Impact of morphological variations on preoperative and postoperative central corneal thickness in congenital cataract: a retrospective observational study.

PURPOSE: Central corneal thickness (CCT) has been reported to be thicker in aphakic and pseudophakic eyes after congenital cataract surgery. We aimed to investigate the factors influencing preoperative CCT and changes in CCT after cataract surgery. METHODS: We included 62 eyes of 39 patients with congenital cataracts who underwent cataract surgery at Hiroshima University between February 2006 and July 2022. Pre- and postoperative CCT were measured using a noncontact specular microscope or ultrasound pachymeter. Cataracts were categorized into five morphologies: anterior, posterior, nuclear, t

IL-1β-Mediated Immunometabolic Adaptation in Corneal Epithelial Cells.

PURPOSE: As an external mucosal surface, the corneal epithelium is subject to a barrage of stressors that are known to trigger inflammation. IL-1β, a master regulator of inflammation, is secreted into the preocular tear film by ocular surface epithelial cells and infiltrating immune cells. While increased levels of IL-1β have been associated with corneal disease, the effects of IL-1β on mitochondrial function in corneal epithelial cells (CECs) is unknown. METHODS: To investigate the effects of IL-1β on mitochondrial function, telomerase immortalized human CECs were cultured in either 50 ng/mL

Peptide DFCPPGFNTK Mitigates Dry Eye Pathophysiology by Suppressing Oxidative Stress, Apoptosis, Inflammation, and Autophagy: Evidence from In Vitro and In Vivo Models.

Dry eye is an ophthalmic disease with an intricate pathomechanism, and there are no effective interventions or medications available. We investigated the effects of a peptide, DFCPPGFNTK (DFC), screened from tilapia skin hydrolysate on dry eye and its underlying mechanisms. In vitro, human corneal epithelial cells (HCECs) were challenged by 100 mM NaCl in a hyperosmotic environment. DFC restored the cell viability of HCECs induced by NaCl, reduced the transition of mitochondrial membrane potential, delayed the apoptosis of damaged cells, reduced the production of reactive oxygen (ROS) and malo

Prospective consecutive case series of patients with neurotrophic keratopathy associated with unilateral total limbal stem cell deficiency caused by severe ocular surface burns.

INTRODUCTION: Limbal stem cell deficiency (LSCD) is commonly caused by ocular surface burns. Corneal nerve damage, often due to infection, inflammation, trauma, leads to neurotrophic keratopathy (NK). This study investigates corneal sensation and nerve damage in patients with unilateral total LSCD before and after autologous cultivated limbal epithelial transplantation (Auto-CLET). METHODS: This prospective, single-centre case series included patients with unilateral total LSCD due to severe burns treated at Royal Victoria Infirmary, UK, between June 2012 and January 2018. Corneal sensation w

Potential Roles of Extracellular Vesicles in Murine Tear Fluids in the Physiology of Corneal Epithelial Cells In Vitro.

Biological extracellular vesicles in tear fluids, such as exosomes, are thought to have physiological functions in the management of healthy ocular surface epithelium, including corneal epithelium. However, the physiological roles of tear extracellular vesicles in the ocular surface remain unclear. In this study, we investigated the physiological function of tear extracellular vesicles in mouse tear fluids in the ocular surface epithelium in vitro. Morphological analysis of the isolated extracellular vesicles from mouse tear fluids was performed using nanoparticle tracking analysis and transmi

Nitrogen mustard causes progressive tissue injury, severe DNA damage, and necroptosis in the cornea.

The eye is sensitive and vulnerable to vesicants such as sulfur and nitrogen mustard (NM). To assess the adverse effects of NM exposure on the ocular surface, we used porcine and rat corneas and NM-wetted filtrate paper disks to assess NM's ex vivo and in vivo effects on corneal health. In cultured porcine corneas (N = 5), 5 mg/ml NM caused exposure time-dependent damage, which progressed from 1 to 2 days post-NM exposure (dpe), with increased programmed cell death, particularly necroptosis at 2 dpe. In rat corneas (N = 4), 5 mg/mL NM caused corneal opacification, punctate keratitis, pupil ani

Corticosteroids elevate intraocular pressure through suppression of TREK-1 signaling.

Clinicians are often forced into the dilemma of whether to battle ocular inflammation or preserve vision imperiled by elevated intraocular pressure (IOP). Anti-inflammatory treatments utilizing glucocorticosteroid regimens may induce glaucoma by chronically elevating IOP via increased trabecular meshwork (TM) resistance to the flow of aqueous humor, but it is not known whether pressure transduction itself is impacted by steroids and how changes in TM mechanosignaling affect conventional outflow resistance and IOP. To address this, we investigated the role of TREK-1 (TWIK-related potassium chan

Prior contact lens wear reduces susceptibility of the superficially-injured cornea to bacterial adhesion.

Contact lens wear in humans and mice is consistently associated with asymptomatic corneal parainflammation. Here, we tested the hypothesis that the corneal response to lens wear alone can function to protect it against bacterial adhesion enabled by superficial injury. One eye only of mT/mG-LysMcre mice (cell membranes red; Lyz2+ cells green) wore a contact lens for 4-6 days. After lens removal, mice were anesthetized and both corneas superficially-injured before bacterial inoculation with either a mouse eyelid commensal (Macrococcus epidermidis) or a corneal pathogen (Pseudomonas aeruginosa).

SA-XV, a 15-amino acid fragment of host defense peptide S100A12, targets mitochondria and is protective against fungal infections.

Fungal infections are a huge emerging crisis, with more than 2 million people infected worldwide annually. Corneal infections caused by fungus are the major cause of vision loss and often warrant corneal transplantation. Both Fusarium spp. and Candida spp. are critical etiological agents of fungal keratitis and also common causes for invasive fungal infections with high mortality rates. In the previous work, we described growth inhibition of Fusarium spp. by S100A12, a host antimicrobial peptide. Here, to optimize a potential therapeutic, we have studied a 15-amino acid fragment of S100A12, SA

Enabling In Vivo Longitudinal Evaluation of Descemet's Membrane Thickness in Wild-type and FECD Mice Using Self-Referenced Optical Coherence Microscopy.

PURPOSE: Descemet's membrane (DM) remodeling is a key factor in the etiology of early-onset Fuchs endothelial corneal dystrophy (FECD). Although various mouse models have been developed to replicate major FECD phenotypes, including DM thickening, there is currently no imaging technique capable of evaluating changes in mice DM thickness in vivo. This work proposed a novel self-referenced optical coherence microscope (OCM) to longitudinally evaluate age-dependent and FECD-dependent changes in DM thickness in mice. METHODS: The self-referenced OCM used the mouse corneal surface as the reference

A novel brain-to-gut communication pathway mediated by astrocyte-derived small extracellular vesicles modulates stress-induced intestinal inflammation.

The connection between stress-induced mood disorders and inflammatory bowel diseases (IBD) has been well-established. However, the cellular mechanisms by which psychological stress can influence intestinal inflammation remain mostly enigmatic. Here, we hypothesize that psychological stress regulates intestinal inflammation through the release of small extracellular vesicles derived from astrocytes (AsEVs). In-utero electroporation performed to selectively express an AsEV-associated membrane recombinant protein in rat forebrain astrocytes reveals that this protein is transferred to the gut-asso
