Integrating genomics and endocrinology to investigate prolactin dysregulation in female African Savannah elephants housed in north American zoos.

The sustainability of the captive African savannah elephant (Loxodonta africana) population in the United States has reached a critical point and is no longer viable under current conditions. A major contributing factor is the high proportion of reproductively aged female African savannah elephants that are not cycling due to prolactin dysregulation (hyperprolactinemia). Because prolactin secretion plays a key role in normal ovarian function in mammals, its dysregulation can negatively affect both individual health and overall reproductive success. No treatments for this condition are currentl

Analysis of the Pupillary Light Reflex Response Against Light Stimuli for Autism Screening in Children.

PURPOSE: Traditional diagnostic methods for autism spectrum disorder (ASD) predominantly rely on behavioral scales, which can be less effective among young children. This delay in diagnosis impedes timely interventions crucial for improving developmental outcomes. Pupillometry, which measures the pupillary light reflex (PLR), serves as a promising technique to address these challenges. It has the potential to provide biological insights into the central nervous system's functioning, indirectly revealing potential ASD risk. METHODS: This pilot proof-of-concept study assessed the PLR in 30 chil

Transcranial Direct Current Stimulation and Cranial Electrotherapy Stimulation in Athletes: Optimizing Athletic Performance and Sleep Quality Through Neuromodulation.

Neuromodulation is a procedure relying on the delivery of specific stimuli to selected sites of the nervous system and hereby influencing its activity. Even though the underlying neurobiological mechanisms are not yet fully elucidated and the research is still ongoing, neuromodulation is already being used as an intervention for various mental conditions and insomnia, or it may even improve the physical performance of athletes. This narrative review summarizes current evidence on the effects of specific neuromodulation techniques on performance and sleep quality in athletes, focusing on crania

Structural network alterations and peripheral immune-inflammatory dysregulation define distinct subtypes of major depressive disorder with prognostic implications.

BACKGROUND: Immune-inflammatory dysregulation and cortical structural vulnerability have been increasingly implicated in major depressive disorder (MDD). However, how they interact with each other, and whether immune heterogeneity in MDD is systematically organized by structural networks, remain unclear. METHODS: Genome-wide association studies data of MDD, gray matter volume (GMV) phenotypes, and 19 immune-related traits were utilized to assess causal relationships. A prospective cohort comprising 122 patients with MDD and 66 healthy controls underwent structural magnetic resonance imaging a

prickle-mediated epilepsy in flies is a brain-derived neurodevelopmental defect.

Damaging DNA variants in PRICKLE genes cause epilepsy from flies to humans. While classic work, primarily in flies, has demonstrated that PRICKLE proteins work with other planar cell polarity proteins to polarize epithelial sheets, the function of PRICKLE proteins in the nervous system is still unclear and may play distinct roles in the nervous system of different organisms. We thus sought to compare and contrast Drosophila prickle with PRICKLE genes in vertebrate systems in order to identify unifying roles across species. First, we find that similar to expression of the mouse Prickles, fly pr

Association of Left Ventricular Outflow Tract Obstruction with Left Atrial Remodeling and Dysfunction in Hypertrophic Cardiomyopathy: A Three-Dimensional Echocardiography and Left Atrial Strain Study.

Adolescents with major depressive disorder (MDD) and non-suicidal self-injury (NSSI) face serious challenges to their mental and physical health. The development of this condition is influenced by genetic, environmental, and brain development factors. In this study, we used the Morphometric Inverse Divergence Gradient Network (MIND-Gd) for the first time to explore brain structural network abnormalities and their transcriptional mechanisms in adolescents with MDD and NSSI. A total of 241 adolescents with MDD participated in this study. Among them, 134 had NSSI, while 107 did not. Using high-re

Ultrasound-induced luminescence imaging for deep tissues.

Ultrasound-induced luminescence imaging enables non-invasive and space-constrained luminescent probe activation, thereby overcoming the limited penetration and high background of traditional light-excited imaging. In this review, we summarize recent advances in the area, focusing on three major paradigms: instant ultrasound-induced luminescence, sonoafterglow luminescence, and Off-On switching luminescence, with detailed explanations of their mechanisms. We also explore various strategies to improve luminescent and biological performance. Additionally, we offer a comprehensive overview of biom

Neighborhood disadvantages and longitudinal progression of multimorbidity among adult survivors of childhood cancer: a single-center St. Jude Lifetime Cohort Study in the U.S.

BACKGROUND: The impact of cancer therapy on chronic health condition (CHC) burden in childhood cancer survivors is well-established; however, the role of neighborhood-level social determinants of health (SDOH) remains underexplored. METHODS: In this single-center cohort study in the U.S., we analyzed 2508 survivors enrolled in the St. Jude Lifetime Cohort Study between 9/13/2007 and 4/20/2020, who were ≥5 years from diagnosis, ≥18 years of age, and had ≥2 clinical assessments. SDOH encompassed social vulnerability (Social Vulnerability Index) and environmental/healthcare access vulnerabilitie

Performance in play: stimulant exposure and broader substance-use vulnerability in mass-participation youth esports.

Youth esports now extends far beyond professional competition, encompassing ranked online play, school clubs, amateur events, livestreaming, and digitally mediated peer communities. These settings expose adolescents to energy drinks and other stimulant-containing products in contexts where alertness, concentration, and prolonged play are valued. Energy drinks merit particular attention because their prominence in gaming culture combines caffeine exposure with, for many products, substantial added sugar. Potential consequences include disrupted sleep, acute stimulant effects, and adverse nutrit

[Increased nervous excitability, anxiety, and sleep disorders in childhood - the role of multicomponent low-dose medications].

Anxiety, emotional excitability, and sleep disturbances are common in pediatric populations, often emerging during stressful periods such as chronic family conflict, parental divorce, initial socialization in kindergarten, and the beginning of formal schooling. These conditions are frequently linked to autonomic nervous system (ANS) dysfunction. During such periods, nervous system instability impairs stress adaptation and may lead to neurotic reactions. Chronic stress and anxiety can trigger neuroinflammatory processes, which further intensify anxiety and ANS dysfunction. Clinically, these mec

Nanoplasmonic Aptasensing Enables Real-Time Optical Monitoring of Neurotransmitters in Living Brainstem Organoids.

Disruption of dopamine signaling is central to Parkinson's disease (PD) pathology and a primary target of symptomatic therapy. However, direct measurement of dopamine dynamics in human neural tissue with high sensitivity, temporal resolution, and compatibility with three-dimensional models remains technically challenging. Here, an organoid-on-a-chip platform that integrates a cavity-coupled nanoplasmonic aptasensor with human brainstem organoids (hBSOs) and microfluidic control is employed for label-free monitoring of the extracellular dopamine response. The plasmonic aptasensor achieves a lim

Illuminating the Disease Landscape: Human Oxidoreductase-Targeted Imaging and Quantification Agents for Precision Imaging and Activity Determination.

Electron-specific reduction reactions afforded by quinone oxidoreductases have long been recognized as essential for safeguarding healthy mammalian cells against oxidative stress induced by environmental insults. The presence of reactive oxygen species (ROS) initiates the upregulation of intracellular redox enzyme activities used to battle the effects of ROS in healthy cells. At the same time, diseased human cells hijack the pathways of certain oxidoreductases, such as the two-electron-specific human NAD(P)H:quinone oxidoreductase isozyme 1 (hNQO1), and use the overexpressed enzymes as standal

ALOX15-Inhibitory Peptides Derived from Coffee Bean Protein Hydrolysates Confer Neuroprotection against Lipid Peroxidation.

Coffee beans are widely consumed worldwide and are known for their central nervous system-stimulating effects, mainly attributed to caffeine and other small molecules. However, the neuroprotective potential of coffee bean-derived peptides remains largely unexplored. In this study, coffee bean protein hydrolysates (CBPH) exhibited protective effects against RSL3- and 6-OHDA-induced neuronal injury. Virtual screening and molecular docking targeting 15-lipoxygenase (ALOX15), a key enzyme involved in oxidative neuronal damage, identified eight candidate inhibitory peptides. Among them, FAR and FPS

Pan-neurofascinopathies: a subgroup that mimic other neuroimmune-mediated diseases.

Pan-neurofascinopathies represent a distinct subset of inflammatory neuropathies characterized by autoantibodies that target multiple neurofascin (NF) isoforms, which are essential for nodal and paranodal architecture in peripheral nerves. These antibodies are associated with a broad clinical spectrum, often mimicking Guillain-Barré syndrome (GBS) or chronic inflammatory demyelinating polyradiculoneuropathy (CIDP), and exhibit significant variability depending on the immunoglobulin G (IgG) subclass involved. The IgG1 and 3 subclasses correlate with acute, fulminant presentations and complement

Antagonizing central GABAA and adenosine receptors reverses the autoresuscitation defects caused by reduced brainstem serotonin.

Sudden Infant Death Syndrome (SIDS) remains a leading cause of infant death, and its incidence is increasing. SIDS infants harbor numerous defects within the brainstem serotonin (5-hydroxytryptamine, 5-HT) system. There is also evidence that in hypoxic conditions SIDS infants do not successfully autoresuscitate, the integrated response to severe hypoxia necessary to reperfuse the brain, restore eupnea, and promote arousal from hypoxic coma. In animals, a loss of 5-HT or 5-HT2A receptor activity compromises autoresuscitation. Caffeine, via undefined mechanisms, rescues the autoresuscitation of

Pharmacological Activation of Autophagy Ameliorates Neuromuscular-Associated Defects and Extends Lifespan in a Caenorhabditis elegans Model of Spinal Muscular Atrophy.

Spinal muscular atrophy (SMA) is a severe neuromuscular disorder caused by homozygous deletions or mutations in the SMN1 gene, resulting in reduced levels of the ubiquitously expressed survival motor neuron protein and progressive degeneration of lower motor neurons. Although autophagy dysregulation has been implicated in SMA pathology, the therapeutic benefit of modulating this pathway remains controversial. Here, we screened autophagy modulators in a previously characterized Caenorhabditis elegans SMA model. We identified that pharmacological activation of autophagy, but not inhibition, sign

Tau isoform mapping across tissues and biofluids reveals a signature distinguishing Alzheimer's disease from amyotrophic lateral sclerosis.

Blood phosphorylated tau 217 (p-tau217) accurately detects Alzheimer's disease (AD) pathology but is also elevated in amyotrophic lateral sclerosis (ALS), likely reflecting peripheral (big) tau released from muscle. Assays selectively measuring brain-derived (BD-) tau and BD-p-tau217 are therefore needed. We analysed serum BD-tau and p-tau217 in ALS (n = 139), AD (n = 101) and controls (n = 75) using direct immunoassays and after BD-Tau immunoprecipitation (BD-Tau-IP), and tau species in brain (n = 16), spinal cord (n = 11) and muscle (n = 10) by targeted mass spectrometry. Clinical Characteristics, 90-Day Functional Outcomes, Antimicrobial Susceptibility, and Exploratory Genomic Analysis of Invasive Human Streptococcus suis Infection: A Multicenter Retrospective Study in China.

BACKGROUND: Streptococcus suis is a zoonotic pathogen; survivors of invasive infection may have persistent auditory or vestibular sequelae. To address limited multicenter evidence from China, we characterized 90-day outcomes, antimicrobial susceptibility, and exploratory genomic findings. METHODS: We retrospectively studied 34 patients with invasive S. suis infection at eight hospitals in four Chinese provinces (2017-2025). Unfavorable outcome was defined as death within 90 days or persistent sequelae at day 90. Minimum inhibitory concentrations were reinterpreted using CLSI viridans-group st

Spinorphin Ameliorates Nitroglycerin-Induced Migraine-Like Responses and Trigeminovascular Neuroinflammation Potentially Through Opioid Receptor-Mediated Mechanisms.

Enhancement of endogenous opioid signaling through inhibition of enkephalin degradation has emerged as a potential strategy for pain control, yet its role in migraine remains unclear. This study investigated whether spinorphin, an endogenous inhibitor of multiple enkephalinases, attenuates trigeminovascular activation and migraine-like responses in experimental migraine. Migraine-like pain was induced in male Wistar rats by nitroglycerin. The effects of spinorphin were evaluated using periorbital and plantar von Frey tests, followed by assessment of c-Fos levels in the trigeminal nucleus cauda

Developing a cps-genotyping scheme for the emerging zoonotic pathogen Streptococcus parasuis through genetic analysis of capsular polysaccharide synthesis gene loci.

BACKGROUND: Streptococcus parasuis is an emerging zoonotic pathogen responsible for pneumonia, meningitis, peritonitis, pleuritis, nephritis, cellulitis, cholecystitis, sepsis, and arthritis in humans. The continuous increase in reported human cases of S. parasuis infection indicates its growing threat to public health and highlights the urgent need to monitor its epidemiology. The lack of high-throughput typing tools has seriously impeded the epidemiological surveillance of S. parasuis. Serotyping based on antigenic differences in capsular polysaccharides (CPSs) synthesized by cps loci is wid
