Somatic gene mutations in the motor cortex of patients with sporadic amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is characterized by the progressive degeneration of cortical and spinal motor neurons. Mendelian germline mutations often cause familial ALS (fALS) but only approximately 10% of sporadic ALS cases (sALS). We leveraged DNA and single-cell RNA sequencing data from autopsy tissue to explore the presence of somatic mosaic variants in sALS cases. Deep targeted panel sequencing of known ALS disease genes in motor cortex tissue revealed an enrichment of low allele frequency variants in sALS, but not in fALS with an identified monogenic cause. In silico analysis pre

Nanomaterial-based therapeutic strategies for spinal cord injury repair: harnessing multifunctionality to overcome pathophysiological challenges.

Spinal cord injury (SCI) is a severe central nervous system (CNS) disorder caused by mechanical trauma, leading to primary injury characterized by irreversible neural damage and secondary injury involving cascades of neuroinflammation, oxidative stress, glial scar formation, and disruption of the blood-spinal cord barrier (BSCB), which collectively result in profound functional deficits. This review synthesizes recent progress (past five years) in nanomaterial-based strategies to address the multifaceted pathophysiology of SCI. Nanomaterials leverage their tunable size, surface functionalizati

Restoration of Microglia-Dependent Signaling Networks Drives Tissue Repair and Functional Recovery After Spinal Cord Injury in Microglia-Depleted Mice.

Regulatory mechanisms underlying microglia-dependent restorative effects during the early stages following spinal cord injury (SCI) remain uncertain. In adult mice, microglia depletion exacerbates injury and impairs functional recovery, suggesting that microglia play a protective role after SCI. We performed RNA sequencing on four spinal cord conditions (uninjured, injured, microglia-depleted uninjured, and microglia-depleted injured) to identify the core transcriptional signature of microglia-dependent restorative functions after SCI. Bioinformatics analysis identified 16 genes as critical mi

Collagen-Based Hydrogel Loaded With Microspheres Encapsulated With Placental Mesenchymal Stem Cells-Derived Exosomes Synergistically Promote Recovery After Spinal Cord Injury in Rats.

Traumatic spinal cord injury (TSCI) is a serious medical issue where there is a loss of sensorimotor function. Current interventions continue to lack the ability to successfully enhance these conditions; therefore, it is crucial to consider alternative effective strategies. Currently, we investigated the effects of collagen-based hydrogel (CbH) loaded with microspheres encapsulated with placental mesenchymal stem cells (PMSCs)-derived exosomes in the recovery of TSCI in rats. Sixty adult male Sprague-Dawley rats were randomly assigned into four groups (n = 15/group): TSCI (injury without treat

Methodological optimization for eliciting robust median nerve somatosensory evoked potentials for realtime single trial applications.

Objective.Single-trial measurement of median nerve somatosensory evoked potentials (SEPs) with noninvasive electroencephalography (EEG) is challenging due to low signal-to-noise ratio (SNR), limiting its use in real-time neurorehabilitation applications. We describe and evaluate methodological optimizations for eliciting reliable median nerve SEPs measurable in real time, with reduced reliance on post-processing.Methods.In twelve healthy participants, two sessions each, SEPs were assessed at three pulse widths (0.1, 0.5, 1 ms), at a low-frequency stimulation (0.5 Hz ± 10%), and at an intensity

Ependymal Yes-Associated Protein Promotes the Neural Regeneration Through Enhancing the Ependymal Cell-Derived Perilesional Glial Borders in Mice After Spinal Cord Injury.

OBJECTIVES: Ependymal cell-derived perilesional glial borders may play a beneficial role in neural regeneration after spinal cord injury (SCI). Yes-associated protein (YAP), a key transcriptional cofactor, is involved in the control of body organ size by regulating cell differentiation, proliferation, growth, and apoptosis; however, it remains unclear whether the roles and underlying mechanisms of YAP signaling regulate the ependymal cell-derived perilesional glial borders after SCI. METHODS: We established a dorsal cord incision injury mouse model. The YAPf/f; FoxJ1-CreERT2 (YAPFoxJ1-ERT2-CK

Unveiling the Genetic Association Between Hemoglobin Concentration and Amyotrophic Lateral Sclerosis.

BACKGROUND: Although previous studies have suggested an association between hemoglobin (Hb) concentration and amyotrophic lateral sclerosis (ALS), the precise cause-and-effect relationship between them is still unclear. This study aims to investigate the causal correlation between Hb concentration and ALS, and explore the potential genes related to their association. METHODS: We extracted summary statistical data of Hb concentration and ALS from genome-wide association studies (GWAS), performed Mendelian randomization (MR) analyses, and conducted RNA sequencing of motor neurons different from

Joint model with latent disease age: Overcoming the need for reference time.

Heterogeneity of the progression of neurodegenerative diseases is one of the main challenges faced in developing therapies. Thanks to the increasing number of clinical databases, progression models have allowed a better understanding of this heterogeneity. Joint models have proven their effectiveness by combining longitudinal and survival data. Nevertheless, they require a reference time, which is ill-defined for neurodegenerative diseases, where biological underlying processes start before the first symptoms. In this work, we propose a joint non-linear mixed-effect model with a latent disease

Homozygosity for the C allele at UNC13A rs12608932 seems to compromise cognition in ALS independently of the cognitive domains.

The common single nucleotide polymorphism (SNP) rs12608932 located at a cryptic splice in the UNC13A gene has been reported to modify the clinical phenotype of ALS, but it is unclear whether homozygosity for the C-allele at UNC13A rs12608932 modifies specific domains of cognition in ALS. We analyzed retrospective data from a German cohort and found that the proportion of cognitively or behaviorally impaired patients was higher in the high-risk group of homozygous C-allele carriers. Patients with C/C alleles had lower scores than controls on verbal fluency, executive functioning, and delayed me

LDHA as a Potential Therapeutic Target for Lactylation Regulation in Spinal Cord Injury: Integrated Bioinformatics Analysis, Experimental Validation, and Drug Prediction.

Lactate metabolism and its epigenetic modification, lactylation, are emerging regulators in neurological disorders, yet their roles in spinal cord injury (SCI) remain unclear. Here, we mined SCI transcriptomes from GEO and identified 13 differentially expressed lactylation-related genes (DE-LRGs), with lactate dehydrogenase A (LDHA) significantly upregulated. Immune-infiltration analysis linked LDHA to T cells gamma delta, and Mfuzz time-series clustering showed dynamic LDHA upregulation during acute SCI, which was validated in GSE45006. Random forest and ROC analyses supported the diagnostic

Video summarization for home-based egocentric footage in spinal cord injury rehabilitation.

BACKGROUND: Wearable cameras provide a means to assess hand function in individuals with spinal cord injury (SCI) beyond clinical settings. Previous studies have found that clinicians acknowledge the potential of egocentric video to monitor and inform rehabilitation. Nonetheless, the need for time-intensive manual review of the footage remains a challenge to its integration into clinical practice. To address this barrier, we investigated the utility of video summarization for egocentric videos of hand use after SCI. METHODS: A dataset comprising 316 egocentric videos from 20 individuals with

Superficial white matter microstructural impairments correlate with functional alterations and disease severity in early-stage amyotrophic lateral sclerosis.

PURPOSE: White matter (WM) damage is a key pathophysiological process in amyotrophic lateral sclerosis (ALS). However, alterations in superficial WM (SWM) have not been systematically explored. This study aimed to assess SWM microstructural changes in early-stage ALS and their associations with cortical functional alterations and disease severity. METHODS: Forty-two early-stage ALS patients and 48 healthy controls were included. Disease severity was evaluated using the revised ALS Functional Rating Scale (ALSFRS-R). The SWM was identified by sampling voxels along the cortical surface, maintai

Lactate Dehydrogenase Inhibition Reverts the Fatty Acid-Induced Neurotoxic Phenotype of Astrocytes.

Astrocytes are central to lipid metabolism in the central nervous system. Due to their morphological and functional characteristics, astrocytes can uptake fatty acids (FAs) from the bloodstream and extracellular space and store them in lipid droplets (LD). LD are dynamic organelles, whose accumulation in astrocytes has been shown to occur upon exposure to various stress stimuli. Different hypotheses proposed to explain motor neuron degeneration in amyotrophic lateral sclerosis (ALS) implicate mitochondrial dysfunction and oxidative stress. Mitochondrial dysfunction in astrocytes is associated

Biologically-constrained spiking neural network for neuromodulation in locomotor recovery after spinal cord injury.

Presynaptic inhibition after spinal cord injury (SCI) has been hypothesised to disproportionately affect flexion reflex loops in locomotor spinal circuitry. Reducing gamma-aminobutyric acid (GABA) inhibitory activity increases the excitation of flexion circuits, restoring muscle activation and stepping ability. Conversely, nociceptive sensitisation and muscular spasticity can emerge from insufficient GABAergic inhibition. To investigate the effects of neuromodulation and proprioceptive sensory afferents in the spinal cord, a biologically constrained spiking neural network (SNN) was developed.

Modeling amyotrophic lateral sclerosis (ALS) in vitro: from mechanistic studies to translatable drug discovery.

Amyotrophic lateral sclerosis is a rapidly progressing, fatal neurodegenerative disease that causes selective degeneration of the corticomotor system. Currently, ALS remains incurable, and the available treatment options offer little in the way of extending life or improving quality of life. This is due, at least in part, to a lack of representative disease models. In vitro modeling offers rapid, experimentally accessible platforms for mechanistic discovery research and drug screening, but modeling the complexity of ALS - a multicellular, multisystem disease - in a dish, is not without its cha
