Widespread use of nitroimidazoles in competition pigeons reduced their effectiveness against Trichomonas gallinae.

Trichomonas gallinae is a protozoan parasite of major concern in avian medicine, particularly in domestic pigeons (Columba livia). This study investigated the nitroimidazole susceptibility and T. gallinae occurrence in domesticated pigeons from Eastern Spain, kept for different competitions. A total of 220 pigeons from 11 lofts were sampled and examined by microscopy and culture, revealing 63.6% of infected birds. Genotyping identified genotype C as predominant, with occasional detection of genotype A, mixed A/C infections, and one isolate of Lineage III. In vitro susceptibility testing of 42

Cannonballs in Trichomonas vaginalis Infection: Morphologic Evidence of Parasite-Associated Neutrophilic Aggregates.

Cannonballs, compact aggregates of neutrophils observed in Papanicolaou (Pap) smears, are frequently associated with Trichomonas vaginalis (T. vaginalis) infection but are generally regarded as nonspecific inflammatory findings. To clarify their morphologic features, we analyzed cervicovaginal liquid-based cytology specimens from 29 cervicitis cases, including six positive for T. vaginalis. Cannonballs were evaluated using immunocytochemistry for T. vaginalis and cytokeratin, cell block analysis, and morphometric analysis, with negative cases as controls. All positive cases contained T. vagina

AI-Assisted Diagnosis of Trichomonas vaginalis from Routine Gram-Stained Vaginal Smears.

Background/Objectives: Trichomonas vaginalis is one of the most prevalent non-viral sexually transmitted infections worldwide. Although Gram staining is routinely performed in clinical microbiology laboratories for the evaluation of vaginal samples, it is not considered a diagnostic method for T. vaginalis, which represents a missed diagnostic opportunity in routine practice. This study aimed to evaluate an artificial intelligence (AI)-assisted diagnostic approach for the identification of T. vaginalis directly from routine Gram-stained vaginal smears. Methods: A retrospective dataset of Gram-

Trichomonas vaginalis pseudocysts are a quiescent cell stage characterized by a differentially regulated transcriptome and intact membranes.

Trichomonas vaginalis is the causative agent of trichomoniasis, the most common non-viral sexually transmitted infection (STI). Despite its prevalence, low levels of public knowledge and research funding and the absence of T. vaginalis screening or control programs have led to its categorization as a "neglected" STI. Unlike other STIs in the USA, prevalence increases with age, peaking among individuals in their 40s. Both a motile trophozoite stage and a non-motile pseudocyst state have been described for the parasite, although it is debated whether the latter is a quiescent stage or a degenera

Trichomonas vaginalis-induced inflammation is associated with Th17 differentiation and tumor-intrinsic IL-17R signaling in prostate cancer progression.

OBJECTIVE: Chronic inflammation associated with infection has been implicated in prostate cancer (PCa) progression, yet the immunological mechanisms linking sexually transmitted pathogens to tumor development remain incompletely understood. Trichomonas vaginalis (Tv), the most prevalent nonviral sexually transmitted parasite worldwide, has been associated with prostatic inflammation and increased PCa risk. However, how Tv-induced inflammation influences tumor progression has not been clearly defined. Here, we investigated whether Tv-induced epithelial inflammation promotes Th17 differentiation

Acceptability and feasibility of one-stop home-based genital self-sampling for female genital schistosomiasis, human papilloma virus and self-testing for trichomonas and HIV: the Zipime Weka Schista study in Zambia.

BACKGROUND: Female genital schistosomiasis (FGS) is a neglected gynaecological disease that affects 56 million girls and women in sub-Saharan Africa. It is caused by the waterborne parasite Schistosoma (S.) haematobium and has been associated with HIV infection, with human papillomavirus (HPV) and cervical precancer. FGS alters the normal sexual and reproductive health of girls and women. Diagnosis and screening are bottlenecked, but previous studies have shown good acceptability of genital self-sampling at home for individual diagnosis of FGS, HPV and selected STIs. Here, we aim to determine

Genotyping of Trichomonas vaginalis in rural Vellore, South India.

INTRODUCTION: Trichomonas vaginalis (T.vaginalis) is a globally prevalent sexually transmitted protozoan parasite associated with a wide spectrum of clinical manifestations ranging from asymptomatic infections to malodorous vaginal discharge, cervicitis, with an increased risk of cervical cancers. Molecular characterization based on actin gene polymorphism has enabled classification of T. vaginalis into distinct genotypes, facilitating exploration of genotype-specific clinical associations and geographic distribution patterns. However, data on circulating genotypes from India are lacking. This

Trichomoniasis associated with amniotic sac rupture in pregnancy: a case report.

The aim of this report is to present a rare case of pre‑labour premature rupture of membranes (PPROM) in a patient in the second trimester of pregnancy, in which an association with a lower genital tract infection caused by Trichomonas vaginalis was observed. The patient was a 26‑year‑old woman, gravida 2, at 25 weeks of gestation, who presented with clinical complaints of abnormal vaginal discharge and fluid leakage.Following a definitive diagnosis of trichomoniasis based on stained smear and culture findings, standard treatment with metronidazole was initiated. The treatment was effective, l

Leveraging current capacity to address the high prevalence of Chlamydia trachomatis, Neisseria gonorrhoeae, and Trichomonas vaginalis in South Africa: Modelling potential costs and benefits of near point-of-care GeneXpert testing for STIs.

South Africa has high sexually transmitted infections (STIs) prevalence and currently implements syndromic management, which has limitations including untreated asymptomatic infections and excess antibiotic use. Diagnostic tools like GeneXpert may offer potential improvements. We evaluated potential costs and benefits of GeneXpert for STIs testing. We developed a static analytical model using existing data over a one-year time horizon (January-December 2024) from a provider perspective. We estimated costs (2024 USD), outcomes, cost-effectiveness, and budget impact of syndromic management and m

Sexually transmitted infection prevalence following azithromycin mass drug administration for trachoma in Nauru: a before-and-after cross-sectional survey.

BACKGROUND: Sexually transmitted infections (STIs) are a major public health problem globally, particularly in low and middle-income countries. In the Western Pacific region, including Nauru, STIs are highly prevalent. Syndromic management misses a high proportion of infections and screen-and-treat strategies are not currently feasible at scale in these settings. Mass drug administration (MDA), widely used for control of neglected tropical diseases, may provide an alternative approach to the population control of STIs. METHODS: We used a before-and-after study design to evaluate the impact of

Molecular Diagnosis of Trichomonas vaginalis in Women and Men: A Systematic Review.

BACKGROUND: Trichomonas vaginalis (T. vaginalis) is a human protozoan parasite and the leading cause of nonviral sexually transmitted infections worldwide. Diagnosis of T. vaginalis is typically performed using vaginal discharge or urine samples, which are examined via wet prep, culture methods, or molecular tests. Compared to traditional culture-based methods, molecular diagnostic techniques have shown higher sensitivity, as they allow more precise identification of T. vaginalis-infected individuals. The present review evaluated the molecular diagnosis of T. vaginalis in male and female popul

Study of microbe-microbe interactions between the sexually transmitted parasite Trichomonas vaginalis with the cervicovaginal bacteria Lactobacillus iners.

Trichomonas vaginalis is the leading cause of non-viral sexually transmitted infections and it is associated with comorbidities that affect female health. Lactobacillus iners is one of the most predominant bacteria in the cervicovaginal microbiome. As so, both microbes are likely to encounter one another upon T. vaginalis infection. To our knowledge, the interaction of both microbes has not been previously investigated. Here, we report that T. vaginalis and L. iners bind to one another at early time points of co-incubation. Using imaging flow cytometry and scanning electron microscopy, we capt

Machine Learning Classification Models for the Design of Novel Nitroimidazole Derivatives with Anti-Trichomonas vaginalis Activity.

Trichomonas vaginalis is the pathogen responsible for trichomoniasis, a widespread sexually transmitted infection worldwide. Treatment currently relies solely on nitroimidazole-class drugs, such as metronidazole and tinidazole, to which resistance is increasingly reported. When resistance or treatment failure occurs, higher doses are prescribed, which in turn heightens the risk of toxic side effects. Therefore, discovering new therapeutic options is urgently needed. To address this, quantitative structure-activity relationship (QSAR) models were developed using nitroimidazole derivatives to an

Cost-Effectiveness of Testing and Screening Strategies for Trichomoniasis among Asymptomatic and Symptomatic Women in U.S. Outpatient Clinical Settings.

BACKGROUND: T. vaginalis, the most common non-viral sexually transmitted infection among women, is associated with multiple adverse health outcomes. Screening asymptomatic women is not recommended except among HIV-infected women. We aimed to evaluate the cost-effectiveness of T. vaginalis testing and screening strategies among asymptomatic and symptomatic women, respectively, in U.S. primary care settings. METHODS: We developed a decision tree model to evaluate 11 testing/screening strategies (using combinations of wet mount microscopy, rapid antigen testing, and T. vaginalis nucleic acid amp

Multiple Strains of Trichomonas gallinae are Widespread in Reservoir Hosts and Environmental Resources in UK Farmland.

Understanding transmission routes of parasites and pathogens is critical for the development of control measures and mitigation of impacts on host populations. Trichomonas gallinae is a widespread parasite in columbids worldwide, and has emerged as a lethal disease in passerines, particularly finches. The postulated transmission route from columbids to passerines via spillover at shared resources has not yet been empirically tested. Here, we screen 363 birds (261 columbids and 102 non-columbids) in the UK and France for the presence and strain identity of T. gallinae. To quantify potential env

Refining Trichomonas vaginalis treatment in women and men: protocol for an open-label randomised comparison of multi-dose oral metronidazole versus single-dose oral secnidazole.

INTRODUCTION: Trichomonas vaginalis is estimated to be the most common non-viral sexually transmitted infection (STI) worldwide with 156 million new cases each year. In 2021, the United States Centres for Disease Control and Prevention (CDC) updated their STI Treatment Guidelines to recommend multi-dose oral metronidazole (MTZ) for all T. vaginalis-infected women. Although multi-dose oral MTZ 500 mg twice daily was found to be superior to single-dose 2 g oral MTZ in multiple trials in women, multi-dose oral MTZ still had unacceptably high rates of breakthrough infection (9%-11%) at test-of-cur
