Evaluating the impact of triple-drug mass drug administration using human and mosquito-based indicators of lymphatic filariasis in Samoa, 2019 - 2024.

BACKGROUND: For lymphatic filariasis (LF) elimination, the World Health Organization recommends multiple rounds of mass drug administration (MDA) to interrupt transmission. While LF antigen (Ag) is routinely used to monitor progress, recent evidence suggests more time-sensitive indicators are needed during the immediate post-MDA period. In Samoa, triple-drug MDA was distributed in 2018 and 2023. This study aimed to evaluate the impact of Samoa's second round of triple-drug MDA on human and mosquito-based indicators of transmission. METHODOLOGY: Surveys were conducted in eight primary sampling

GCMS profiling and larvicidal potentials of the extracts of Terminalia catappa leaf, Eucalyptus citriodora seed and stem against malarial, dengue and filarial vector mosquitoes (Diptera: Culicidae).

The worldwide proliferation of vector-borne infections, coupled with advancing insect defense mechanisms against standard synthetic pesticides, compels the exploration of ecological, vegetation-derived countermeasures. This investigation sought to characterize the secondary metabolite distribution, prominent chemical fractions, and destructive impact of Terminalia catappa leaf ethyl acetate extract (TLEE), alongside Eucalyptus citriodora stem ethyl acetate extract (LSEE) and seed ethyl acetate extract (LEEE), against Aedes aegypti, Anopheles gambiae, and Culex quinquefasciatus. Retroperitoneoscopic lymphatic disconnection for non-filarial chyluria in Vietnamese adults: a case series.

INTRODUCTION: Retroperitoneal laparoscopic surgery is a widely applied technique in the management of renal, adrenal, ureteral, and retroperitoneal lymphatic disorders. A representative condition is chyluria, a rare disease caused by a lymphatic-urinary fistula, most commonly involving the ureter and renal pelvis. The retroperitoneal approach provides direct access to the affected structures while minimizing intra-abdominal manipulation, thereby reducing the risks of bowel adhesions and visceral injury. This technique enables ligation, excision, or suturing of abnormal lymphatic vessels, poten

Tick-borne diseases in French Guiana: A comprehensive review pinpointing differences from mainland France.

French Guiana (FG), an Amazonian French territory, represents a unique ecological and epidemiological setting for tick-borne diseases (TBDs). In contrast to mainland France, where TBDs are well-documented, data from FG remain scarce. We carried out a systematic review using the PRISMA criteria for our search of the PubMed, ScienceDirect, and SciELO data bases. We identified 68 articles, of which 43 met our inclusion criteria. Data from FG were compared with those originating from mainland France. Albeit highly prevalent in Europe, and notwithstanding detection of a distinct Borrelia species in

Human filarial infection reshapes CD8+ T-cell transcription and function at homeostasis and following viral activation.

The role of CD8+ T cells in filarial infections remains poorly understood. Given the chronic nature of these infections, individuals living in endemic regions are frequently exposed to other pathogens including viruses. Because CD8 cells are essential for antiviral immunity, understanding how filarial infections shape the cytotoxic compartment is essential to elucidate their impact on bystander immunity. We evaluated the phenotype and function of CD8+ T cells from Loa loa-infected (Fil+) and -uninfected (Fil-) individuals at baseline and following cytomegalovirus (CMV) re-stimulation. Filarial

Spatial Clustering of Lymphatic Filariasis in Relation to Potential Mansonia Mosquito Habitats in Endemic Communities of Belitung Regency, Indonesia.

BACKGROUND: Lymphatic filariasis (LF) remains endemic in several island communities in Indonesia despite mass drug administration (MDA) campaigns. Investigating the spatial relationship between human cases and potential mosquito breeding sites may help explain the persistence of LF transmission. METHODS: A cross-sectional survey was conducted in 2019 across four sub-villages in two LF-endemic villages in Belitung Regency, Indonesia: Suak Gual village (Suak Gual sub-village) and Lasar village (Batumana, Dudat, and Ulin sub-villages). Thick blood smears were collected at night from 671 particip

Correction: Transplantation of older DCD livers in the machine perfusion era: a U.S. cohort study.

The spread of dirofilariosis, as a newly emerging zoonosis, due to global changes has been documented in several Central European countries. In Slovakia, the first autochthonous case of dirofilariosis was recorded in dogs in 2005. The first case of human subcutaneous dirofilariosis caused by Dirofilaria repens was diagnosed in a 60- year-old patient from an area of western Slovakia. We report another case of dirofilariosis in a 37-year-old woman living in the same area of south-western Slovakia. The infection manifested as a painful, inflamed nodule on the back of the left hand, containing an

Pan filarial-nested PCR with gelified tubes: a new approach towards point-of-care testing for human filariasisFIRST REVIEW ROUND - REVIEWERS' COMMENTSREVIEWERS' COMMENTS.

BACKGROUND: In recent years, traditional point-of-care tests (POCTs) for diagnosing human filariasis have demonstrated substantial limitations, raising concerns regarding their reliability in the current epidemiological context, in which numerous countries are approaching the elimination of human filariasis. Consequently, there is a need for new molecular tests that allow the simultaneous and reliable detection of multiple human filarial species, even in cases of very low microfilaremia. OBJECTIVES: The objective of this study was to convert the Fn-PCR from its original manual format to the n

Developing a scoring tool for assessing adherence to morbidity management and disability prevention (MMDP) practices in patients with filarial lymphedema: A pilot study.

BACKGROUND: Morbidity Management and Disability Prevention (MMDP) practices are essential in mitigating the progression of lymphedema and in preventing episodes of adenolymphangitis. Despite their importance, patients' adherence to these prescribed practices has frequently been found to be suboptimal. Currently, there is a lack of a standardized tool for the objective assessment of patients' adherence to MMDP practices in patients with filarial lymphedema. This study was undertaken to develop a scoring system for assessing patients' adherence to MMDP and to validate this among patients attendi

Microfilaremic loiasis is associated with T cell hyporesponsiveness against SARS-CoV-2.

BACKGROUND: Loiasis is a chronic filarial infection endemic to Central and West Africa. Although long considered benign, increasing evidence links loiasis to substantial morbidity and mortality. The infection is associated with immune modulation, including Th2-skewed responses and elevated regulatory cytokines. Clinically, loiasis is classified as microfilaremic (presence of circulating microfilariae) or amicrofilaremic ("occult") disease, the latter defined by a history of eyeworm migration without detectable microfilaremia. This study investigated how chronic L. loa infection influences anti

Stomoxys as vectors of Stephanofilaria dinniki in rhinoceros.

We took advantage of rhinoceros translocations and entomological studies in the 1990s to elucidate some aspects of the biology of Stephanofilaria in Kenya, which remains largely undocumented. Blood samples from 88 black and 27 white rhinos were screened for microfilariae during routine haematology. Microfilariae matching S. dinniki were found in three individual black rhinos on single occasions and were not found in white rhinos; the parasite was not transmissible to tsetse or laboratory mice. Thousands of proboscides of 19 species of Stomoxyinae and other Diptera were dissected from Nairobi N

Secreted filarial acetylcholine analogs modulate parasite reproduction and host physiology.

Lymphatic filariasis, caused by Brugia malayi, leads to chronic lymphatic dysfunction and lymphedema. The molecular mechanisms underlying interactions between filarial worms and their hosts remain poorly understood. Using untargeted metabolomics, we identify previously unknown acetylcholine analogs synthesized from leucine and isoleucine that are secreted by Brugia adult males. In the mammalian host, these metabolites selectively activate host M2 muscarinic acetylcholine receptors, alter lymphatic endothelial cell integrity, and trigger constriction of lymphatic vessels. These metabolites adde

A Preliminary Investigation on the Utility of Embedding Fluency-Building Strategies within the TAGteach Procedure.

UNLABELLED: TAGteach is a trademarked intervention package designed to promote skill acquisition by combining the use of an audible stimulus as immediate feedback with several other behavior analytic procedures. Although TAGteach effectively improves the accuracy of sports performance, previous researchers have noted that TAGteach does not reliably promote synchronized, coordinated (i.e., fluent) sport movements. To date, no study has evaluated the efficacy of embedding fluency-building strategies within the standard TAGteach procedure. We used a concurrent multiple baseline across skills desi

In silico DNA barcoding surpasses whole genome sequencing for species identification from vector surveillance pools.

Mosquito-borne diseases are responsible for over 600,000 deaths annually, mainly in sub-Saharan countries. Mosquito surveillance is a crucial element of vector control programmes to assure they remain effective. This study focused on optimizing the use of the MinION sequencer for interrogating mosquito pools from surveillance programmes, with simplified bioinformatic workflows for use in African laboratories nearer to the field. Mosquito pools were created using different human disease vector species, mimicking possible field trap contents. Some pools were spiked with DNA from the malaria para

Molecular detection of filarial parasites in black flies (Diptera: Simuliidae) in Laos.

Filarial parasites cause diseases in humans and other animals and are transmitted primarily by hematophagous Diptera, including black flies (Simuliidae). However, knowledge of the vectorial role of black flies in transmitting filarial parasites in tropical Asia is limited. We used molecular approaches to detect filarial parasites in two common black flies, Simulium asakoae and S. khelangense, in Laos. Among 483 black flies, 15 were infected with filarial parasites. Based on sequence similarity analyses of the COI and 12S rDNA genes, the parasites were closely related to two filarial taxa: one

"String sign"- acute lymphangitis in the forearm following triple drug therapy for bancroftian filarial infection.

Localized adverse events following mass drug administration for lymphatic filariasis elimination are often selflimiting. However, they are of significant concern to the participants and may affect community drug compliance if not managed appropriately. Here, we report a rather uncommon adverse event, the 'string sign' in a south Indian woman with bancroftian filarial infection treated with the triple drug regimen (IDA: ivermectin, diethylcarbamazine and albendazole). String sign is the manifestation of acute lymphangitis as a cordlike subcutaneous swelling due to the swollen and inflamed lymph

In cold blood: screening wild Alaskan moose (Alces alces gigas) for filarial nematode (Filarioidea: Onchocercidae) infections using a deep-amplicon sequencing approach.

Moose are significant ecological, economical and cultural animals for the stakeholders of Alaska, USA. Thus, the impact of pathogens, like filarial nematodes, is a critical area of moose research. These vector-borne parasites, including Setaria yehi, and Rumenfilaria andersoni, can lead to severe health consequences (e.g., peritonitis). However, little is known about filarial nematode distribution, diversity and its associated life cycle with Alaskan moose hosts. Newly developed next-generation sequencing techniques offer the ability to efficiently screen multiple species of co-infecting filar

Fulminant septic shock from clinically occult deep soft tissue infection in chronic lymphedema: A clinicopathological correlation.

Chronic lymphedema is a well-recognized risk factor for recurrent cellulitis. However, deep soft tissue infections in lymphedematous limbs may present without classical cutaneous signs, leading to delayed diagnosis and rapid clinical deterioration. We report a 35-year-old woman with chronic filarial lymphedema and prior recurrent cellulitis who presented with a three-day history of fever, myalgia, and arthralgia without localizing symptoms. Initial examination revealed no evidence of cellulitis. She rapidly developed severe neutropenia, thrombocytopenia, acute kidney injury, severe metabolic a

Filarial Nematode Infestation Presenting as Subcutaneous Swellings in a Teaching Hospital in Kerala.

Filarial nematode infection presenting as subcutaneous swellings is uncommon and often mimics other benign subcutaneous swellings. While individual cases are documented, few case series explore this specific presentation. This study aims to examine patients with histopathologically or radiologically confirmed filarial nematode infection manifesting as subcutaneous swellings of various parts of the body. A retrospective analysis was conducted at a teaching hospital in Kerala from January 2015 to December 2020, reviewing all excised subcutaneous swellings confirmed as nematode infection. Since f
