AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

Conclusions and Therapeutic Implications of AA.

AA is essential for several tissues in the body, as they are involved in many physiological and pathological processes. Its regulatory role in inflammation, immune response regulation, cell cycle, and membrane structural organization and its ability to control the expression and function(s) of several receptors and convey messages to and from the nucleus are of particular significance. Fine-tuning the metabolism of AA may lead to the development of prevention and management strategies for several diseases, including but not limited to sepsis, inflammatory bowel disease, diabetes mellitus, canc

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

The spliceosome component SNRPC promotes glioma progression by sustaining mitochondrial function and TNFAIP2 signaling.

Human glioma is a devastating primary brain tumor with a dismal prognosis, necessitating the identification of novel therapeutic targets. The small nuclear ribonucleoprotein C (SNRPC), a core spliceosome component, is implicated in cancer, but its role in glioma remains unexplored. This study aimed to delineate the expression, function, and underlying mechanisms of SNRPC in human glioma. The bioinformatic analysis revealed that SNRPC is significantly upregulated in glioma tissues, with expression levels correlating strongly with higher tumor grade, aggressive molecular subtypes (IDH wild-type,

Repurposing triamterene as chloride intracellular channel 1 inhibitor via ligand-based approach for glioblastoma.

Identification of effective therapies for glioblastoma is an unmet need in clinical oncology. The antidiabetic drug metformin has been recommended as a repurposed treatment for glioblastoma, mainly acting as a blocker of chloride intracellular channel 1 (CLIC1), a key regulator of glioblastoma stem cell (GSC) proliferation. However, metformin's complex pharmacokinetics and low antitumor potency have limited its clinical translation. Using a ligand-based approach and computational studies, we identified the diuretic triamterene as a previously unrecognized CLIC1 blocker. Whole-cell electrophysi

[Impact of epilepsy-related stigma on employability: A review of the evidence for primary care practice].

INTRODUCTION: Epilepsy is a common chronic neurological disorder that entails a substantial psychosocial and occupational impact mediated by stigma. Available studies show that, compared with the general population, people with epilepsy have higher rates of unemployment, work disability, absenteeism and presenteeism, as well as poorer health-related quality of life. The aim of this study is to analyze the impact of stigma on the employment situation of people with epilepsy and its implications for primary care and occupational medicine. METHOD: A structured narrative review with critical appr

Enhanced Glioblastoma Detection Through 13C Hyperpolarized MRI and Two-Dimensional Statistics.

PURPOSE: To develop a method for statistically significant tumor delineation. Glioblastoma (GBM) remains resistant to current therapeutic strategies and is unequivocally associated with a dismal prognosis. Hyperpolarized 13C MRI (hpMRI) provides unique insights into tissue metabolism, enabling outlining of tumor boundaries. Statistical confidence of this outline may help guide surgical resection, inform therapeutic decisions, and evaluate treatment efficacy. METHODS: The new method was applied to time-resolved hyperpolarized [1-13C]pyruvate data from a previous study in a rat glioma model as

Calcaneal Lengthening Osteotomy in Cerebral Palsy: Does Age at Intervention Influence Outcomes?

BACKGROUND: The optimal age for calcaneal lengthening osteotomy in children with cerebral palsy remains controversial. Younger age has been suggested as a risk factor for recurrence or overcorrection, but long-term clinical data are limited. The primary purpose of this study was to determine whether age at surgery influences long-term radiographic maintenance of correction and clinical outcomes. METHODS: A retrospective review was performed of 102 patients (155 feet) with cerebral palsy who underwent lateral column lengthening as part of a foot reconstruction procedure between 1990 and 2020 w

Design, synthesis and anti-glioblastoma multiforme activity of N-methoxy-2-(pyrimidin-4-ylamino)benzamide derivatives as focal adhesion kinase inhibitors.

Glioblastoma multiforme (GBM) remains a lethal brain cancer with poor prognosis, largely due to the limitations of current therapies. Focal adhesion kinase (FAK) is a promising therapeutic target, given its central role in driving GBM progression, invasion, and microenvironment adaptation. While several FAK inhibitors, such as TAE-226, have entered clinical trials, none have achieved approval, underscoring the need for novel, more effective agents. Through a structure-based molecular hybridization strategy that integrated key pharmacophores from TAE-226 and the clinical candidate GSK2256098, a

Health literacy among patients with subarachnoid hemorrhage.

INTRODUCTION: Patients with subarachnoid hemorrhage (SAH) often experience cognitive deficits and require extensive and complex medical care. However, there is minimal literature investigating how SAH impacts health literacy and how this may affect care. Accordingly, we aim to leverage the NIH's All of Us Research Program to assess how SAH relates to health literacy among a large and diverse patient cohort. METHODS: Using the NIH's All of Us Research Program, respondents with SAH were matched to those without SAH via nearest-neighbor propensity score matching. Only those with SAH prior to sur

Structure-Aware Attention Prototype Network for Cross-Modality Few-Shot Brain Tumor Segmentation.

Multi-modal magnetic resonance imaging (MRI) plays a crucial role in brain tumor diagnosis. However, the substantial physiological sensitivity discrepancies across imaging modalities create a severe domain gap that challenges current cross-modality segmentation methods. Many unsupervised domain adaptation (UDA) approaches reduce this gap through image style translation or distribution alignment, which have shown promising results but may struggle to preserve modality-specific anatomical and pathological information. In contrast, few-shot segmentation (FSS) provides a promising alternative para

Predicting postoperative hospice enrollment following stereotactic brain biopsy: a retrospective study and risk score development.

OBJECTIVE: Stereotactic brain biopsy (SBB) is a key diagnostic tool in neuro-oncology. Due to the poor prognosis for many tumors, a significant subset of patients transition to hospice care shortly after the procedure. Current tools inadequately predict which patients are less likely to benefit from SBB. This study aimed to define preoperative factors that limit enrollment in adjuvant treatment in SBB candidates by characterizing patients who enrolled in hospice soon after the procedure. Understanding the safety profile and diagnostic utility of SBB in the context of hospice enrollment is esse
