Design and Practice of Undergraduate Biochemistry Experiments Based on the DOK Model: A Case Study of HPLC Quantification of TMZ in Mouse Tissues.

While biochemistry laboratory instruction continuously evolves to enhance student learning, effectively cultivating higher-order thinking skills in complex experimental settings remains an ongoing challenge. By applying the Depth of Knowledge (DOK) model, we designed and implemented a comprehensive experiment titled "Determination of Temozolomide Content in Mouse Tissues Using High-Performance Liquid Chromatography (HPLC)" for undergraduate bioscience majors. This experiment helps students understand drug tissue distribution mechanisms while filling a curricular gap in teaching pharmacokinetic

Plasma Lipidome Is Dysregulated in Amyotrophic Lateral Sclerosis and Associated With Life-Course Adiposity Polygenic Risk.

OBJECTIVE: Although both pre-symptomatic adiposity and dysregulated lipid metabolism are implicated in amyotrophic lateral sclerosis (ALS), the mechanistic pathways linking adiposity to disease remain unclear. This study aimed to investigate how genetic predisposition to adiposity might influence ALS through specific lipid metabolic pathways. METHODS: We conducted mass spectrometry-based quantitative lipidomics on plasma from 104 ALS patients and 78 age- and sex-matched controls. Life-course adiposity polygenic risk scores (PRS) for birth weight, body fat percentage, appendicular lean mass an

Serum p-tau181 and p-tau217 levels in adolescents and adults with 5q spinal muscular atrophy.

BACKGROUND: Reliable blood biomarkers for longitudinal monitoring in adolescent and adult 5q spinal muscular atrophy (SMA) are lacking. Phosphorylated tau (p-tau) species are elevated in serum in amyotrophic lateral sclerosis (ALS), where they may reflect peripheral nerve and muscle pathology. We aimed to investigate serum p-tau181 and p-tau217 as longitudinal biomarkers in adolescent and adult SMA and assessed associations with clinical parameters. METHODS: In this retrospective multi-centre study, we assessed serum p-tau181 and p-tau217 levels in adults with 5q SMA under treatment with nusi

Single-nucleotide polymorphism-directed design of allele-specific antisense oligonucleotides for spinocerebellar ataxia type 3.

Spinocerebellar ataxia type 3 (SCA3), also called Machado-Joseph disease, the most common form of autosomal dominant cerebellar ataxia, is caused by a CAG repeat expansion in ATXN3. The mutant protein is prone to aggregation and exerts toxic gain-of-function effects, whereas the wild-type protein has important neuroprotective roles as a deubiquitinase. Current treatment strategies for SCA3 lack specificity for the mutant allele. Identifying genetic variants such as single-nucleotide polymorphisms (SNPs) in linkage with the pathogenic CAG expansion offers a promising avenue to develop allele-sp

Intradural extramedullary venous hemangioma mimicking meningioma: a case report and literature review.

BACKGROUND: Intradural extramedullary hemangiomas of the spinal canal are rare, and the venous subtype is particularly uncommon. Their imaging appearance can overlap with more frequent intradural extramedullary tumors, which increases the risk of misdiagnosis. This case report aims to describe an intradural extramedullary venous hemangioma that mimicked meningioma on MRI and to highlight key surgical considerations. CASE PRESENTATION: A 49-year-old man presented with a five-month history of progressive numbness, radicular pain, and weakness in the left lower limb, which progressed to gait dis

ABCD-Trial: study protocol for a randomized, double-blind, placebo-controlled study in patients with Amyotrophic lateral sclerosis for the evaluation of efficacy and safety of B-Cell Depletion with rituximab (ABCD).

BACKGROUND: The exact disease-causing mechanisms in ALS remain incompletely understood. A possible cause is inflammation of neurons. Compared to other neurodegenerative diseases, neuroinflammation is pronounced in ALS, presumably leading to disease progression and neuronal death. Patients with ALS can have neuronal autoantibodies and most notably antibody-producing cells in the cerebrospinal fluid. The CD20-binding therapeutic antibody rituximab targets these antibody-producing B-cells. Rituximab is a medication commonly used in neurology. Single patients with ALS have already been treated wit

DNA methylation in motor neuron biology and epigeneticopathy in amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS), the 3rd most frequent neurodegenerative disease, is fatal. Skeletal muscles, neuromuscular junctions, and motor neurons in the cerebral cortex, brainstem, and spinal cord that control swallowing, breathing, and movement degenerate, causing paralysis, respiratory arrest, and death. Clinically meaningful disease mechanisms are unknown, and no effective therapies are available. ALS is a polygenic heterogeneous disease seen in 5%-10% of patients as inherited, but most patients have no inheritance pattern. ALS appears to manifest as a complex interplay between m

Caspase-1 and Inflammasome Signaling in Spinal Cord Injury: Mechanistic Insights and Translational Gaps.

Caspase-1 is a central mediator of canonical inflammasome signaling and drives neuroinflammatory injury through interleukin-1β (IL-1β) and interleukin-18 (IL-18) maturation, gasdermin D (GSDMD)-mediated pyroptosis, and secondary inflammatory cascades. In spinal cord injury (SCI) and other neurologic disorders, experimental evidence links caspase-1 activation to blood-spinal cord barrier (BSCB) or blood-brain barrier (BBB) disruption, glial activation, neuronal and oligodendroglial injury, and worse functional outcomes. However, despite mechanistic rationale, the clinical role of caspase-1 as a

Peripheral and cerebrospinal fluid inflammatory markers in sporadic amyotrophic lateral sclerosis an exploratory cross-sectional association study.

Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disorder with an ambiguous etiology, whose onset may be linked to innate immune system activation outside the central nervous system (CNS). We investigated the association between peripheral blood and CNS inflammatory parameters in 45 sporadic ALS patients compared to 30 controls, analyzing peripheral blood tests (C-reactive protein [CRP], blood count, fibrinogen) and cerebrospinal fluid (CSF) immunological measures (Tumor Necrosis Factor alpha [TNF-α], TNF receptor type 1 [TNFR-1], CRP) alongside clinical characteristics.

Feasibility and potential benefits of daily online adaptive radiotherapy for head and neck cancer without margin reduction using an RTT-only workflow.

INTRODUCTION: Online adaptive radiotherapy (oART) may improve head and neck cancer (HNC) treatment, where anatomical changes during radiotherapy (RT) are common. However, oART is resource-intensive and clinical data are limited. We conducted a prospective feasibility trial implementating an RT technologist (RTT)-led workflow to evaluate dosimetric outcomes, workflow strategies, and intrafractional motion in daily oART. MATERIAL AND METHODS: Ten patients receiving curatively intended oART (33-34 fractions) using standard PTV margins were included (NCT: 07506902). For each fraction, a scheduled

Protein disulphide isomerase protects against ALS/FTD phenotypes associated with cyclin F in zebrafish and cellular models.

BACKGROUND: Mutations in CCNF, encoding cyclin F, cause a small proportion of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) cases. Mutant cyclin F mislocalises from the nucleus to the cytoplasm, impairs the ubiquitin proteasome system, disrupts endoplasmic reticulum (ER) to Golgi transport, and triggers ER stress and apoptosis. Protein disulphide isomerase (PDI), a redox-active ER chaperone of the thioredoxin superfamily, protects against ALS phenotypes induced by mutant SOD1, TDP-43, and FUS, but its activity against mutant cyclin F has not been examined. OBJECTIVES:

Stress, diet, and aging: a comprehensive analysis of allostatic load in biological aging.

BACKGROUND: As the global population continues to age, biological aging has emerged as a significant public health challenge. It is imperative to identify modifiable factors and underlying mechanisms that can promote healthy aging. Allostatic load (AL), a comprehensive indicator of chronic stress, is linked to numerous adverse outcomes in behavioral health. However, there is a paucity of comprehensive analyses and mechanistic investigations regarding the relationship between AL and biological aging across the entire population. This study seeks to elucidate the relationship between AL and biol

Microglial Labeling in Mouse Spinal Cord Injury: A Review of Timing, Efficacy, and Specificity.

Traumatic spinal cord injury (SCI) is a complex and disabling condition with no curative therapy currently available. The mechanical damage of the primary injury phase is followed and exacerbated by a secondary injury phase, characterized by the release of free radicals and pro-inflammatory cytokines that lead to chronic inflammation. Microglia, as the resident immune cells of the central nervous system, play a critical role in orchestrating this inflammatory response. Studying their dynamics after SCI not only deepens our understanding of injury pathology but also advances the development of

AT-RvD1 liposomes effectively accumulate in spinal cord and reprogram microglia via ANGPTL4-SUCLG1-propionate axis for treating neuropathic pain.

Neuropathic pain (NP) remains a major clinical challenge as current medications are ineffective in treating the underlying cause, and the clinical use of these drugs also has adverse reactions such as addiction. Therefore, specialized pro-resolving mediators (SPMs) may represent a promising therapeutic strategy. Microglia-mediated neuroinflammation is central to NP pathogenesis, yet feasible strategies for precise modulation of microglial inflammatory responses remain limited. Here, we reveal that aspirin-triggered resolvin D1 (AT-RvD1) promotes a shift of microglia toward an anti-inflammatory

Targeting the gut microbiota to promote motor rehabilitation after spinal cord injury: mechanisms, evidence, and translational challenges.

Spinal cord injury (SCI) is a devastating central nervous system trauma that frequently results in permanent motor dysfunction. Over the past decade the gut microbiota has emerged as a potentially modifiable contributor to secondary injury and neural repair, giving rise to the concept of the gut-spinal cord axis. In this narrative review we synthesize current understanding of how SCI-associated gut dysbiosis may relate to the recovery of motor function, focusing on immune modulation, microbial metabolites such as short-chain fatty acids and bile acids, and gut-central nervous system signaling.

BACKGROUND: Surgical decompression is regarded as standard treatment for degenerative cervical myelopathy (DCM). However, the timing of surgery remains a critical yet unresolved issue in clinical practice. Therefore, this meta-analysis aimed to evaluate the impact of symptom duration on surgical outcomes and determine the optimal timing for surgery. METHODS: A systematic search was conducted on 16th November 2025 using PubMed, Ovid, Scopus, and Web of Science databases to identify eligible articles. Studies comprising patients with DCM undergoing surgery, assessing outcome by mJOA score at a

Diverging diagnostic trajectories of degenerative cervical myelopathy and cervical radiculopathy in South Korea: a nationwide time-series analysis of claims data, 2010-2024.

BACKGROUND: Degenerative cervical myelopathy (DCM) is the leading non-traumatic cause of spinal cord dysfunction in adults, and its burden is projected to grow as populations age. Cervical radiculopathy shares the same degenerative substrate but follows a separate diagnostic pathway. Long-term national trends in the two conditions, and their relation to surgical practice, are unquantified in an East Asian system with universal coverage. METHODS: This nationwide repeated cross-sectional ecological analysis used aggregate open data from the Korean Health Insurance Review and Assessment Service,

Phenotypic Profiling of SOD1-G93A Mice: Integrating Sciatic Nerve Histopathology with Systemic Chemokine Dynamics During ALS Progression.

Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disorder characterized by the progressive loss of motor neurons, leading to muscle atrophy and fatal respiratory failure. While neuroinflammation is a hallmark of ALS, the temporal and spatial relationship between systemic inflammation and peripheral nerve degeneration remains poorly understood. In the present study, the transgenic SOD1-G93A mouse model was employed to conduct a longitudinal analysis of amyotrophic lateral sclerosis (ALS) progression. We integrated morphometric and ultrastructural examinations of the sciati
