Integrated multi-omics analysis combined with clinical validation reveals that HLA-DRB5 and ODAPH are causal risk genes for keratoconus.

This study aimed to elucidate the genetic determinants of keratoconus (KC) using a multi-omics approach, with a focus on identifying key causal genes to address gaps in understanding the disease’s pathological mechanisms. An integrated multi-omics study combining bioinformatics analysis of transcriptome data with clinical validation. This approach does not include a traditional control group but utilizes public databases and patient samples to assess genetic associations. Five KC patients from the First Affiliated Hospital of Xi’an Jiaotong University provided corneal tissues and blood samples

William W. Cooper and his epidemiological study "On the conical cornea" of 1850.

PURPOSE: We present the life and work of William White Cooper, with a focus on his 1850 study On the conical cornea, the earliest known multicentre epidemiological investigation of keratoconus. We contextualise Cooper's contribution within mid-19th-century ophthalmology, reassessing his findings in light of modern epidemiological knowledge. METHODS: Historical research was conducted using Gallica (the digital platform of the National Library of France (BnF)) and the historical resources of The Lancet website. The keywords used were 'conical cornea' or 'keratoconus,' which were the most common

[Corneal crosslinking for keratoconus: real-life data and Committee for Refractive Surgery (KRC) recommendations].

Keratoconus is a chronic corneal ectasia characterized by a progressive course frequently starting in young age, in which in advanced stages often only a corneal transplantation can help to restore vision. Since the early 2000s, crosslinking has been available as a surgical procedure that can halt the progression of keratoconus. Crosslinking is a low-risk procedure with a high chance of success. To ensure that the costs of crosslinking are covered by the statutory health insurance in Germany, medical evidence of progression must be provided. In the assessment of the Federal Joint Committee on

Timing of corneal cross-linking in eyes undergoing intracorneal ring segments for keratoconus: same-day versus staged CXL: a systematic review and meta-analysis.

BACKGROUND: In keratoconus managed with intracorneal ring segments (ICRS) plus corneal cross-linking (CXL), ICRS primarily reshapes the cornea while CXL provides biomechanical improvement and shape stabilization. However, clinical practice varies regarding whether CXL should be performed on the same day as ICRS or delayed, and it remains uncertain whether CXL timing modifies the postoperative optical and topographic outcomes of ICRS. METHODS: We systematically searched PubMed, Web of Science, Scopus, and the Cochrane Library from inception to October 2025 and included seven comparative studie

Corneal infiltration due to late endothelial perforation by an intrastromal ring segment.

Intrastromal rings have a high safety profile, with late complications being rare. A clinical case is presented of a patient who attended the emergency department due to photophobia and redness in the right eye with a two-day onset. A relevant medical history includes the implantation of an intrastromal corneal ring segment for keratoconus 11 years earlier. Examination revealed a deep whitish stromal infiltration around the ring. After several weeks of antibiotic treatment, the decision was made to explant the ring. Preoperative OCT demonstrated endothelial perforation at the site of the infil
