Reciprocal effects of conditioned medium on gene and protein expression of limbal epithelial cells and limbal fibroblasts in congenital aniridia.

Congenital aniridia is marked by substantial inflammatory changes to the ocular surface. However the exact mechanisms of epithelial-stromal interaction are not fully understood. The purpose of this study was to investigate inflammatory cytokine expression in limbal epithelial cells and fibroblasts following exposure to each other's conditioned medium (CM). Healthy primary limbal epithelial cells (pLECs) and healthy (LFC) or aniridia primary limbal fibroblasts (AN-LFC) were isolated. A PAX6-deficient limbal epithelial cell line (mut-LSCs) modeled aniridia. pLECs underwent siRNA-mediated PAX6 kn

ATAD3 duplications bridge mitochondrial diseases and Aicardi-Goutières syndrome.

A recurrent 68-kb heterozygous duplication of the ATAD3 locus has been implicated in a mitochondrial disorder characterized by prenatal or neonatal onset and rapidly fatal course with cardiomyopathy, hyperlactataemia, cataract, and encephalopathy. We analysed the clinical, neuroimaging, and molecular spectrum associated with duplication of the ATAD3 gene cluster in nine patients (four males, five females; age range: 3 days-3 years, median: 11 days, mean: 7.8 months, SD: 1 year 1 month). Five patients presented with prenatal signs (intrauterine growth restriction in four of nine and cardiac abn

Cardiac arrhythmia, developmental delay, epilepsy and ichthyosis due to Xp22.31 deletion: review of literature and case report.

BACKGROUND: The broad phenotypic variability observed in patients with Xp22.31 deletion, traditionally associated with X-linked ichthyosis (XLI), is increasingly recognized as encompassing a wider spectrum of clinical manifestations. While ichthyosis, caused by STS gene deletion, remains the hallmark feature, recent studies and genomic analyses (e.g., chromosomal microarray and whole genome sequencing) have revealed additional extracutaneous phenotypes. These include corneal opacification, cryptorchidism, autism spectrum disorders, intellectual disability, epilepsy, developmental delay, renal

Collagen IV in Gould syndrome and Alport syndrome.

Collagen IV is a basement membrane component that is encoded by six genes in mammals (COL4Α1-COL4A6). The α-chains encoded by these genes assemble into three known heterotrimers - collagen α1α1α2(IV), α3α4α5(IV) and α5α5α6(IV) - that provide structure and act as multifunctional signalling platforms. The ancestral collagen superfamily members collagen alpha-1(IV) chain (COL4Α1) and collagen alpha-2(IV) chain (COL4Α2) are present throughout the animal kingdom and in all developing and most mature mammalian tissues. Consistent with this broad distribution, variants in COL4A1 and COL4A2 cause a co

Zebrafish adamtsl4 knockout recapitulates key features of human ADAMTSL4-related diseases: a gene involved in extracellular matrix organization, cell junctions and development.

Loss-of-function mutations in ADAMTSL4, a gene encoding an extracellular matrix-associated protein with incompletely understood biological roles, are linked to autosomal recessive disorders predominantly characterized by lens dislocation. Pupil ectopia, increased intraocular pressure, retinal detachment, cataracts, and skeletal abnormalities are also observed in some patients. To investigate ADAMTSL4 biology and related diseases we established a zebrafish knockout line using CRISPR/Cas9 genome editing. The generated zebrafish model harboured the c.234-351del mutation in adamstsl4, reducing its

Protein arginine methyltransferase 1 stimulates basal cell proliferation and migration to maintain corneal epithelial homeostasis.

The corneal epithelium is a constantly self-renewing, stratified squamous tissue that protects the inner eye from external stimuli. The organization of the corneal epithelium involves multiple biological activities, including basal cell proliferation and centripetal migration. However, the underlying molecular mechanisms remain unclear. Herein, we identify protein arginine methyltransferase 1 (PRMT1) as a key regulator of corneal epithelial homeostasis. We exploited an inducible Prmt1 knockout mouse model and observed apparent disruption in the corneal epithelial homeostasis. PRMT1-deficient m

Congenital aniridia: European COST action ANIRIDIA-NET guidelines for diagnosis, management and care.

Congenital aniridia is a rare ocular disorder affecting the majority of eye structures and can be associated with systemic manifestations. The main visible phenotypic characteristic is the partial or complete absence of the iris; however, foveal hypoplasia is a more frequent and reliable clinical sign. Other ocular comorbidities are associated with the disease, such as cataract, keratopathy and optic nerve hypoplasia. Mutation in the PAX6 gene is the most common cause of congenital aniridia, but other causative mutations exist. The main ocular symptoms experienced by those with congenital anir

Prolonged aftersensations to thermal noxious stimulation as a screening tool for central contributions to chronic ocular pain.

Painful aftersensations (AS) are sensations that persist after termination of a noxious stimulus, and prolonged AS may reflect abnormal somatosensory processing. We aimed to validate AS as indicators of central nervous system processing abnormalities in individuals with chronic ocular pain (COP). We examined two cohorts (Groups 1; n=278 and 2; n=64) who underwent an ocular surface examination, quantitative sensory testing (QST), and pain symptom assessment. Individuals were asked to rate pain intensity and unpleasantness (0-100) of AS at 15 s and 30 s after termination of a repeated stimulus p

Guiding first-line treatment decisions in advanced urothelial carcinoma: a global survey.

BACKGROUND: Enfortumab vedotin combined with pembrolizumab (EV-P) has become the new standard first-line therapy for patients with advanced urothelial carcinoma (aUC), based on its superior efficacy over platinum-based chemotherapy. As this regimen is increasingly adopted in routine care, treatment decisions may often occur in sites without dedicated genitourinary oncology expertise. This global survey aimed to explore how physicians perceive clinical factors that may influence the safe and effective use of EV-P in daily practice. MATERIAL AND METHODS: A panel of international physicians with
