Purine metabolism disruption links pathology and cognitive decline in Alzheimer's disease.

Purine metabolism dysregulation has been implicated in Alzheimer's disease (AD), but its temporal dynamics and mechanistic links to core pathologies remain unclear. This translational study integrated a longitudinal preclinical AD mouse model and clinical cohort investigations to define the role of purine metabolism in AD progression. In APPswe/PSEN1dE9 (APP/PS1) mice, serum-untargeted metabolomics identified purine metabolism as the most notably altered pathway, with metabolite levels (e.g., adenine, adenosine) shifting dynamically across disease stages alongside increasing amyloid-β (Aβ) bur

Artificial intelligence for facial soft-tissue landmark localization, measurement, and classification in orthodontics: A systematic review.

Microglial polarization critically influences neuroinflammation and recovery after ischemic stroke. The role of C-type lectin domain family 7 member A (CLEC7A), a potential regulator in this process, remains unclear. Integrated bioinformatics analysis of Gene Expression Omnibus datasets identified CLEC7A as a hub gene. Its functional role was investigated using in vitro oxygen-glucose deprivation (OGD) models in BV2 microglia/HT22 neuron co-cultures and in vivo rat middle cerebral artery occlusion (MCAO) models. CLEC7A was knocked down in vitro and in vivo. Functional outcomes were assessed vi

Combined PET imaging of tumor hypoxia and proliferation in Glioblastoma using a single injection of 18F-FMISO and 18F-FLT: Association with sites of subsequent recurrence.

BACKGROUND: Glioblastoma (GBM) is characterized by intra-tumoral heterogeneity, rapid proliferation, hypoxia-driven radio-resistance, and early recurrence. Gadolinium-Enhanced MRI, the standard of care imaging modality for surgical and radiotherapy planning, lacks specificity for viable tumor burden, particularly in non-enhancing regions. PURPOSE: This pilot study aimed to evaluate whether preoperative dual-tracer PET using a combined dose of 18FMISO (hypoxia) and 18FLT (proliferation) improves tumor delineation and predicts subsequent local recurrence compared with MRI. METHODS: Seven patie

Outdoor extreme heat-related dementia risk and the role of socioeconomic disparities: evidence from 20 years of data on US older adults.

INTRODUCTION: Extreme heat is increasingly recognized as a significant threat to brain health. However, human evidence from large, longitudinal studies remains limited. We examine whether outdoor heat exposure is associated with incident dementia and whether socioeconomic factors moderate this risk. METHODS: Leveraging data from the 2000 to 2020 Health and Retirement Study (N = 28,512), we assigned census-tract-level 5-year exposure to the number of "Extreme Caution" (90°F to 102°F [32.2°C to 39.3°C]) and "Danger+" (≥103°F [39.4°C]) heat days, adjusting for moves. [Tourettic obsessive-compulsive disorder: A clinical-phenomenological dilemma at the intersection of neurology and psychiatry].

In recent years, the notion of Tourettic obsessive-compulsive disorder (Tourettic OCD), viewed as a distinctive endophenotype of Gilles de la Tourette syndrome, has garnered increasing attention. Despite this growing interest, clinical observations of affected patients remain sparse in the literature. This article presents the clinical case of a teenager diagnosed with Gilles de la Tourette syndrome, whose condition evolved to domination of obsessive-compulsive and behavioral disorders over the previously dominant multiple motor and vocal tics. The question of the nosological independence of T

[Pseudo-remission in childhood absence epilepsy. Objective assessment of seizure control and the role of ethosuximide].

Absence epilepsy is a common form of pseudo-remission, in which patients and observers may incorrectly believe seizures have ceased while paroxysms persist in subtle form. Three primary factors contribute to this phenomenon: the brief duration (typically less than 10 seconds) and indistinct semiology of typical absences; the limited reliability of parental observation (sensitivity up to 43%); and the occurrence of generalized spike-wave discharges without significant clinical manifestations, which are nonetheless associated with fluctuations in wakefulness. Verification of remission requires a

[Clinical heterogeneity and prediction complexity associated with nucleotide sequence abnormalities in the SCN2A gene].

Numerous variants of the SCN2A gene have been identified, including missense mutations, deletions, insertions, frameshifting, nonsense mutations, and splice site mutations. The emergence of severe phenotypes is generally associated with de novo and missense variants. The clinical manifestations linked to SCN2A abnormalities exhibit considerable diversity, including varying severities of epileptic episodes with onset in neonatal or subsequent periods, autism spectrum disorders, schizophrenia (including catatonic manifestations), and episodic ataxia. With rare exceptions, all identified phenotyp

Clinical Diagnostic and Prognostic Evaluation of miR-136-5p in Children with Severe Pneumonia: A Case-Control and Cohort Study.

Severe pneumonia is a leading cause of childhood morbidity and mortality. MicroRNA‑136‑5p (miR‑136‑5p) plays critical roles in inflammatory diseases, yet its expression, clinical relevance, and regulatory mechanisms in children with severe pneumonia remain unclear. This study recruited 114 children with severe pneumonia and 106 healthy controls; peripheral blood miR‑136‑5p and Argonaute 1 (AGO1) were detected by quantitative real‑time polymerase chain reaction (qRT‑PCR). Receiver operating characteristic (ROC) curves, Kaplan‑Meier analysis, and multivariate Cox regression were used to evaluate

Inherited and early-life vulnerability in functional neurological disorder and functional somatic symptoms: A retrospective cohort and case-control study from a large electronic health record database.

BACKGROUND: Biological factors affecting motor, sensory, and integrative processing may predispose to functional neurological disorder (FND) and functional somatic symptoms. While early-life influences have traditionally been conceptualized within psychological frameworks, the contribution of early-life biological vulnerabilities to symptom expression remains unclear. METHODS: Using TriNetX, a large electronic health records database, we assessed proportions of inherited, congenital, perinatal, and neurodevelopmental diagnoses, capturing a range of early-life conditions. We compared 188,868 i

Multivariate GWAS Analysis Reveals a Common Genetic Factor Linking Dry Eye Disease to Gastrointestinal Comorbidities.

PURPOSE: Dry eye disease (DED) frequently overlaps clinically with irritable bowel syndrome, gastroesophageal reflux, and peptic ulcer, yet their shared genetic architecture and pathogenic pathways remain elusive. METHODS: Univariate genome-wide association studies (GWAS) were conducted for DED and four gastrointestinal diseases. Genomic structural equation modeling defined a shared genetic factor mvDED-GI. FUMA annotated peak variants for subsequent gene-set, tissue, and cell enrichment. Transcriptome-wide association study (TWAS) plus fine-mapping prioritized causal genes, and multi-layer e

A Cell Culture Insert-Based Protocol for Constructing a Simplified Multicellular In Vitro Blood-Tumor Barrier Model of Breast Cancer Brain Metastasis.

Effective treatments for breast cancer brain metastases (BCBM) remain limited, largely because the blood-tumor barrier (BTB) restricts the entry of many chemotherapeutic and targeted agents into metastatic lesions within the central nervous system. To model selected features of this complex microenvironment in vitro, this study presents a standardized protocol for constructing a simplified multicellular three-dimensional BTB model. The method comprises three core steps. First, a blood-brain barrier (BBB) compartment is constructed using cell culture inserts with a 0.4 μm pore size. Human brain

Vascular Cell Adhesion Molecule 1 Is Associated With Immune Remodeling, Glioma Progression, and Tumor Cell Survival in U87 and U251 Cells.

The immunosuppressive microenvironment of glioma remains a major barrier to effective treatment. Vascular cell adhesion molecule 1 (VCAM1) regulates leukocyte adhesion and inflammatory signaling, but its relevance to glioma remains unresolved. Clinical and RNA sequencing (RNA-seq) data from The Cancer Genome Atlas (TCGA), gene co-expression information from LinkOmics, and protein expression data from the Human Protein Atlas (HPA) were integrated. Kaplan-Meier analysis, Cox regression, receiver operating characteristic (ROC) analysis, and nomogram construction were used to evaluate prognostic v

Pre-surgery statin use and outcomes in operated chronic subdural haematomas: a nationwide cohort study.

BACKGROUND: Chronic subdural haematoma (cSDH) is common in older adults, and medication exposures may influence postoperative outcomes. We assessed whether pre-surgery statin use is associated with short- and long-term outcomes. METHODS: We conducted a nationwide registry study of adults undergoing surgical evacuation of traumatic or non-traumatic cSDH (6/2004-12/2018) in Finnish university hospitals. Pre-surgery statin use was defined from national prescription records using a pill-counting approach. The primary outcome was 30-day all-cause mortality. Secondary outcomes were 30-day reoperati

Vascular endothelial growth factor A induces proliferation of adult neural progenitor cells in non-neurogenic periventricular regions of the adult mouse hindbrain.

BACKGROUND: In addition to the well-established neurogenic niche of the ventricular-subventricular zone (V-SVZ) of the lateral ventricles, neural stem/progenitor cells (NPCs) have been identified in the periventricular regions (PVRs) throughout the adult mouse ventricular system. The potential role of these NPCs in neuroplasticity or neurorestoration, however, remains largely unexplored. OBJECTIVE: Given that vascular endothelial growth factor A (VEGF-A) has been shown to promote stem cell proliferation in the V-SVZ, this study aimed to investigate VEGF-A effects on NPC proliferation in mid-

Description of Corynebacterium nasorum sp. nov. and Corynebacterium hallucis sp. nov. isolated from human nasal passages and skin.

Strains of two novel Corynebacterium species were cultured from samples of human nostrils and skin collected in the USA and Botswana. These strains demonstrated growth on Columbia Colistin-Nalidixic Acid agar with 5% sheep blood and in liquid media (brain-heart infusion and tryptic soy broth) supplemented with Tween 80, a source of the fatty acid oleic acid. Cells were Gram-positive, non-spore-forming, non-motile bacilli that showed catalase but not oxidase activity. Major fatty acids in both of these species were C18 : 1 ω9c (oleic acid), C16 : 0 (palmitic acid) and C18 : 0 (stearic acid). An

TSPO-PET binding in vivo reflects increased phagocytic microglia at post mortem in people with frontotemporal dementia.

Brain inflammation is a key feature of frontotemporal dementia (FTD). Positron emission tomography (PET) of the 18 kDa translocator protein (TSPO) is widely used as an in vivo proxy for neuroinflammation, but whether the elevated signal reflects microglial, astrocytic or vascular pathology remains controversial, and this has not previously been resolved in FTD using matched ante mortem imaging and post-mortem tissue. We paired ante mortem [11C]PK11195 TSPO-PET with quantitative post-mortem neuropathology in 10 individuals with FTD (5 FTLD-tau, 5 FTLD-TDP) and 5 age- and sex-matched controls, c
