MYC-driven gliosis impairs neuron-glia communication in amyotrophic lateral sclerosis.

Chronic activation of glial cells leads to the dysfunction and degeneration of motor and cortical neurons in amyotrophic lateral sclerosis and frontotemporal dementia with an unknown mechanism. To shed light on the molecular pathogenetic processes underlying the exordium and contribution of gliosis to disease onset and progression, we used cells, mice and patient-derived cells modelling TDP-43, SOD1 and C9ORF72-linked and sporadic ALS. Our data reveal a sequential disease progression, starting with enhanced glial reactivity and proliferation, and transitioning into inflammation with upregulati

A next-generation HDAC6 inhibitor for amyotrophic lateral sclerosis and frontotemporal dementia.

Dysregulated proteostasis and intracellular transport contribute to neurodegeneration. Histone deacetylase 6 (HDAC6), a therapeutic target of interest for neurodegenerative diseases, acts at a nexus modulating both proteostasis and intracellular transport. Inhibition of HDAC6 deacetylase activity promotes autophagic clearance of protein aggregates and increases α-tubulin acetylation, thereby enhancing microtubule resiliency and motor protein-microtubule binding, which facilitates intracellular transport and, subsequently, proteostasis. Despite these benefits, advancement of HDAC6 inhibitor the

Profiling cognition and brain metabolism in amyotrophic lateral sclerosis and frontotemporal dementia.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are described as a disease continuum, given their shared clinical, genetic and pathological characteristics. Comparisons of clinical and biomarker features within the ALS and behavioural variant FTD (bvFTD) spectrum are of utmost importance for diagnostic and prognostic purposes. This study investigated biomarker differences between cognitively normal patients with ALS (ALS-cn), ALS-FTD patients and bvFTD patients. Participants, genetically screened for known ALS- and FTD-associated mutations, underwent neuropsychological as

Improved Sensation With Transcutaneous Spinal Cord Stimulation Following Chronic Spinal Cord Injury.

INTRODUCTION: Following spinal cord injury (SCI), recovery of sensation is often neglected despite its critical roles in motor task execution and injury prevention. The Up-LIFT trial recently found targeted transcutaneous spinal cord stimulation (tSCS) combined with rehabilitation significantly improved sensation in addition to upper extremity (UE) strength and function. Here, we explore the clinical and functional significance of these sensory improvements. METHODS: Sixty participants with chronic, incomplete cervical SCI completed a two-month rehabilitation run-in phase followed by two mont

Single-nucleus transcriptomics of spinal ependymoma subtypes recognizes intratumoral heterogeneity.

BACKGROUND: The 2 most prevalent types of spinal ependymal tumors are myxopapillary ependymomas (MPE) and spinal ependymomas (SP-EPN). Based on molecular data from bulk tumor samples, we previously identified clinically relevant subtypes with poor (MPE-A; SP-EPN-A) and favorable progression-free survival (MPE-B; SP-EPN-B). However, detailed cellular composition, molecular heterogeneity, and features of tumor progression are largely unknown. METHODS: We performed singlenucleus transcriptomic sequencing of 25 formalin-fixed and paraffin-embedded spinal ependymal tumors, including all MPE and SP

A Novel Brain-Computer Interface Application: Precise Decoding of Urination and Defecation Motor Attempts in Spinal Cord Injury Patients.

Patients with spinal cord injury (SCI) often face urinary and defecation dysfunction, and existing treatments have limited effectiveness. Brain-computer interface (BCI) technology has been shown to have positive effects on the rehabilitation of SCI patients, but its application in promoting the recovery of urinary and defecation functions has not been explored. This study proposes a new BCI application approach and develops an accurate decoding model targeted at urination and defecation motor attempt tasks. Specifically, we designed a Bidirectional Temporal Convolutional Network (UDCNN-BiTCN)

Somatic gene mutations in the motor cortex of patients with sporadic amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is characterized by the progressive degeneration of cortical and spinal motor neurons. Mendelian germline mutations often cause familial ALS (fALS) but only approximately 10% of sporadic ALS cases (sALS). We leveraged DNA and single-cell RNA sequencing data from autopsy tissue to explore the presence of somatic mosaic variants in sALS cases. Deep targeted panel sequencing of known ALS disease genes in motor cortex tissue revealed an enrichment of low allele frequency variants in sALS, but not in fALS with an identified monogenic cause. In silico analysis pre

Nanomaterial-based therapeutic strategies for spinal cord injury repair: harnessing multifunctionality to overcome pathophysiological challenges.

Spinal cord injury (SCI) is a severe central nervous system (CNS) disorder caused by mechanical trauma, leading to primary injury characterized by irreversible neural damage and secondary injury involving cascades of neuroinflammation, oxidative stress, glial scar formation, and disruption of the blood-spinal cord barrier (BSCB), which collectively result in profound functional deficits. This review synthesizes recent progress (past five years) in nanomaterial-based strategies to address the multifaceted pathophysiology of SCI. Nanomaterials leverage their tunable size, surface functionalizati

Restoration of Microglia-Dependent Signaling Networks Drives Tissue Repair and Functional Recovery After Spinal Cord Injury in Microglia-Depleted Mice.

Regulatory mechanisms underlying microglia-dependent restorative effects during the early stages following spinal cord injury (SCI) remain uncertain. In adult mice, microglia depletion exacerbates injury and impairs functional recovery, suggesting that microglia play a protective role after SCI. We performed RNA sequencing on four spinal cord conditions (uninjured, injured, microglia-depleted uninjured, and microglia-depleted injured) to identify the core transcriptional signature of microglia-dependent restorative functions after SCI. Bioinformatics analysis identified 16 genes as critical mi

Collagen-Based Hydrogel Loaded With Microspheres Encapsulated With Placental Mesenchymal Stem Cells-Derived Exosomes Synergistically Promote Recovery After Spinal Cord Injury in Rats.

Traumatic spinal cord injury (TSCI) is a serious medical issue where there is a loss of sensorimotor function. Current interventions continue to lack the ability to successfully enhance these conditions; therefore, it is crucial to consider alternative effective strategies. Currently, we investigated the effects of collagen-based hydrogel (CbH) loaded with microspheres encapsulated with placental mesenchymal stem cells (PMSCs)-derived exosomes in the recovery of TSCI in rats. Sixty adult male Sprague-Dawley rats were randomly assigned into four groups (n = 15/group): TSCI (injury without treat

Methodological optimization for eliciting robust median nerve somatosensory evoked potentials for realtime single trial applications.

Objective.Single-trial measurement of median nerve somatosensory evoked potentials (SEPs) with noninvasive electroencephalography (EEG) is challenging due to low signal-to-noise ratio (SNR), limiting its use in real-time neurorehabilitation applications. We describe and evaluate methodological optimizations for eliciting reliable median nerve SEPs measurable in real time, with reduced reliance on post-processing.Methods.In twelve healthy participants, two sessions each, SEPs were assessed at three pulse widths (0.1, 0.5, 1 ms), at a low-frequency stimulation (0.5 Hz ± 10%), and at an intensity

Ependymal Yes-Associated Protein Promotes the Neural Regeneration Through Enhancing the Ependymal Cell-Derived Perilesional Glial Borders in Mice After Spinal Cord Injury.

OBJECTIVES: Ependymal cell-derived perilesional glial borders may play a beneficial role in neural regeneration after spinal cord injury (SCI). Yes-associated protein (YAP), a key transcriptional cofactor, is involved in the control of body organ size by regulating cell differentiation, proliferation, growth, and apoptosis; however, it remains unclear whether the roles and underlying mechanisms of YAP signaling regulate the ependymal cell-derived perilesional glial borders after SCI. METHODS: We established a dorsal cord incision injury mouse model. The YAPf/f; FoxJ1-CreERT2 (YAPFoxJ1-ERT2-CK

Unveiling the Genetic Association Between Hemoglobin Concentration and Amyotrophic Lateral Sclerosis.

BACKGROUND: Although previous studies have suggested an association between hemoglobin (Hb) concentration and amyotrophic lateral sclerosis (ALS), the precise cause-and-effect relationship between them is still unclear. This study aims to investigate the causal correlation between Hb concentration and ALS, and explore the potential genes related to their association. METHODS: We extracted summary statistical data of Hb concentration and ALS from genome-wide association studies (GWAS), performed Mendelian randomization (MR) analyses, and conducted RNA sequencing of motor neurons different from

Joint model with latent disease age: Overcoming the need for reference time.

Heterogeneity of the progression of neurodegenerative diseases is one of the main challenges faced in developing therapies. Thanks to the increasing number of clinical databases, progression models have allowed a better understanding of this heterogeneity. Joint models have proven their effectiveness by combining longitudinal and survival data. Nevertheless, they require a reference time, which is ill-defined for neurodegenerative diseases, where biological underlying processes start before the first symptoms. In this work, we propose a joint non-linear mixed-effect model with a latent disease
