Advanced machine learning approaches for predicting Neglected Tropical Disease co-endemicity in Kenya: A focus on soil-transmitted helminths, schistosomiasis, and lymphatic filariasis.

BACKGROUND: Neglected Tropical Diseases (NTDs) affect 1.5 billion people worldwide with 39% of the global burden occurring in Africa. In Kenya, NTDs remain endemic despite control efforts, with co-endemicity of soil-transmitted helminths (STH), schistosomiasis (SCH), and lymphatic filariasis (LF) complicating intervention strategies. This study developed machine learning models to predict high-risk co-endemic areas using demographic and Water, Sanitation, and Hygiene (WASH) indicators. METHODOLOGY: The study analyzed Kenya's 2022 NTD co-endemicity data from the Expanded Special Project for El

Efficacy of a novel chewable tablet (Credelio Quattro™) containing lotilaner, moxidectin, praziquantel, and pyrantel for the prevention of heartworm disease (Dirofilaria immitis) in dogs.

BACKGROUND: Canine heartworm (Dirofilaria immitis), a filarial parasite endemic in many countries worldwide, can cause life-threatening disease in dogs if left untreated. Two laboratory studies and one field study were conducted to evaluate the efficacy of Credelio Quattro, a novel chewable tablet containing moxidectin in combination with lotilaner, praziquantel and pyrantel, for the prevention of heartworm disease caused by D. immitis. METHODS: In the laboratory studies, dogs were inoculated with 50 D. immitis L3 larvae on Day - 30. Eight dogs per group were randomized to receive either a pl

Public-private partnership offers a strategic opportunity for lymphatic filariasis elimination in India by 2030.

India carries a significant burden of lymphatic filariasis. Although the national elimination programme has been implemented since the early 2000s, LF remains endemic in 345 districts. The economic and operational demands of large-scale interventions, such as mass drug distribution, morbidity management, and post-MDA surveillance, place a considerable strain on the public health infrastructure. Moreover, the disability, stigma and productivity loss associated with lymphedema impose substantial socio-economic burden. Given the large presence of private and non-profit sectors in India, public-pr

Gene regulation in response to host sex and infection route in Brugia pahangi with new genome annotation.

Male rodents are almost exclusively used for experimental maintenance of filarial nematodes because they have higher susceptibility to parasitism than female rodents. This study investigates the effects that rodent host sex has on the worm transcriptome for subcutaneous (SQ) infections and intraperitoneal (IP) infections. Significantly more adult worms were recovered from IP-infected gerbils (median = 78 worms/infected animal) relative to SQ-infected gerbils (median = 2 worms/infected animal). Microfilaria production in male gerbils infected via the SQ route (mean = 6.62 microfilariae (mf)/20

First human infection with Onchocerca takaokai (Spirurida: Onchocercidae) presenting as creeping eruption in Japan.

Zoonotic onchocerciasis is a rare human infection caused by Onchocerca species that normally parasitize non-human mammals. In Japan, all previously reported human cases have been attributed to Onchocerca japonica and have presented as localized, non-migratory subcutaneous nodules. Here, we report the first human infection caused by Onchocerca takaokai Uni et al., 2015. A 24-year-old male presented with linear migratory erythema on the forearm, clinically consistent with creeping eruption. Histopathological examination revealed an adult female filarial nematode with polymyarian-coelomyarian mus

Modeling the impact of community information dissemination on the transmission dynamics of lymphatic filariasis.

The World Health Organization has underscored the importance of implementing sustained and integrated interventions to interrupt the transmission of lymphatic filariasis (LF); however, achieving this goal requires a deeper understanding of the dynamics of the disease. In this study, we propose a deterministic mathematical model to investigate the transmission and control of LF, incorporating key factors including treated and untreated individuals, community information dissemination, immature and mature mosquito populations. We fitted the LF model to data using reported LF cases in Ghana from

Efficacy of oxfendazole treatment against infective larvae of Litomosoides sigmodontis.

Filarial infections are prevalent worldwide and affect both humans and animals. Two filarial infections, lymphatic filariasis and onchocerciasis, are listed among the neglected tropical diseases by the WHO and targeted for their elimination via large-scale public health campaigns in the affected countries. New drugs are urgently needed to facilitate this goal, and the WHO has outlined the need to develop novel adult worm killing (=macrofilaricidal) compounds for onchocerciasis as one of the critical actions in their roadmap for neglected tropical diseases for 2020-2030. Oxfendazole (Oxf) is an

Development of Recombinase Polymerase Amplification and CRISPR-Cas12a-Enhanced Isothermal Amplification Assays for Strongyloides stercoralis DNA Detection: A Pilot Study.

Soil-transmitted helminth (STH) infections are prevalent worldwide, but the true burden of strongyloidiasis is unclear due to lack of sensitive and field-friendly diagnostic tools. Diagnosis is often based on serological assays that are typically not point-of-care (POC). Although polymerase chain reaction (PCR) tests are sensitive and specific, the need for expensive equipment and highly skilled personnel limits their use in resource limited areas. Isothermal amplification assays are largely instrument-free, making them simpler to implement without loss of either sensitivity or specificity. We

Strengthening lymphatic filariasis elimination programmes with molecular xenomonitoring: critical utility and unmet operational needs.

Molecular xenomonitoring (MX)-the detection of filarial DNA in mosquito vectors-has become an important component of post- mass drug administration surveillance within the Global Programme to Eliminate Lymphatic Filariasis (GPELF). MX offers non-invasive, high-sensitivity detection of residual or recrudescent Wuchereria bancrofti transmission, particularly where human microfilaraemia and antigenemia surveys have limited power. However, the current operational model relies heavily on gravid-trap-based sampling, which captures a physiologically narrow, behaviourally biased subset of mosquitoes,

Polymorphic marker regions support divergence of Mansonella sp. "DEUX" and M. perstans.

BACKGROUND: Though being prevalent worldwide, Mansonella parasites are among the most neglected filarial nematodes. The true prevalence and genetic diversity of this genus have yet to be fully understood. Mansonella sp. "DEUX" is a recently described filarial nematode infecting humans and other primates in Gabon and Cameroon, although its status as distinct species has been controversial. We investigated four different polymorphic regions to further explore the genetic differences between Mansonella species and to support their status as distinct species. METHODS: We screened whole blood samp

First molecular characterization of Dirofilaria vector species and the distribution of canine dirofilariasis in Gampaha district, Sri Lanka.

BACKGROUND: Dirofilariasis is a vector-borne parasitic infection caused by filarial nematodes belonging to the genus Dirofilaria. Sri Lanka has reported the highest prevalence of human dirofilariasis cases in Asia. Molecular-based detection of Dirofilaria vector species has not yet been conducted in Sri Lanka, which could provide more sensitive information by directly detecting and analyzing genetic material. The present study aimed to analyze the distribution of canine dirofilariasis and to determine the Dirofilaria vector species in Gampaha district, Sri Lanka, using molecular-based techniqu

Clinico-pathological and renal morphological findings in dogs naturally infected with Dirofilaria repens.

BACKGROUND: Dirofilaria repens is a mosquito-borne filarial nematode that causes subcutaneous dirofilariasis in dogs and is closely related to Dirofilaria immitis. Infection with D. immitis can lead to immune-mediated glomerulonephritis characterized by immune complex deposition along the glomerular basement membrane, resulting in proteinuria and renal dysfunction. Reported histopathological changes include membranous glomerulonephritis with potential chronic progression to chronic interstitial nephritis, glomerulosclerosis, and amyloidosis. Despite the close relationship between these two Dir

Molecular and Phylogenetic Characterization of Onchocerca flexuosa in Red Deer from South-Eastern Europe.

Onchocerca flexuosa is a vector-borne filarial nematode infecting red deer (Cervus elaphus) throughout Europe. Despite numerous reports from Central, Northern, and Southern Europe, its occurrence in South-Eastern Europe has remained largely undocumented. This study provides the first molecularly confirmed report and the first systematic epidemiological assessment of O. flexuosa in red deer in Croatia. During the 2024-2025 hunting season, 110 legally harvested red deer from central Croatia were examined for subcutaneous nodules. Nodules were evaluated morphologically, and adult nematodes were i

Global Research Trends in Emerging Zoonosis Due to (the Filarial Nematode) Dirofilaria repens (1955-2025): A Bibliometric Analysis of a Climate-Driven Expansion.

Dirofilaria repens is the leading cause of subcutaneous (dogs) and subcutaneous/ocular dirofilariosis (humans) in the Old World. Despite its rapid geographical spread, driven by climate change, the emergence of new invasive vectors (Aedes albopictus) and growing interest in its study due to the emergence of new cases in areas previously free of the parasite, amongst other factors, scientific research into this pathogen remains limited. This study provides the first longitudinal bibliometric analysis of global research on D. repens (1955-2025). Data from Web of Science and Scopus were processed

The moving cervical mass: A surprising discovery.

Human dirofilariasis is a rare zoonotic infection caused by filarial nematodes of the genus Dirofilaria, with Dirofilaria repens being the most common species affecting humans. Humans are accidental hosts, typically presenting with solitary subcutaneous nodules that can mimic other benign or malignant conditions. Cervical (neck) localization is particularly uncommon, making diagnosis challenging, especially in regions where the disease is rarely reported. We report the 23rd confirmed case of human dirofilariasis in Tunisia, occurring in a 72-year-old woman from a rural area who presented with

Comparison of an integrated versus stand-alone approach to post-validation surveillance for lymphatic filariasis in Niue: a micro-costing study, 2025.

BACKGROUND: Lymphatic filariasis (LF) is a mosquito-borne parasitic disease which, in cases of chronic infection, may result in lymphoedema, elephantiasis, and hydrocele, imposing significant physical, social, and economic burdens. Global initiatives have reduced the prevalence of this condition, with Niue, a self-governing Pacific Island country, being validated as having eliminated LF as a public-health problem in 2016. To ensure transmission does not re-emerge, the World Health Organization recommends post-validation surveillance. As an integrated approach where LF-PVS is embedded within ex

First ever programmatic implementation of triple drug therapy in the prevention of lymphatic filariasis in Haiti: lessons learned in improving the quality and coverage of locally led programming in lymphatic filariasis-persistent and conflict-affected endemic communes.

Although significant strides have been made towards lymphatic filariasis (LF) elimination in Haiti, which has experienced ongoing multidimensional crises, 19 out of 140 communes still require mass drug administration (MDA) despite multiple rounds using the two-drug regimen of diethylcarbamazine (DEC) and albendazole. This includes the commune of Limonade, which has experienced persistently high LF prevalence despite nine rounds of two-drug MDAs since 2010. In September and October 2023 and July 2024, the Haitian Ministry of Public Health and Population, with support from IMA World Health and t

Lessons from the field: adapting lymphatic filariasis morbidity mapping and patient care amidst conflict in Ethiopia.

Conflict in Ethiopia has prevailed in recent years and is impeding the implementation of lymphatic filariasis elimination activities for morbidity management and disability prevention. This led to the need for alternative approaches to ensure safe and effective implementation. Here, we outline the challenges that were faced (e.g., insecurity, restricted travel to external field teams, limited in-person training, direct patient care). First, we describe what potential solutions were discussed and decided. Second, we summarise the planned adapted strategy and how it was implemented through centr

Subclinical Infections of Babesia and Dirofilaria in Dogs Presented to a Veterinary Teaching Hospital: Evidence for a Silent Reservoir of Infection in Sri Lanka.

PURPOSE: Canine filariasis and babesiosis are serious vector-borne diseases, often undetected in asymptomatic dogs. This hidden prevalence complicates disease control, as subclinical carriers may continue transmission. This study determined the prevalence of asymptomatic infections and explored potential links between breed types. METHODS: Blood samples were collected from dogs brought to the Veterinary Teaching Hospital for illnesses, vaccination, sterilization, and regular health check-ups. Babesia and filarial worms were morphologically identified through Giemsa-stained thin blood smears a
