AA and Parkinson's Disease.

Parkinson's disease (PD) is due to degeneration of dopaminergic neurons in substantia nigra pars compacta. PD neuropathology is characterized by the presence of Lewy bodies that contain aggregated α-synuclein, which has been implicated in a range of interactions with phospholipid membranes and free fatty acids. I propose that α-synuclein interacts with arachidonic acid (AA) to regulate the function of dopaminergic neurons. α-Synuclein sequesters AA, whereas AA induces the formation of oligomers that are alpha-helical; resistant to fibril formation; more prone to disaggregation, enzymatic diges

AA in Brain-Memory, Dementia, Depression, Anxiety, Alzheimer's Disease, Affective and Psychiatric Disorders, and Coronary Heart Disease (CHD).

The human brain is rich in AA and DHA, which are crucial for normal brain growth and development and memory. They form a major structural component of neurons of the cerebral cortex and photoreceptor cells in the retina. AA and DHA support synaptic plasticity and contribute to efficient neurotransmission. Adequate amounts of AA and DHA support neuronal growth and the formation of new synapses and thus are crucial for memory and cognitive function.Depression, anxiety, schizophrenia, and other neurological diseases occur more frequently in those with coronary heart disease (CHD) and vice versa.

Systematic Dissection of Key Driver Perturbation Signatures in Single Cells via ECCITE-seq.

CRISPR screens, such as expanded CRISPR-compatible cellular indexing of transcriptomes and epitopes by sequencing (ECCITE-seq), enable the simultaneous measurement of transcriptomes, gRNA identity, and cell-surface protein expression at single-cell resolution to systematically interrogate gene function. This platform provides a powerful and scalable experimental approach for validating disease-associated regulators identified by large-scale association studies and other computational methods, including network-based analyses of multi-omics data. Here, as an example application, we describe an

NR3C1 promotes group 4 medulloblastoma invasion via activating VCAN.

Approximately 30% of group 4 medulloblastomas (G4-MBs) present with metastasis at diagnosis, yet the molecular mechanisms remain unclear. To elucidate differences of genes' expression between primary tumors of metastatic (M+) and non-metastatic (M0) G4-MB, we performed multi-omics profiling, including RNA sequencing, proteomics, single-nucleus RNA sequencing (snRNA-seq), and spatial transcriptomics on tumor samples. Integrative analyses identified VCAN, a chondroitin sulfate proteoglycan, as the most significantly up-regulated gene in primary tumors of M+ G4-MB. High expression of this gene wa

Comment on 'Evidence Mapping of Clinical Practice Guidelines Recommendations and Quality for Nutritional Management in Dementia'.

This commentary critically appraises the evidence-mapping review by Su et al. on clinical practice guideline recommendations for nutritional management in dementia. Although the review provides a valuable synthesis using AGREE II and RIGHT checklist methodology, several interpretive limitations warrant attention before its recommendations are translated into practice. First, the included guidelines represent structurally distinct sources: dementia-specific guidelines incorporating nutritional advice and nutrition-focused guidelines extrapolating recommendations to dementia populations. Present

Artificial Intelligence-Based CTA Software for Real-World Detection of Large-Vessel Occlusion in Acute Ischemic Stroke.

INTRODUCTION: Artificial intelligence (AI)-based tools for CT angiography (CTA) have been introduced to support rapid detection of large vessel occlusion (LVO) in acute ischemic stroke. Although regulatory approval has relied mainly on controlled clinical studies, evidence from routine clinical practice remains limited. This study aimed to assess the real-world diagnostic performance and workflow impact of the Brainomix e-CTA software. METHODS: We conducted a retrospective, single-center observational study including consecutive patients with suspected acute ischemic stroke who underwent mult

Correlation between resumption of employment and overall survival following awake mapping-based surgery for IDH-mutant grade 2 glioma: a consecutive series with 515 patients.

OBJECTIVE: Although the life expectancy after surgery for IDH-mutant grade 2 glioma (G2G) is now approximately 20 years, the resumption of postresection employment has received little attention. The aim of this study was to identify factors associated with return to work (RTW) following awake mapping-guided resection and its potential impact on long-term outcomes in a homogeneous cohort of patients with G2G. METHODS: Patients with G2G who were employed before surgery and had ≥ 2 years of postoperative follow-up were identified from a prospective database of connectome-based awake surgical cas

Low-frequency bipolar versus high-frequency monopolar cortical and subcortical motor mapping in craniotomies for brain tumors.

OBJECTIVE: Maximal safe resection of intra-axial brain tumors near the motor cortex is aided by intraoperative motor mapping. Here, the authors compared outcomes in patients undergoing either low-frequency bipolar stimulation (LFBS) or high-frequency monopolar stimulation (HFMS) for cortical and/or subcortical motor mapping during brain tumor resection. METHODS: A retrospective analysis of patients undergoing an asleep craniotomy with motor mapping or an awake craniotomy with motor and speech mapping with a single surgeon was performed. Three cohorts were compared: 1) asleep LFBS (AsLFBS), 2)

Fathers' perspectives on brain health education and the promotion of dementia risk reduction for their primary school aged children.

BACKGROUND: Fathers occupy a distinctive and influential role in shaping children's developmental trajectories and health-related behaviours. However, their perspectives on brain health education and dementia risk reduction remains largely absent from the literature. Despite growing recognition that modifiable risk factors for dementia operate across the life course, little is known about how fathers view brain health or perceive their role in fostering protective behaviours in their children. The present study seeks to examine how fathers conceptualise brain health, their attitudes towards de

TET1 Inhibition Promotes Therapeutic Sensitivity in TP53-Mutant GBM by Influencing Genome Fragility and Altering TAMs Biology.

TP53 mutations (TP53mut) are associated with therapeutic resistance in glioblastoma (GBM) patients, yet the underlying mechanisms remain incompletely understood. Here, we identified an association between reduced P53 function and increased expression of the epigenetic regulator TET1 in GBM models. In TP53mut GBM cells, TET1 knockdown influenced genome fragility, including DNA damage, senescence, and telomere shortening. Specifically, our findings are consistent with a model in which TET1 binds to the ROS1 promoter and may help maintain ROS1 expression, likely by keeping the promoter in a hypom

Identification of potential biological biomarkers for acute ischemic stroke based on integrated bioinformatics analyses.

Ischemic stroke is a major cause of disability and mortality worldwide, accounting for approximately 66% of all stroke cases. Stroke has been reported to cause approximately 6.5 million deaths annually, and the global disability-adjusted life years attributable to stroke are projected to exceed 200 million by 2030. Therefore, identifying reliable diagnostic biomarkers for acute ischemic stroke (AIS) and exploring their underlying molecular mechanisms are of great importance for early disease recognition and clinical intervention. Transglutaminases (TGases) are widely distributed in the central

Contrast-enhanced ultrasound for reclassification of moderate-risk carotid plaques (Plaque-RADS 3).

BACKGROUND: The Carotid Plaque-RADS system has been proposed to standardize carotid plaque evaluation recently. However, the carotid plaques classified into plaque-RADS 3 share considerable heterogeneity, which poses challenges for clinical management. This research aimed to investigate the supplementary value of contrast-enhanced ultrasound (CEUS) in assessing these moderate-risk carotid plaques designated as plaque-RADS 3. METHODS: A retrospective diagnostic analysis was conducted on 194 patients (270 plaques) who underwent carotid plaque CEUS from January 2020 to May 2024. Plaque features

Machine learning-enabled behavioural and psychological symptoms of dementia management intervention for dementia caregivers: protocol for a hybrid factorial SMART-MRT trial.

BACKGROUND: Behavioural and psychological symptoms of dementia (BPSD) affect over 90% of people living with dementia and are a major contributor to stress and adverse health outcomes among family caregivers. Caregiver‑led behavioural management interventions based on the Antecedents-Behaviour-Consequences (ABC) framework are effective, yet their overall impact is modest. This may be due to the highly individualised and fluctuating nature of BPSD and insufficient adaptive support to sustain engagement. Mobile health interventions offer scalable support; however, engagement and adherence remain

Association between lactate-to-albumin ratio and the short-time mortality of patients with ischemic stroke in intensive care unit.

BACKGROUND: Patients with severe acute ischemic stroke (IS) often face poor outcomes and extended hospital stays. Early prognosis prediction aids in creating personalized treatment plans. The lactate-to-albumin ratio (LAR) is linked to poor outcomes in various illnesses, but its connection to all-cause mortality in ischemic stroke patients is not well understood. This study aims to investigate the prognostic value of LAR in these IS patients in intensive care unit. METHODS: This retrospective cohort study utilized MIMIC-IV (v2.2) data to identify intensive care unit (ICU) patients with acute

Dual targeting of interlocked PTK2B and MAPK signaling triggers synergistic ferroptosis via autophagic flux disruption in BRAFV600E glioma.

BACKGROUND: BRAFV600E mutation is one of the most common oncogenic drivers in gliomas, sharing the highest incidence in pediatric low-grade gliomas (~ 20%) and being frequently associated with poor prognosis. Although combination therapies targeting both BRAFV600E and downstream MAPK signaling have been developed, their efficacy is substantially limited by acquired drug resistance. Consequently, identifying the underlying mechanisms of resistance and novel therapeutic vulnerabilities remains an urgent need. METHODS: Using patient gene expression profiles, immunohistochemistry on glioma sample

Pilot report of an anti-PD-1/VEGF bispecific antibody for treating recurrent H3K27M-mutant diffuse midline gliomas.

BACKGROUND: H3K27M-mutant diffuse midline gliomas (DMGs) are highly aggressive, immunosuppressive tumors that are resistant to conventional therapies. Median overall survival is typically 8-11 months after diagnosis; fewer than 10% of patients survive beyond 2 years, and the 5-year survival rate is generally below 1%. Ivonescimab, a novel bispecific antibody that simultaneously targets PD-1 and VEGF, may help overcome this resistance by restoring T cell immunity and normalizing tumor vasculature. We report the first use of ivonescimab as salvage therapy for recurrent DMG. METHODS: Two patient

Association between cigarette smoking and carotid intima-media thickness among young Saudi adults: a cross-sectional ultrasonographic study.

OBJECTIVES (BACKGROUND): Atherosclerosis is a major contributor to cardiovascular disease and is strongly associated with cigarette smoking. Carotid intima-media thickness (CIMT) is a well-established noninvasive marker of subclinical atherosclerosis; however, limited evidence exists regarding the association between smoking intensity and CIMT among young adults in the Middle East. This study assessed the association between cigarette smoking and CIMT in Saudi adults aged 20-30 years compared with non-smoking. METHODS (SETTING AND DESIGN): This cross-sectional comparative study was conducted
