Hippo pathway disruption and YAP1 activation in anogenital papillary squamous intraepithelial lesion.

AIMS: Papillary squamous intraepithelial lesion (PSIL), also known as papillary immature metaplasia, is a rare anogenital lesion associated with low-risk human papillomavirus (HPV) infection. The molecular pathogenesis of PSIL remains unclear. This study aims to investigate the activation of YAP1, a Hippo pathway co-activator, in PSIL and compare it to condyloma and papillary HSIL. METHODS AND RESULTS: YAP1 activation was evaluated by immunohistochemistry using an antibody specific for activated/unphosphorylated YAP1 (act-YAP1) in 36 PSIL cases, 20 condyloma and 2 papillary HSIL cases. HPV ge

Comparative efficacy of topical garlic extract and 20% podophyllin solution in the treatment of female genital warts: a triple-blind randomized clinical trial.

BACKGROUND: Genital warts caused by human papillomavirus are common sexually transmitted infections, with no single therapy providing consistent clearance. Garlic extract has antiviral and antiproliferative properties and may offer a low-cost treatment. OBJECTIVE: To compare the efficacy and safety of topical 5% garlic extract with 20% podophyllin solution in women with genital warts. METHODS: Seventy-four women were randomized to 5% garlic extract (n = 37) or 20% podophyllin (n = 37), applied weekly for four weeks. Wart size and count were assessed using marginal models (generalized estimat

Human Papillomavirus Condylomas of Extragenital Site in Immunocompetent Man, New York, USA, 2025.

Human papillomavirus types 6 and 11 are associated with mucosal condylomata acuminata, with rare reports of extragenital manifestations. We report a case of condyloma localized to the neck of an immunocompetent heterosexual man, without concurrent anogenital lesions. Our findings challenge theories of the site-specific tropism of these human papillomavirus types.

Primary Breast Pure Metaplastic Squamous Cell Carcinoma: A Breast Abscess With Squamous Metaplasia and Malignant Transformation That Mimics Condylomata Acuminata.

Pure metaplastic squamous cell carcinomas (MSCC) of the breast are rare. We report the case of a 57-year-old woman who presented with a breast abscess in whom a papillomatous lesion was identified during incision and drainage. The initial biopsy demonstrated hyperplastic squamous epithelium with koilocytic atypia, raising the possibility of condylomata acuminata. Subsequent wide excision revealed extensive squamous metaplasia with invasive squamous cell carcinoma arising within a cystic papillary lesion consistent with pure MSCC of the breast. Immunohistochemistry revealed positivity for p63 a

Real-world effectiveness and impact of human papillomavirus vaccination in males: a comprehensive review.

OBJECTIVES: Gender-neutral vaccination (GNV) for human papillomavirus (HPV) has increasingly been implemented worldwide. However, real-world evidence of vaccine effectiveness (VE) in males and the population-level impact of GNV remains heterogeneous. We summarized the real-world VE of HPV vaccination in males and the impact of GNV. METHODS: A comprehensive review was performed using Embase and PubMed for studies published from January 2007 to May 2024 that evaluated quadrivalent or nonavalent HPV vaccines. Eligible studies assessed VE or GNV impact on HPV infections, precancerous lesions, can

Sequential thulium laser vaporization and catheter-guided 5-aminolevulinic acid photodynamic therapy for long-segment anterior urethral condyloma acuminatum: A case report.

Long-segment intraurethral condyloma acuminatum (CA) is difficult to treat because the urethral lumen limits visual access, instrument maneuverability, and uniform contact between a topical agent and the mucosa. Treatment must balance immediate removal of visible disease against cumulative mucosal injury: repeated or deep destructive procedures can cause bleeding, ulceration, pain, scarring/fibrosis, stenosis, and urethral stricture [1,2]. Electrocautery can rapidly destroy discrete accessible lesions but may produce thermal injury when applied deeply or circumferentially. Cryotherapy is usefu

ALA-PDT for giant condyloma acuminatum in an immunodeficient child with BCL11B mutation: safety and efficacy.

BACKGROUND: The treatment of giant condyloma acuminatum in immunodeficient children remains a challenge. In immunodeficient children, human papillomavirus infection may progress to extensive condyloma acuminatum (CA. However, 5-aminolevulinic acid photodynamic therapy (ALA-PDT) has shown favorable normal tissue-sparing properties in combination with high rates of lesion control. This case aims to highlight the safety and efficacy of ALA-PDT on giant condyloma acuminatum in an immunodeficient child induced by BCL11B mutation. METHODS: This report examines two affected siblings presenting with

[Vaccine prevention of HPV-associated diseases].

Early prophylactic human papillomavirus (HPV) vaccination of girls and boys leads to long-lasting sterile immunity against the HPV types covered by the respective vaccine. Both in clinical trials and in population-based studies, virus-like particle (VLP)-based HPV vaccines have exceeded all expectations regarding immunogenicity and efficacy. In clinical trials, HPV vaccines induced protection rates of 95-100% against high-grade cervical intraepithelial neoplasia (CIN) caused by vaccine-covered types, and nearly 100% against HPV6/11-induced anogenital warts. Numerous studies have demonstrated t

Intraepithelial Penile Lesions.

Intraepithelial penile lesions encompass non-neoplastic and preneoplastic lesions. Non-neoplastic lesions comprise condyloma acuminatum, including giant condyloma acuminatum, also known as Buschke-Löwenstein tumor. It is usually the low-risk human papillomavirus (HPV) types 6 and 11 that are most prevalent in penile condylomas. However, high-risk HPV types can be detected along with low-risk types in a subset of patients. Penile intraepithelial neoplasia (PeIN) is a preneoplastic lesion that is either HPV-associated or HPV-independent. HPV-associated PeIN represents the majority of PeIN in reg

Impact of intralesional tuberculin PPD injection on anogenital warts: Interferon-α, interleukin-2, and treatment outcomes.

The clinical manifestation of anogenital warts (AGW) resulting from Human Papillomavirus (HPV) infection is influenced by host immune responses. Interferon (IFN)-α contributes to antiviral activity, while Interleukin (IL)-2 plays an important role in cellular immunity. Tuberculin purified protein derivative (TPPD) has been investigated as an immunotherapeutic agent. This exploratory quasi-experimental study analyzed 12 AGW patients treated with intralesional TPPD (5 tuberculin units weekly for six injections). Tissue samples were collected at baseline and week 2, and serum samples at baseline

Integrative metabolomic and single-cell transcriptomic analysis of recurrent condyloma acuminatum in humans.

Condyloma acuminatum (CA), primarily caused by low-risk HPV6/11, is a benign proliferative disease that is difficult to cure and prone to recurrence. However, the molecular and immune mechanisms underlying relapse remain unclear. We combined metabolomic profiling with single-cell RNA sequencing to investigate recurrence-associated changes. Metabolomics revealed dysregulation of ascorbate and aldarate, glycerophospholipid, purine, and arginine/proline metabolism in recurrent CA. Single-cell analysis identified altered expression of metabolism-related genes (AMD1, GSTM3, ALDH3A1, GPX1, GPX4) in
