Treatment of Graves' disease (GD) is complex and controversial. Conventional therapy of GD includes antithyroid drugs (ATDs), surgery, and radio-iodine therapy (RIT) and each has limitations. ATDs are associated with high rates of relapse and side effects. Definitive therapies result in permanent hypothyroidism and increased risk of Graves' ophthalmopathy. With advancements in the understanding of precise autoimmune mechanisms causing GD, novel therapies targeting immune pathways are in the pipeline of development. Newer drugs switch off a part of the immune system that is causing the disease,

The prevalence of thyroid dysfunction is rising in India and thus so is Graves' disease (GD). The phenotype of Indian GD is different than in Caucasians and is characterized by lower age at onset, lower female to male ratio, delayed diagnosis, more signs and symptoms, lower body mass index (BMI) and weight, lower prevalence of Graves' orbitopathy (GO), more inactive GO, lower doses of carbimazole being more effective, different markers of remission, and other things. Mechanistic studies are urgently needed to characterize the pathophysiology of Indian GD. Prevention, early diagnosis, and adequ

While it is well known that hyperthyroidism is associated with decreased bone mineral density, other Dual Energy X-ray Absorptiometry (DXA) derived indices, such as proximal hip geometry and body composition parameters, have not been well studied in India. This study was undertaken to comprehensively assess DXA-derived parameters and bone turnover markers in premenopausal women aged 25-45 years, and to compare these with age- and BMI-matched female controls. This was a case-control study done over a period of one year. Premenopausal women aged 25-45 years with a diagnosis of Graves' disease we

Autoimmune-associated orbital inflammation refers to the inflammation of orbital tissues resulting from immune system dysregulation. Interleukin-17 (IL-17) plays a critical role in immune defense, tissue repair, inflammation, and tumor progression. Given its immunomodulatory functions, this study aimed to investigate whether single-nucleotide polymorphisms (SNPs) in IL17-related genes contribute to susceptibility and clinical manifestations of orbital autoimmune diseases. A total of 60 patients with orbital autoimmune disease and 60 healthy controls were recruited. Candidate SNPs in IL17A, IL1

Thyroid-associated ophthalmopathy (TAO), the most common orbital disease in adults, is a specific autoimmune condition closely associated with thyroid dysfunction, primarily affecting orbital fat and extraocular muscles. Multiparametric magnetic resonance imaging (MRI) holds unique value in diagnosing TAO, staging disease activity and progression evaluation by providing multidimensional information on tissue morphology, edema, fat infiltration, and fibrosis. This review examines conventional and advanced MRI techniques-such as T1WI, T2WI, diffusion-weighted imaging, dynamic contrast-enhanced M

INTRODUCTION: Thyroid eye disease (TED) is an autoimmune-mediated inflammatory condition of the orbit leading to hypertrophy and inflammation of the orbital extra-ocular muscles (EOMs). Previous studies have sought to quantify this enlargement, but due to the anatomical proximity and limitations in imaging quality, have measured the superior rectus (SR) and Levator Palpebrae Superioris (LPS) complex together rather than measuring the individual muscles.
METHODOLOGY: Retrospective manual measurements from high-field (3-Tesla) fat-suppressed contrast-enhanced T1-weighted magnetic resonance imag

BACKGROUND: Significant progress has been made in the management of thyroid eye disease (TED), based on the elucidation of important pathogenic mechanisms. This has led to novel therapeutics validated in randomized clinical trials. Autoreactive antigens that elicit specific orbital immune reactions have not yet been identified, although it has been shown that fibrocytes, circulating stem cells that differentiate into fibroblasts, are expressing the thyroid-stimulating hormone receptor (TSHR) and insulin-like growth factor-1 receptor (IGF-1R) and may be stimulated in the orbit by a cascade of i

This case report characterizes the molecular pathology of two synchronous IDH mutant gliomas in a 28-year-old female patient. The patient exhibited symptoms of dizziness, retro-orbital pain, headache, and numbness with paresthesia in her right arm. MRI imaging revealed two distinct non-enhancing T2/FLAIR hyperintense lesions in the left frontal and parietal lobes. Histopathologic and molecular analyses, including whole exome sequencing, were performed on the resected specimens from each location. The left parietal tumor was diagnosed as a grade 4 astrocytoma with an IDH1 R132H mutation, while

Graves' disease (GD) is a common autoimmune thyroid disorder and is often accompanied by Graves' orbitopathy (GO), an inflammatory eye disease that can significantly reduce the quality of patients' life. Despite understanding of GD and GO has progressed, the mechanisms driving disease progression remain incompletely defined. Emerging evidence highlights extracellular vesicles (EVs), particularly exosomes, as important mediators of immune regulation and tissue remodeling in autoimmune disorders, including GD and GO. This review summarizes current knowledge of EVs biogenesis and molecular compos

OBJECTIVES: Thyroid eye disease (TED) is a debilitating autoimmune condition frequently associated with Graves' disease (GD). Both disorders may impair glucose metabolism, yet the risk of hyperglycemic events and complications remain unknown. We evaluated the incidence, prevalence, and predictors of these outcomes in patients with TED or GD versus the general population (GP).
METHODS: This retrospective cohort study using the Merative MarketScan Research Claims Database (2014-2019) identified adults (≥18 years) in TED, GD, or GP cohorts via diagnostic codes. The primary outcome was a composit

The first 1000 days of a child's life represent a critical window for brain development, during which nutrition exerts profound effects on the trajectories of neurodevelopment. Human Milk Oligosaccharides (HMOs), a major component of human milk, are largely indigestible by infants and are known to influence immunity, microbiome composition, and gut-brain signaling, but their direct role in neurodevelopment remains poorly understood. Here, we investigated the impact of HMOs on human cortical organoids, a physiologically relevant in vitro model of early brain development. We found that HMO treat
