Valosin-containing protein counteracts ATP-driven dissolution of FUS condensates through its ATPase activity in vitro.

Fused in sarcoma (FUS) forms phase-separated condensates implicated in amyotrophic lateral sclerosis (ALS). Although millimolar ATP concentrations paradoxically dissolve FUS condensates through hydrotropic activity, condensates nevertheless persist in cells, suggesting active regulatory mechanisms. Here, using a reconstituted system, we show that the AAA+ATPase valosin-containing protein (VCP) counteracts ATP-driven dissolution of FUS condensates. VCP preserved both wild-type and ALS-linked P525L condensates under high ATP conditions, and this protection required catalytic ATPase activity rath

Neurofilaments support the differentiation of amyotrophic lateral sclerosis from degenerative cervical myelopathy.

The differential diagnosis between degenerative cervical myelopathy (DCM) and early amyotrophic lateral sclerosis (ALS) is often clinically challenging; laboratory tests that support this distinction could be highly valuable. This study examined the concentrations of neurofilament light chain (NfL) and the phosphorylated neurofilament heavy chain (pNfH), in CSF and serum of carefully selected patients with DCM, spinal canal stenosis (SCS) and ALS, after exclusion of major neurological confounders. Patients were assigned to five groups according to MRI findings and ALS diagnosis. All patients w

Bringing cellular clarity to the cortical component of ALS with a high-density multi-electrode array system.

There is selective vulnerability in diseases, requiring the understanding of cell-type specific aspects of neurodegeneration with cellular resolution. Single-cell electrophysiology enables direct investigation of neuronal excitability, ion-channel dynamics, and synaptic transmission with high temporal precision, which is crucial in understanding neuronal circuitries and how they are affected in diseases. Microelectrode arrays (MEAs) have emerged as powerful platforms enabling long-term, parallel, and non-invasive extracellular measurements. The advent of complementary-metal-oxide-semiconductor

Revisiting the muscle spindle in ALS: Implications for spinal motor network dysfunction.

Amyotrophic lateral sclerosis (ALS) is typically characterized by progressive degeneration of upper and lower motor neurons. However, the contribution of afferent sensory pathways to spinal motor network disorganization has received little attention. Converging human histological evidence from multiple independent laboratories demonstrates that fusimotor (gamma) motor neuron innervation of muscle spindles degenerates in ALS while sensory endings remain morphologically intact at the end organ, although the afferent neurons and ascending sensory pathways are also affected. This dissociation crea

Treadmill Exercise Can Modulate the Inflammatory Microenvironment and Promote Regeneration After Compressive Spinal Cord Injury in Female Mice.

Traumatic spinal cord injury (SCI) leads to severe sensorimotor impairments and reduced quality of life. Physical rehabilitation has emerged as a key therapeutic strategy to improve function and mitigate complications such as muscle atrophy and joint contractures, partly through modulation of the inflammatory microenvironment. This study investigated the effects of a structured exercise protocol on inflammation, neuroplasticity and sensorimotor recovery following compressive SCI in mice. Female C57BL/6 mice (8-12 weeks) were subjected to extradural spinal cord compression and divided into thre

Spinal microglia mediate chronic itch by promoting ferroptosis.

Chronic itch, affecting 8-10% of the global population, is a debilitating condition linked to dermatoses, systemic diseases, and neurological disorders, with limited therapeutic efficacy due to unclear central mechanisms. Spinal microglia, central nervous system-resident immune cells, regulate neuroinflammation and synaptic plasticity in chronic pain and neurodegeneration, but their role in chronic itch remains unknown. This study investigated the role of spinal microglia in chronic itch. Using an allergic contact dermatitis (ACD) mouse chronic itch model, we found robust microglial activation

Targeting astrocyte-mediated neurotoxicity induced by ALS/FTD-associated RNA binding proteins.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are neurodegenerative disorders characterized by reactive astrocytes that contribute to neuronal injury through TAR DNA-binding protein 43 (TDP-43)-or fused in sarcoma (FUS)-driven neuroinflammatory signaling. Dehydrocostus lactone (DHE), a blood-brain barrier-permeable sesquiterpene lactone with established anti-inflammatory activity, represents a promising but unexplored therapeutic candidate for ALS/FTD. METHODS: The therapeutic effects of DHE were evaluated in primary mouse and human astrocytes expressing AL

PTPRS mediates invasion of tetanus neurotoxin into the central nervous system.

Tetanus neurotoxin (TeNT) invades the central nervous system (CNS) via retrograde transport through motoneurons. Whether a host factor is needed to mediate TeNT trafficking has remained unknown. Here, using pull-down and mass spectrometry analysis, and by developing a luciferase-based in vivo transport assay, we identify receptor protein tyrosine phosphatase sigma (PTPRS; PTPσ) as the retrograde transport receptor for TeNT. Global or motoneuron-specific PTPRS knockout (KO) abolishes TeNT entry into the CNS in mouse models. PTPRS-TeNT interactions occur in endosomes, prevent toxin translocation

Beyond rehabilitation: a qualitative exploration of sport participation context and FES-cycling user experience in spinal cord injury.

BACKGROUND: While functional electrical stimulation (FES)-cycling benefits for spinal cord injury (SCI) are documented, user acceptance and sustained engagement remain underexplored. Understanding how sport participation history and values shape FES-cycling experiences is essential for predicting technology adoption beyond rehabilitation. OBJECTIVE: To explore FES-cycling user experiences within the context of participants' sport participation history, examining how sport-related values, post-injury adaptations, and existing barriers influence device acceptance and attitudes towards sustained

Population-Level Nucleotide Diversity and Genetic Differentiation at SOD1 Across Global Human Populations.

BACKGROUND: The SOD1 gene encodes superoxide dismutase 1, an antioxidant enzyme in which pathogenic variants cause a subset of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Although the clinical and molecular consequences of SOD1 variants are well established, its locus-wide population genetic architecture has not been systematically characterized across global populations. OBJECTIVE: To characterize patterns of genetic diversity, population differentiation, and evolutionary constraint across the SOD1 locus in worldwide human populations. METHODS: We analyzed genomic

Annexin A1 inhibits neuronal apoptosis in amyotrophic lateral sclerosis by restraining the Hippo pathway.

Annexin A1 (ANXA1) is a multifunctional regulatory protein involved in neuroinflammation and cellular homeostasis, but its role in amyotrophic lateral sclerosis (ALS) remains unclear. In this study, we investigated the relationship between ANXA1 and Hippo pathway signaling in ALS. Bioinformatic analysis of transcriptomic data from iPSC-derived motor neurons showed that ANXA1 expression was reduced in ALS and associated with apoptosis and Hippo pathway-related changes. Decreased ANXA1 expression was confirmed in hSOD1^G93A transgenic mice and NSC34 motor neuron-like cells. Phosphorylation level

Splicing modulation of ATXN2 as a therapeutic strategy to regulate Ataxin-2 protein levels in ALS.

ATXN2 is associated with an increased risk of Amyotrophic Lateral Sclerosis (ALS), while down-regulation of ATXN2 has been shown to mitigate TDP-43 proteinopathy in ALS models. In this study, we demonstrated that Ataxin-2 protein levels were upregulated in rNLS8 mice following doxycycline withdrawal, which coincided with TDP-43 overexpression, phosphorylation, and aggregation. To reduce Ataxin-2 protein levels, we explored the approach of splicing modulation at the RNA level. Through bioinformatical analysis, we identified an alternative 5' splicing site of ATXN2 in intron 8. This alternative

Cholinergic inputs to motoneurons in the spinal cord augment focal muscle spasticity in rodents.

BACKGROUND AND PURPOSE: Cholinergic premotor inputs to motoneurons support high levels of motor output when required. However, the possible role of these inputs in pathologically elevated muscle tone has not been characterised. To address this, we examined the modulatory effect of spinal acetylcholine (ACh) on experimental spastic paralysis. EXPERIMENTAL APPROACH: Rats were injected with tetanus neurotoxin (TeNT, 33-100 pg, im) into gastrocnemius to induce sustained rigid hind-paw extension and resistance to passive ankle flexion. The effects of spinal ACh on experimental hypertonia were prob

PANoptosis In Microglia and Neurons Following Traumatic CNS Injury: A Systematic Review of Pathological Features and Therapeutic Implications.

BACKGROUND: Traumatic brain injury (TBI) and spinal cord injury (SCI) induce PANoptosis, a pro-inflammatory programmed cell death modality orchestrated by the PANoptosome. This modality integrates pyroptosis, apoptosis, and necroptosis molecular machinery and contrasts with canonical single-pathway cell death. METHODS: This systematic review (PROSPERO CRD420261383435) followed PRISMA and searched PubMed, Web of Science, and Cochrane Library. The in vivo studies confirming PANoptosis at the protein level were included; quality was assessed using CRIME-Q, and shared genes/protein interactions w

Serum Neurofilament Light Chain and GFAP in Amyotrophic Lateral Sclerosis on a Commercial ECLIA Platform.

INTRODUCTION/AIMS: In amyotrophic lateral sclerosis (ALS), SIMOA-based studies show that baseline serum neurofilament light chain (NfL) predicts the revised ALS Functional Rating Scale (ALSFRS-R) decline rate and survival. The Roche Elecsys electrochemiluminescence immunoassay (ECLIA) reports values approximately six-fold lower; cross-platform comparison confirms comparable performance, but serial clinical-practice data remain limited. We assessed whether ECLIA NfL correlates with the ALSFRS-R decline rate under the sampling conditions of clinical practice, and whether GFAP or S-100B adds prog

Trunk-mediated spinal cord stimulation: A step toward clinical implementation after spinal cord injury.

In this issue of Med, Agnesi et al., show that residual trunk movements after spinal cord injury can modulate the effects of epidural spinal cord stimulation (SCS) to enable participants to volitionally initiate and control stepping.1 This trunk-mediated interface offers a practical strategy for bringing epidural SCS one step closer to widespread clinical adoption.

Agrin-Enriched Neonatal Spinal Cord-Derived Extracellular Vesicles Bypass Age Barriers to Organize Spinal Cord Repair.

Spinal cord injury (SCI) poses a formidable challenge in regenerative medicine, with current therapeutic strategies proving largely unsuccessful in achieving functional repair. Notably, paediatric patients and neonatal mice exhibit spontaneous and efficient regeneration after SCI, while the mechanisms remain elusive. Through integrated multi-omics analysis in neonatal, adult and aged mice with SCI, we discovered that the regeneration time window, inflammatory response, and intercellular communication are all age-dependent. Neonatal spinal cord-derived extracellular vesicles (nSC-EVs) were iden
