and well-being among older adults. METHODS: We used two datasets from the Japan Gerontological Evaluation Study (JAGES). Dataset 1 used three-wave panel data (2016, 2019, 2022; n = 41,758) for 42 self-reported outcomes. Dataset 2 linked 2016-2019 panel data to municipal long-term care insurance records through 2022 (n = 56,153), capturing mortality, dementia, and
