Human-machine Interface using functional electrostimulation and inertial sensors for lower limb rehabilitation in spinal cord injury individuals: a proof of concept.

A spinal cord injury (SCI) is a neurological disorder that impairs motor and physiological functions and leads to a reduced quality of life and autonomy for the person affected. In this scenario, human-machine interfaces (HMIs) have emerged as an effective tool to leverage residual motor capabilities and benefit injured persons. This work aims to develop a closed-loop HMI system for lower-limb rehabilitation composed of an in-house multi-channel Functional Electrical Stimulation (FES), which is activated by considering gait and pedaling cycles measured by an Inertial Measurement Unit. Two expe

Hydrogen treatment attenuates ferroptosis and alleviates spinal cord ischemia-reperfusion injury by activating the Nrf2/HO-1 signaling pathway.

PURPOSE: Spinal cord ischemia-reperfusion injury (SCIRI) can lead to significant losses in sensory and motor functions. The precise role of hydrogen (H2) as an antioxidant in the process of ferroptosis is not fully determined. MATERIALS AND METHODS: This study used an abdominal aorta ligation technique to establish a SCIRI model in rats. Following oxygen-glucose deprivation/reoxygenation (OGD/R), HT22 cells were treated with H2 to assess its impact on ferroptosis. Hindlimb motor function was evaluated using the motor deficit index (MDI) and Basso, Beattie, Bresnahan (BBB) scoring, while neuro

The multistep pathogenic hypothesis of amyotrophic lateral sclerosis is incompatible with the epidemiological data.

Amyotrophic lateral sclerosis (ALS) is a multifactorial neurodegenerative disease whose incidence increases with age. According to the gene-time-environment hypothesis, ALS onset occurs through the interaction between genes and environmental exposures during ageing, which may involve a continuous accumulation process. Alternatively, the multistep pathogenic hypothesis, based on the Armitage-Doll multistep model from cancer research, posits that a discrete number of specific sequential "hits" are necessary to trigger ALS. Here we analyzed three large population-based epidemiological datasets of

An ALS-associated mutant SOD1 protein can be eliminated in microglia culture by selective autophagy.

The acquired toxicity of the familial amyotrophic lateral sclerosis (ALS)-associated mutant Zn-superoxide dismutase 1 (SOD1) protein has been implicated in motoneuron death, and cytosolic aggregates or inclusions have been observed in the cytoplasm of motoneurons, astrocytes, and neuronal axons but not in that of microglia. This study elucidates the mechanisms by which mutant SOD1 does not aggregate in and is cleared by microglia. We generated pcDNA3-Venus-tagged SOD1 constructs: wild-type SOD1 and mutant SOD1 were used as controls, and the A4V, D90A, and G93A SOD1 mutants were used as disease

C9orf72 in myeloid cells prevents an inflammatory response to microbial glycogen.

Gut dysbiosis and neural inflammation occur in patients with amyotrophic lateral sclerosis (ALS), including those with a causal mutation in chromosome 9 open reading frame 72 (C9ORF72). How gut commensals interact with common ALS genotypes to impart risk of neural degeneration remains unclear. Here, we identify 10 phylogenetically diverse bacterial strains that promote cytokine release in a C9orf72-dependent manner. Metatranscriptomics implicated the glycogen biosynthesis pathway as a driver of inflammation. Colonization of germ-free C9orf72-deficient mice with Parabacteroides merdae that prod

TRIM9 and TRIM26 Interact with UBQLN2P497H to Modulate Its Proteasomal Degradation.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by progressive motor neuron loss. ALS-linked mutations in UBQLN2 promote protein aggregation and disrupt proteostasis, yet the mutation-specific protein interactomes and their functional relevance remain poorly defined. We employed APEX2 proximity labeling, together with affinity enrichment of biotinylated peptides and LC-MS/MS analysis, to profile the interactomes of wild-type UBQLN2 and two ALS-linked variants, UBQLN2P497H and UBQLN2P497S. We identified 785 unique biotinylated proteins, many of which exhi

Multifunctional Diselenide-Based Antioxidative Nanozymes Promote Spinal Cord Repair via Oxidative Stress Alleviation and Immune Modulation.

Spinal cord injury could trigger an excessive reactive oxygen species formation and a sustained inflammatory response, both of which disrupt neural repair processes. Therefore, it is urgent that exploiting a therapeutic intervention that can simultaneously neutralize ROS and restore immune balance. This study reports a hyaluronic acid-based diselenide cross-linked nanogel (Se-Se@HA) designed to integrate catalytic ROS scavenging and immunomodulatory properties to reconstruct the damaged microenvironment and promote repair. Se-Se@HA exhibits significant structural stability and selectively resp

Lipid Metabolic Mediators Bridge Ischemic Heart Disease and Amyotrophic Lateral Sclerosis.

While epidemiological studies have linked cardiovascular disease (CVD) and amyotrophic lateral sclerosis (ALS), the causal pathways remain unclear. This study aims to clarify the causal relationship between CVD and ALS, with a focus on lipid metabolism as a potential mediator. We conducted a bidirectional two-sample Mendelian randomization (MR) analysis to investigate the causal relationship between CVD and ALS. Furthermore, we utilized mediation MR, summary-data-based MR analysis (SMR), the Kyoto Encyclopedia of Genes and Genomes (KEGG), Gene Ontology (GO) pathway analysis, miRNA interaction

Tumor-associated macrophages enhance peripheral nerve tumor infiltration and spinal cord repair.

Tumor-associated macrophages (TAMs) enhance cancer progression by promoting angiogenesis, extracellular matrix remodeling, and immune suppression. Nerve infiltration also contributes to tumor growth. However, the role of TAMs in promoting intratumoral nerve growth remains unclear. In this study, we have shown that TAMs express a distinct neural growth gene signature. TAMs actively enhanced neural growth within tumors and directly promoted in vitro neurite outgrowth. We identified secreted phosphoprotein 1 (SPP1) as a required mediator of TAM-driven neural growth and mTORC2 activation. Leveragi

Modulation of the miR-485-3p/PGC-1α Pathway by ASO-Loaded Nanoparticles Attenuates ALS Pathogenesis.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder characterized by progressive motor neuron degeneration with limited treatment options. In this study, we investigated the pathological role of microRNA-485-3p (miR-485-3p) in ALS, particularly its regulation of PGC-1α, a transcriptional coactivator essential for mitochondrial function and neuroprotection. We also evaluated the therapeutic potential of BMD-001S, a nanoparticle-based formulation encapsulating an antisense oligonucleotide targeting miR-485-3p. Our results demonstrated that miR-485-3p expression was signific

Peripheral Immune Remodeling After Spinal Cord Injury: Marked by Expansion of CD38+, CTLA-4+, and PD-1+ T and NK Cells.

BACKGROUND AND OBJECTIVES: Spinal cord injury (SCI) is associated with severe immunologic changes, such as SCI-induced immune deficiency syndrome, which heightens susceptibility to infections. However, the immune components underlying this immune reorganization remain poorly defined. This study aimed to characterize immune remodeling in patients with SCI across different time points postinjury. METHODS: High-dimensional flow cytometric profiling was performed on peripheral blood samples from patients with SCI in a cross-sectional observational study to assess immune changes at different posti

VKORC1L1 Downregulation Induced Vitamin K Cycle Disorder Exacerbates Neuronal Ferroptosis After Spinal Cord Injury.

Ferroptosis contributes significantly to neuronal loss and functional decline after spinal cord injury (SCI). Emerging evidence links the vitamin K (VK) cycle to ferroptosis regulation, but its status and role in SCI remain unknown. Therefore, this study aims to elucidate the role and underlying mechanism of the VK cycle in neuronal ferroptosis post-SCI, thereby providing insights for developing novel therapeutic strategies. We quantified VK2 levels and examined VK cycle enzyme expression in injured spinal cords after SCI to identify mechanisms of VK2 reduction. The role of VKORC1L1 in neurona

Understanding liquid-liquid phase separation through TDP-43: fundamental principles, subcellular compartmentalisation, and role of solid inclusion formation.

Phase separation is an important process in biology associated with formation of membraneless organelles but possibly related to the emergence of solid inclusions. TDP-43 is a largely studied paradigmatic case, as it forms neuronal cytoplasmic inclusions in neurodegenerative diseases and is an essential component of many membraneless organelles. Here, we review the physicochemical fundamentals of liquid-liquid phase separation (LLPS) of TDP-43 and its fragments in vitro, showing that full-length TDP-43 requires RNA or chaperones to form stable liquid droplets. We describe TDP-43-containing mem

Ganglioglioma in the spinal cord of a steer.

An 18-mo-old Aberdeen Angus steer was unable to rise without assistance; and, when standing, the steer walked with flexed forelimbs. Due to the poor prognosis, it was euthanized and autopsied. Grossly, the cervical spinal cord was markedly enlarged at C5-C6 by a yellow, soft, well-demarcated nodule that affected ~70% of the parenchyma. Microscopically, the expansive, multinodular, unencapsulated neoplasm affected mostly the white matter and consisted of 2 distinct cell populations. Small, pleomorphic cells with scant cytoplasm predominated; nuclei were positive for oligodendrocyte transcriptio

In Situ Forming ROS-Scavenging Hydrogel with a STING Inhibitor Improves Recovery After Acute Spinal Cord Injury by Modulating Inflammatory Microenvironment.

The inflammatory cascade initiated by acute spinal cord injury (SCI) is a crucial element contributing to subsequent pathological damage. Uncontrolled or excessive inflammatory responses aggravate neural tissue destruction and hinder the regenerative process. Stimulator of interferons genes (STING) is a key regulator in the innate immune signaling pathway that promotes the generation of inflammatory mediators by activating the TBK1 and IRF3 signaling pathways, thereby shaping the post-injury microenvironment. In this study, we developed a hyaluronic acid-phenylboronic acid-polyvinyl alcohol-ba

Concentration-dependent cytoplasmic phase separation of TDP-43 drives aggregation and proteinopathy.

TDP-43 mislocalization and aggregation are common features of several neurodegenerative diseases, including amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD). However, the mechanisms underlying the transition of nuclear TDP-43 to cytoplasmic aggregates, and their contribution to disease pathogenesis, remain poorly understood. To address this gap, we present a methodology to chemically control the assembly and disassembly of cytoplasmic TDP-43 condensates. By fusing TDP-43 to a phase separation-prone protein scaffold, we can induce the formation of cytoplasmic TDP
