Integrating genomics and endocrinology to investigate prolactin dysregulation in female African Savannah elephants housed in north American zoos.

The sustainability of the captive African savannah elephant (Loxodonta africana) population in the United States has reached a critical point and is no longer viable under current conditions. A major contributing factor is the high proportion of reproductively aged female African savannah elephants that are not cycling due to prolactin dysregulation (hyperprolactinemia). Because prolactin secretion plays a key role in normal ovarian function in mammals, its dysregulation can negatively affect both individual health and overall reproductive success. No treatments for this condition are currentl

Lack of cathepsin B increases severity in Niemann-Pick disease, type C1 mice.

Neurodegeneration is a hallmark of Niemann-Pick disease type C1 (NPC1). One component in the pathology of neurodegeneration is increased lysosomal membrane permeability, especially in microglia. Leakage of lysosomal proteases, such as cathepsin B, into the cytoplasm and ultimately into the interstitial space can lead to activation of apoptosis and neuroinflammation. In line with previous reports showing the involvement of cathepsin B in neurodegeneration and the benefit of inhibiting cathepsin B to prevent neurodegeneration, we tested whether disease severity would be reduced in NPC1 mutant mi

HSP27 Provides Stress-Adaptive Proteostasis Buffering in Glioblastoma and Is Therapeutically Vulnerable.

Glioblastoma, isocitrate dehydrogenase (IDH)-wildtype (GBM), remains one of the most aggressive primary brain tumors, and its proteostasis machinery represents a potential therapeutic target. Here we identify HSP27 as a key regulator-associated proteostasis in glioblastoma, IDH-wildtype, where it is linked to protein synthesis, folding, stress-granule dynamics and ubiquitin-mediated turnover. Ribo-seq showed HSP27 depletion reduced global protein synthesis, while TME labeling coupled to mass spectrometry revealed increased accumulation of unfolded proteins, indicating proteotoxic stress. Purine Metabolism Profiling Reveals Distinct Associations of Uric Acid, Xanthine, and Hypoxanthine with Neurological Severity and Functional Outcome After Acute Ischemic Stroke.

Purine metabolism and oxidative stress are increasingly recognized as important contributors to ischemic brain injury following acute ischemic stroke (AIS). Although uric acid has been extensively investigated because of its dual antioxidant and pro-oxidant properties, comparatively limited data exist regarding the prognostic relevance of upstream purine metabolites such as xanthine and hypoxanthine. The present study aimed to evaluate the association between serum uric acid, xanthine, and hypoxanthine concentrations and neurological severity and functional outcome following AIS. We performed

Feature-Based Machine-Learning Spike Marking in Signal and Source Space EEG: A Proof-of-Principle Case Study of Focal Epilepsy.

Accurate detection of interictal epileptiform discharges (IEDs) in electroencephalography (EEG) plays a crucial role in epilepsy diagnosis. Our work investigates the classification of IEDs using Artificial Neural Networks (ANNs) trained on EEG data represented in both signal and source space, in a proof-of-principle case study of focal epilepsy. Source waveforms were computed using a standard clinical source analysis procedure for the dipolar IED topographies, an equivalent current dipole model in a three-compartment head volume conductor model fitted using either a 1-parameter fixed-orientati

[Association of tumor metabolic parameters on amino acid PET/CT with EGFR expression].

BACKGROUND: According to the 2021 WHO classification, molecular-genetic profiling of CNS tumors is the standard for diagnosis. Beyond IDH mutation status, EGFR receptor alterations are significant factors associated with unfavorable prognosis and treatment resistance. Identifying correlations between genetic abnormalities and amino acid PET/CT parameters is important for discovering non-invasive surrogate markers of aggressive phenotype. OBJECTIVE: To analyze the relationship between EGFR / EGFRvIII gene expression and metabolic PET/CT parameters in patients with glial tumors. MATERIAL AND M

Direct probabilistic IMPT treatment planning with setup and range errors for neuro-oncological patients.

Objective. To show computational feasibility of a previously proposed probabilistic planning approach on a neuro-oncological patient group, we compared probabilistic plans with (automated) robust plans for one patient (group A) that could achieve sufficient clinical target coverage and for four patients (group B) where target coverage had to be compromised due to organ-at-risk (OAR) dose constraints.Approach. The probabilistic approach is percentile-based and uses the fact that a (dose) percentile can be approximated as a linear combination of its expected value and standard deviation. This al

Biomechanical assessment of repetitive brain injury in pedestrian collisions based on wavelet packet energy analysis.

PURPOSE: Existing brain injury criteria mainly focus on assessing injuries in a single collision and lack of assessment of injuries in real-world multiple collisions. METHODS: Based on the head kinematics signals of rats in repeated collisions and pedestrian accident data, the frequency domain characteristics of linear and rotational motion signals were analyzed by wavelet packet transform. Finally, the ROC curve was used to evaluate the effectiveness of injury index prediction. RESULTS: The results of animal experiments show that as the impact intensity increases, the energy in the frequenc

Preterm Risk Factors, Epigenetic Age, and Neurodevelopmental Outcomes.

IMPORTANCE: Studies examining biological factors associated with neurodevelopment in the preterm population are limited. It was previously demonstrated that epigenetic age acceleration was associated with better neurodevelopmental outcomes among children born preterm. It is unclear whether this association is modified by clinical and environmental preterm risk factors. OBJECTIVE: To examine whether the association between epigenetic aging and neurodevelopmental outcomes is modified by preterm brain injury and socioeconomic status. DESIGN, SETTING, AND PARTICIPANTS: This was a prospective coh

Interventions to reduce cognitive decline post-stroke: an umbrella review.

INTRODUCTION: Post-stroke cognitive impairment (PSCI) remains a common problem that can lead to significant disability and dependency. Previous work suggested that methodologically robust trials were lacking and that there is continued uncertainty on the benefits and limitations of pharmacological and non-pharmacological interventions. This umbrella review aims to update and synthesise evidence on pharmacological and non-pharmacological interventions for PSCI. PATIENTS AND METHODS: This review was pre-registered on PROSPERO (CRD42024611909) with searches from 2019 to 2024. Eligible studies we
