Changes in circulating filarial antigen status in previously positive individuals: Lessons for treatment monitoring and pre-transmission assessment surveys.

BACKGROUND: The Global Programme to Eliminate Lymphatic Filariasis has made significant gains through mass drug administration (MDA) of Ivermectin/Albendazole. Periodic evaluation of the MDA programme in lymphatic filariasis elimination is particularly useful in determining end points for stopping the programme. This is a follow-up study that sought to examine the effects of additional time and MDA intake on antigenemia seroreversion in persons who had previously tested positive for LF using the Filarial Test Strip (FTS) and the TropBio ELISA over a period of 1-5 years. METHODOLOGY/PRINCIPAL

Lymphatic filariasis elimination in Kenya: Tracing the journey from 2002-2024 and pathways to achieving 2030 target.

Lymphatic filariasis (LF) is a parasitic neglected tropical disease (NTD) that can lead to debilitating conditions such as lymphoedema (elephantiasis) and hydrocele, impacting affected individuals' quality of life and economic productivity by interfering with their physical and psychological health. Like most countries, Kenya established its National Programme to Eliminate Lymphatic Filariasis in 2000. It launched its first Mass Drug Administration (MDA) in Kilifi District in 2002, with subsequent expansions to Kwale, Malindi, Tana River, Lamu, and Mombasa counties in Kenya's coastal region. T

A review of filarial nematodes parasitizing tick vectors: unraveling global patterns in species diversity, host associations, and interactions with tick-borne pathogens.

Ticks are well-known vectors of pathogens, including filarial nematodes, which can affect animal and human health. This review synthesizes current knowledge of filarial nematodes that utilize ticks as vectors, focusing on the Onchocercidae. Five genera of onchocercid filarial nematodes (Cercopithifilaria, Cherylia, Cruorifilaria, Monanema, and Yatesia) have demonstrated vector-parasite relationships with ticks, while Acanthocheilonema has only demonstrated molecular detection without confirmed vector competence. Vector competence has been experimentally proven for several species through docum

Unveiling the gendered abyss: exploring the case of women with lymphatic filariasis and the path to inclusive care in India.

INTRODUCTION: Lymphatic filariasis (LF), a neglected tropical disease (NTD), affects tropical regions marked by poor socioeconomic conditions and often results in non-curable filarial lymphedema. The impact of LF is often disproportionate, adversely affecting women due to a multitude of intersecting disadvantages. There has been little effort to understand the unique illness experience of women with LF. METHODS: To explore the interplay between gender and the illness experience of women afflicted with filarial lymphoedema, this qualitative study mapped the experience of 18 women and the perce

Postelimination Cluster of Lymphatic Filariasis, Futuna, 2024.

After detection of 2 clinical lymphatic filariasis (LF) cases in a postelimination context in 2023 on the island of Futuna (Wallis and Futuna archipelago), the Wallis and Futuna Health Agency conducted a LF prevalence survey in Futuna in May 2024. This cross-sectional study, carried out among schoolchildren <18 years of age, identified 5 children with antigenemia, indicating an estimated antigenemia prevalence in Futuna children nearing 2%. The study also confirmed a spatial cluster of cases in the village of Taoa, where the child antigenemia prevalence reached 7.5% (95% CI 2.1%-18.2%), and de

Major basic protein and eosinophil peroxidase support microfilariae motility inhibition by eosinophil ETosis.

Eosinophils are a hallmark of filarial infections. They are primary effector cells and can attack filariae by releasing extracellular traps that contain toxic cationic proteins, such as eosinophil peroxidase and major basic protein. Previous studies demonstrated that the extracellular traps of eosinophils are induced by the microfilariae of Litomosoides sigmodontis and that they inhibit their motility. In this project, we aimed to investigate the role of these cationic proteins during the extracellular trap-mediated immobilization of microfilariae. Our results indicate that extracellular DNA t

Human dirofilariasis - A potentially significant nematode zoonosis in an era of climate change.

Dirofilariasis is a mosquito-borne zoonosis caused by several species of the genus Dirofilaria. This disease can manifest as nodular lesions in subcutaneous tissues, various structures of the eye, the lungs and/or visceral organs. The Dirofilaria species and the vectors responsible for transmitting infection differ among various geographical regions. The most competent reservoirs of infection are domestic and wild canids (for Dirofilaria repens and Dirofilaria immitis), raccoons (for Dirofilaria tenuis) and bears (for Dirofilaria ursi), and humans represent aberrant or accidental hosts. Recent

Model development to assess the impact of a preventive treatment with sarolaner and moxidectin on Dirofilaria immitis infection dynamics in dogs.

BACKGROUND: Dirofilaria immitis is a mosquito-transmitted filarial parasite causing heartworm disease in dogs. The parasite may cause a significant disease burden to the dog population in high prevalence areas and is mainly managed through prophylactic treatments. METHODS: In this modelling study, the effect of a prophylactic treatment with moxidectin and sarolaner on heartworm disease dynamics was investigated in dogs. A compartmental model was developed to investigate different epidemiological settings considering different values for prevalence and host preference. RESULTS: When the mosqu

Low Wolbachia incidence in Bactrocera and Zeugodacus species from Thailand and genome analysis of Wolbachia associated with Zeugodacus apicalis.

Wolbachia are bacterial endosymbionts found widely in arthropods and filarial nematodes. Infecting about half of all arthropod species, Wolbachia manipulate their hosts in various ways, including cytoplasmic incompatibility. Here, we investigated Wolbachia diversity in Bactrocera and Zeugodacus, two prevalent tephritid fruit fly genera, using molecular methods. Wolbachia was only detected in Zeugodacus apicalis (de Meijere) (Diptera: Tephritidae) and not in the other 7 studied species. This newly discovered strain, named wZap, belongs to supergroup B with a 1.3 Mb genome containing 1,248 genes

Circulating Cell-Free Nucleic Acid Detection for Wuchereria bancrofti Infections.

Human lymphatic filariasis (LF) is primarily caused by the helminth parasites Wuchereria bancrofti (Wb) and Brugia malayi (Bm). Detecting parasitic circulating cell-free nucleic acids (ccfDNA/ccfRNA) in plasma is a promising approach for detection of active infections. Plasma RNA sequencing analyses from individuals with active Wb infection and uninfected controls identified 6 RNA targets that were specific to Wb and/or Bm. Assays developed to detect either ccfDNA and ccfRNA were successfully used in Wb-infected individuals, but DNA-based assays targeting WbTR1 were found to be the most sensit

Filariasis of parti-colored bats: phylogenetic analysis, infection prevalence, and possible vector mite identification.

INTRODUCTION: The epidemiology of filarial infections is a neglected area of bat research, with little information on filarial species diversity, life cycles, host ranges, infection prevalence and intensity, parasite pathogenicity, or competent vectors. Furthermore, molecular data for filarial worms are largely lacking. METHODS: Here, we examined 27 cadavers of parti-colored bat (Vespertilio murinus) from Czech rescue centers for filarial infection using gross necropsy. We also used nested polymerase chain reactions targeting partial mitochondrial cytochrome c oxidase subunit I (COI) partial

Access, utilisation and barriers to lymphedema care: A mixed method study in Puducherry, India.

Lymphatic filariasis (LF) causes chronic conditions such as lymphedema and hydrocele, which require continued care to prevent disease progression and reduce the risk of secondary infections. The delivery and utilisation of lymphedema care services are shaped by various socio-economic and systemic factors. Although India has implemented the strategy of mass drug administration (MDA) across all 345 endemic districts, it still lags in the implementation of morbidity care to affected individuals. Healthcare access and treatment barriers among lymphedema patients are less explored domains. This mix

Evaluation of Microfilaremic Individuals after Mass Drug Treatment with Ivermectin, Diethylcarbamazine, and Albendazole for Lymphatic Filariasis in Papua New Guinea.

After mass drug administration (MDA) for lymphatic filariasis, which involved a single coadministered dose of ivermectin plus diethylcarbamazine and albendazole (IDA), concerns arose regarding individuals who remained microfilaremic. This situation raised questions about the efficacy of the drugs and whether some individuals had not ingested them. In East New Britain Province, Papua New Guinea (PNG), where 81.7% of the population received IDA, 10 individuals were found to have microfilaremia 12 months after the first round of MDA in an area that had a high baseline of microfilaremia (n = 29 mi

Xenomonitoring as an epidemiological tool supporting post-stop surveillance of albendazole-ivermectin mass drug distribution in the Bougouni-Yanfolila evaluation unit, Sikasso, Mali, in 2023.

INTRODUCTION: Mali and Guinea share a border and are both endemic for lymphatic filariasis (LF). However, their progress towards eliminating this disease varies. Mali is currently in the LF transmission assessment survey phase (TAS), while Guinea continues to implement mass drug administration (MDA). As the populations of these two countries are closely related, and vectors are present, the emergence of LF is theoretically possible in the Bougouni-Yanfolila evaluation unit (EU). This XenoFil study, which combines xenomonitoring and serosurveillance in health facilities, was used as a surveilla

Unraveling the Dynamics of Human Filarial Infections: Immunological Responses, Host Manifestations, and Pathogen Biology.

Lymphatic filariasis (LF), or elephantiasis, is a neglected tropical disease caused by filarial worms, primarily Wuchereria bancrofti, transmitted through mosquito bites. It often begins in childhood but may not show symptoms until later, leaving many individuals asymptomatic for long periods. LF disrupts the lymphatic system, causing severe swelling in the limbs and genitals, leading to deformities and disabilities. The World Health Organization estimates that around 51 million people are affected globally, with 36 million suffering from chronic conditions like lymphedema and hydrocele. In 20

Nutritional predictors of lymphatic filariasis progression: Insights from a machine learning approach.

Lymphatic filariasis (LF) is a mosquito-borne neglected tropical disease that causes disfiguring of the affected extremities, often leading to permanent disability and stigma. Described as a disease of poverty, the impact of socioeconomic indicators such as nutrition on LF remains largely unexplored. This cross-sectional study investigates nutritional predictors implicated in the progression of LF using machine learning methods in the Ahanta West Municipality, Ghana. There were 109 participants with a mean age of 50.72±13.8, and three-quarters being females. Only 14 (12.8%) each had comorbidit
