Multifunctional theranostic nanocarrier platform for microglia-selective cytochalasin D delivery to modulate neuroinflammation after acute brain injuries.

Targeting microglia to modulate neuroinflammation after acute brain injuries (ABIs) is promising but limited by poor blood-brain barrier (BBB) penetration and systemic toxicity of candidate agents. In addition, multifunctional platforms combining targeted therapy with real-time traceability remain limited. We developed and characterized a multifunctional theranostic, folate-functionalized, polydopamine (PDA)-coated mesoporous silica nanoparticle (FA-NP) system for microglia-selective delivery and intracellular release of cytoskeleton inhibitor Cytochalasin D (CytoD). FA-NPs exhibited ideal phy

Case Report: Myelin oligodendrocyte glycoprotein antibody-associated cortical encephalitis masked by peri-ictal magnetic resonance imaging abnormalities.

One phenotype of cerebral cortical encephalitis in myelin oligodendrocyte glycoprotein (MOG) antibody-associated disease (MOGAD) is FLAMES (fluid-attenuated inversion recovery-hyperintense lesions in anti-MOG-associated encephalitis with seizures). FLAMES is characterized by its imaging features and epileptic seizures. We present a case in which these findings were masked by peri-ictal magnetic resonance imaging abnormalities (PMAs) associated with status epilepticus, making differentiation difficult. A middle-aged woman presenting with persistent right upper limb weakness and aphasia was admi

Impact of Obstructive Sleep Apnea on Blood-Brain Barrier Permeability and Membrane Proteins.

The blood-brain barrier (BBB) carefully regulates the transport of molecules between the blood and the nervous tissue. Obstructive sleep apnea (OSA) is a chronic condition that induces structural and functional changes in the BBB. These include alterations in its permeability and in protein expression in the BBB capillary endothelium. Moreover, OSA alters blood levels of transcription factors, such as hypoxia-inducible factor 1 (HIF-1), which are suggested to be responsible for these changes. As a review article, this paper focuses on the most significant effects of OSA on the BBB, including d

Integrated Identification of NAD⁺ Metabolism-Associated Candidate Genes.

Migraine is one of the most disabling neurological diseases globally; however, a significant proportion of patients do not respond to current treatments, or treatment efficacy diminishes over time, highlighting the need to identify additional molecular candidates for further investigation. Recent studies have increasingly focused on the role of neuro-immune interactions in migraine. This study aimed to identify peripheral immune-related candidate genes associated with NAD+ metabolism-related transcriptional features in migraine through integrated bulk and single-cell transcriptomic analyses. Increased impulsivity and higher odds of compulsive shopping among cabergoline-treated patients with prolactinoma: a case-control study.

UNLABELLED: Dopamine agonists, particularly cabergoline, are the first-line treatment for prolactinomas, but have been associated with impulse control disorders (ICDs). However, data on impulsivity and related behaviors in this population remain limited and inconsistent. OBJECTIVE: To evaluate impulsivity and ICDs in patients with prolactinoma treated with cabergoline and to compare these findings with healthy controls. METHODS: This case-control study included 131 patients with prolactinoma receiving cabergoline and 131 healthy controls. Impulsivity was assessed using the Barratt Impulsiven

Arachnoid granulation morphologies and β-amyloid and tau pathology in older adults.

INTRODUCTION: Arachnoid granulations (AG) prominently enlarge with age, yet limited studies evaluate AG in neurodegeneration. Here, we investigate the associations of AG morphologies with Alzheimer's disease (AD). METHODS: Macroscopic AG properties were evaluated along dorsal post mortem brain specimens (n = 882). Regression models were used to analyze their associations with AD neuropathology and dementia, adjusting for demographics. RESULTS: Participants died at mean age of 90.62 (SD = 6.98) years. β-amyloid (odds ratio [OR], 0.79 [95% confidence interval {CI}, 0.66-0.95]) and neurofibrill

Spared Motor Neurons Enable Control of a Robotic Sixth Finger for Functional Grasping in Tetraplegia.

Restoring hand function is a top priority in spinal cord injury (SCI), yet current assistive technologies remain limited in usability and lack intuitive control. We developed a non-invasive neuromechatronic interface that translates spared motor neuron activity from individuals with tetraplegia into the motion of a supernumerary robotic sixth finger. Three individuals with chronic, motor complete cervical SCI (C5-C6 level, > 8 years post-injury), unable to voluntarily move their fingers, participated in the study. High-density surface electromyography (HDsEMG) signals were recorded from paraly

Targeting white matter astrocytes with a Gfap-disruptive Cas9 nuclease benefits a murine model of Alexander disease.

Astrocytes are intriguing therapeutic targets due to their crucial role in initiating and promoting demyelination in the central nervous system. By efficient on-target editing, we can permanently disrupt detrimental genes and restore normal physiological processes. Here, we have optimized an AAV platform to deliver the CRISPR/Cas9 system to white matter astrocytes. A single neonatal intra-CSF administration of an all-in-one AAV-DJ8 vector expressing the Staphylococcus aureus Cas9 enzyme under an astrocyte-specific promoter resulted in effective downregulation of Glial Fibrillary Acidic Protein

Endoscopic Craniofacial Resection for Anterior Skull Base Tumors: A Singapore Center Review.

INTRODUCTION: Anterior skull base (ASB) tumors are rare and comprise a variety of pathologies. In recent years, the surgical management of ASB tumors has undergone a paradigm shift from open craniofacial resection (CFR) to minimally invasive endoscopic approaches. Endoscopic-assisted (EaCFR) and entirely endoscopic (ECFR) CFR have shown equivalent oncological outcomes without the morbidity of open surgery. This study presents our tertiary center experience and outcomes with endoscopic approaches for ASB tumors. METHODS: A retrospective review of all patients who underwent EaCFR and ECFR for A

The positive feedback interaction between mast cell-derived tryptase and NETs formation in the pathogenesis of sepsis-associated encephalopathy.

BACKGROUND: Sepsis-associated encephalopathy (SAE) is a serious neurological complication resulting from sepsis, marked by considerable neuroinflammation and deficits in cognitive function. While neutrophil extracellular traps (NETs) have been implicated in various neuroinflammatory conditions, the specific mechanisms underlying NETs formation in SAE, particularly the role of brain-resident mast cells (MCs) and their interplay with neutrophils, remain poorly understood. METHODS: The cecal ligation and puncture (CLP) procedure was employed to create the SAE model in male C57BL/6 mice. Comprehe
