Discovery of Trolox amide derivatives as potent membrane-targeted ferroptosis inhibitors for corneal injury therapy.

Vitamin E and its water-soluble analogue Trolox are potent radical scavengers but require high concentrations to inhibit ferroptosis effectively. The discovery of a Trolox amide derivative 1 showing 95-fold greater potency inspired the systematic synthesis of a focused library. Most amide derivatives demonstrated significantly enhanced potency, with IC50 values in the sub-micromolar range. Notably, compound 20, which features an indazole moiety, emerged as the most potent inhibitor, exhibiting a 556-fold improvement in activity (IC50 = 0.036 μM) over Trolox and a favorable safety index. Therapeutic Efficacy of Rapamycin in an Experimental Mouse Model of Corneal Alkali Burn.

Corneal alkali burn induces severe inflammation and tissue damage, leading to loss of corneal transparency and vision impairment. In this study, we evaluated the therapeutic potential of rapamycin (RAPA) compared with cyclosporine A (CsA) in a mouse model of corneal alkali burn, focusing on nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB)-mediated inflammatory signaling and its impact on corneal wound healing and repair. Notably, RAPA robustly suppressed NF-κB activation, reduced infiltration of F4/80 macrophages and MPO neutrophils, and downregulated pro-inflammatory cyt

Topical Application of Preserved and Unpreserved Bevacizumab Eye Drops Improves Ocular Surface Parameters in Dogs with Chronic Keratitis: A Pilot Study.

Corneal neovascularization (CNV) in dogs causes chronic ocular surface inflammation, which may disrupt tear film stability and meibomian gland function. As bevacizumab inhibits abnormal vessel growth and affects tear composition, it may benefit dogs with ocular surface disorders. The aim of this pilot study was to evaluate the effects of bevacizumab eye drops on the ocular surface in dogs with chronic CNV. Fourteen eyes from nine client-owned dogs were included. Dogs received either preserved (BBAC-group) or unpreserved (B-group) 0.25% bevacizumab eye drops twice daily for 28 days. Re-examinat

Dynamic regulation mechanisms of macrophages and advances in targeted therapy for corneal injury repair.

Corneal injury is a major cause of visual impairment worldwide and involves a tightly regulated cascade of inflammation, tissue repair, and remodeling. This process typically progresses from an early inflammatory phase to proliferative repair and ultimately to stromal remodeling, with potential outcomes including corneal neovascularization and fibrosis that compromise transparency and vision. Accumulating evidence highlights macrophages as central regulators across these stages. In the early phase, pro-inflammatory (M1-like) macrophages mediate pathogen clearance and immune activation through

Traumatic canalicular laceration and complex eyelid injury in a newborn delivered by forceps.

We present a neonatal case of complex upper eyelid lacerations with canalicular injury following a forceps-assisted vaginal delivery. Mother's insistence on vaginal delivery despite obstetric advice of caesarean section led to obstructed labour requiring forceps delivery. The neonate presented with four full-thickness lacerations of the right upper eyelid, including injury to the upper canaliculus. Immediate surgical repair with canalicular stenting was performed. A corneal ulcer developed secondary to stent-induced mechanical trauma. Early stent removal with topical fortified antibiotics led

Inhibition of PRMT5 reduces angiogenesis in a corneal neovascularization model by blocking NLRP3-mediated pyroptosis.

Protein arginine methyltransferase 5 (PRMT5) is a histone methyltransferase crucial for cell proliferation, differentiation, and inflammation. However, the biological functions of PRMT5 and its underlying molecular mechanisms in corneal neovascularization (CNV) remain unclear. This study utilized corneal alkali burn and vascular endothelial growth factor (VEGF)-induced HUVEC models to examine the role of PRMT5 in CNV. We found that PRMT5 expression was significantly upregulated following corneal alkali burn. Experiments both in vitro and in vivo showed that PRMT5 knockdown or inhibition lowere

Development of Miniprotein Binders to Inhibit VEGF-Induced Corneal Neovascularization.

PURPOSE: Corneal neovascularization (CNV) disrupts corneal transparency and impairs vision and is driven primarily by vascular endothelial growth factor (VEGF-A165). Current therapies rely on off-label anti-VEGF agents, which are limited by poor corneal penetration and the need for repeated dosing. Herein, we aimed to rationally design and validate novel hyperstable VEGF-A165 miniprotein inhibitors against VEGF-A165 for preventing CNV. METHODS: VEGF-A165 miniprotein binders were generated using a de novo diffusion-based protein design protocol and expressed in Escherichia coli (E. coli). Sele
