A genetic variant of BAFF is associated with the risk of lymphoma in Sjögren disease.

OBJECTIVES: B-cell activating factor (BAFF) of the Tumors Necrosis Factor (TNF) family is involved in the pathogenesis of Sjögren's disease (SjD). A functional variant (BAFF variant [BAFF-var]) of the BAFF gene (TNFSF13B), leading to increased levels of BAFF, has been described. The aim of this study was to investigate the association between BAFF-var and clinical phenotype and risk of SjD. METHODS: A case-control study including cases from Paris Saclay and the Assessment of Systemic Signs and Evolution in Sjögren's syndrome (ASSESS) cohort and controls (blood donors from Etablissement França

Safety and effectiveness of rituximab therapy in Sjögren's disease: a PORTRESS - the Portuguese registry of Sjögren's disease-based study.

INTRODUCTION: Sjögren's Disease (SjD) is an immune-mediated disorder characterised by lymphocytic infiltration of exocrine glands and, frequently, systemic involvement. Although B-cell hyperactivity provides a rationale for using rituximab (RTX) therapy, randomised clinical trials have failed to demonstrate consistent efficacy, while real-world data suggest benefits in systemic manifestations. OBJECTIVES: To evaluate the effectiveness and safety of RTX in patients with SjD registered in PORTRESS, the Portuguese Registry of Sjögren's disease. METHODS: We conducted a multicentre, retrospective

Comparative efficacy of pranoprofen with sodium hyaluronate or polyvinyl alcohol in postoperative dry eye and corneal endothelial function.

BACKGROUND: Dry eye following eye surgery represents a common obstacle to recovery and general life satisfaction. Pranoprofen, an anti-inflammatory drug, frequently co-administered with adjuvants such as sodium hyaluronate and polyvinyl alcohol, represents a common response to dry eye. The relative efficacy of pranoprofen and sodium hyaluronate compared with pranoprofen and polyvinyl alcohol as a treatment for dry eye and as an aid to corneal endothelial function will be ascertained. OBJECTIVES: To assess how pranoprofen with sodium hyaluronate (Group B) compares with pranoprofen with polyvin

Deciphering the molecular landscape of Sjögren's disease, mucosa-associated lymphoid tissue lymphoma, and thyroid cancer: unraveling the complexities of disease mechanisms and diagnostic biomarkers.

BACKGROUND: Sjögren's disease (SjD), mucosa-associated lymphoid tissue lymphoma (MALT lymphoma), and thyroid cancer (THCA) are clinically distinct yet immunologically intertwined diseases characterized by chronic inflammation and aberrant immune activation. However, the shared molecular basis linking autoimmunity and oncogenesis remains poorly defined. METHODS: We performed an integrative transcriptomic analysis across three datasets (TCGA-THCA, GSE40611/GSE127952 for SjD, and GSE25638 for MALT lymphoma), totaling 749 samples. Analyses included differential expression, Gene Ontology (GO)/Kyot

Impact of Diabetic Peripheral Neuropathy on Corneal Sensitivity and Ocular Surface.

INTRODUCTION: Diabetes mellitus (DM) is frequently associated with microvascular complications, including diabetic peripheral neuropathy (DPN). The cornea, one of the most densely innervated tissues in the body, has been proposed as a surrogate marker for small fiber neuropathy. Patients with DM often present with ocular surface disease (OSD) and corneal sensitivity impairment, but the relationship between DPN, ocular surface alterations, and inflammatory biomarkers remains unclear. This study aimed to evaluate ocular surface parameters between patients with type 2 DM with and without DPN and

Immune thrombocytopenia in patients with connective tissue disease.

Immune thrombocytopenia (ITP) is a secondary autoimmune complication that can develop in patients with connective tissue diseases (CTDs). Its pathogenesis involves autoantibody-mediated platelet destruction and immune complex deposition, constituting a distinct clinical manifestation of CTD-related autoimmune abnormalities in the hematologic system. Among the subtypes of CTD-related immune thrombocytopenia (CTD-ITP), systemic lupus erythematosus (SLE) and primary Sjögren's syndrome (pSS) are the most prevalent. CTD-ITP occurs within the unique clinical context of being a secondary condition to

A Narrative Review of Salivary Gland Extracellular Matrix and Sjögren's Syndrome: Research Status and Future Prospects.

Sjögren's syndrome (SS) is a chronic systemic autoimmune disease characterized by the main clinical manifestation of oral and ocular dryness, predominantly affecting middle-aged and elderly women. As the most commonly affected target organs in SS, pathological changes in the salivary glands (SGs) and their underlying mechanisms are of great significance for understanding the disease progression. Recent studies have revealed that a dynamic imbalance of the extracellular matrix (ECM) in the SGs plays a crucial role in the pathogenesis of SS. Dysregulation of matrix metalloproteinases (MMPs) and
