A Study of the Causal Association Between Autoimmune Diseases and Blinding Eye Diseases.

INTRODUCTION: Autoimmune diseases (ADs) are known to affect multiple organs, including the eyes. This study aims to evaluate the causal associations of rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), and ankylosing spondylitis (AS) with cataracts, age-related macular degeneration (AMD), and glaucoma using Mendelian randomization (MR). METHODS: Genome-wide association study (GWAS) data were utilized to identify single- nucleotide polymorphisms (SNPs) strongly associated with AS, SLE, RA, cataracts, AMD, and glaucoma, which served as instrumental variables (IVs). The basic analys

Assessing the relationship between gout and the risk of cataract in community-dwelling older adults: mediation and moderation analysis.

OBJECTIVE: This study aimed to assess whether gout is a risk for cataract and identify important factors contributing to the association. METHODS: A total of 381,402 individuals from the UK Biobank were enrolled at baseline (2006-2010). Cataract was ascertained using hospital inpatient and self-reported data until early 2021. Gout was determined by ICD-9, ICD-10, self-report, and medication at baseline. The Cox regression model was used to estimate the hazard ratio (HR) and 95% confidence intervals (CIs) for the risk of cataract. RESULTS: The risk of cataract was significantly increased in p

Comparative Evaluation of Visual Outcomes and Patient Satisfaction After Eyecryl SERT Versus TECNIS Eyhance Intraocular Lens Implantation in Cataract Patients.

Background Intraocular lenses (IOLs) that extend a single focal point are designed to improve intermediate vision while maintaining the optical clarity of traditional monofocals. Unlike multifocal designs, they aim to reduce visual disturbances such as glare and halos, offering a more comfortable visual experience. These lenses are particularly beneficial for modern tasks like computer use, cooking, or dashboard viewing, where intermediate vision plays a critical role. This study is part of a larger prospective investigation comparing visual outcomes, patient satisfaction, and safety of the Ey

Causal Association Between Commonly Used Medicines and Diabetes-Related Eye Diseases: Univariable and Multivariable Mendelian Randomization Study.

BACKGROUND: Diabetic patients require long-term polypharmacy, yet the causal effects of these medications on ocular complications-specifically diabetic retinopathy (DR), diabetic maculopathy (DMac), and glaucoma-remain unclear due to confounding in observational studies. METHODS: We performed a Mendelian randomization (MR) study using GWAS summary statistics for 23 common medication classes and five diabetic eye diseases. Multivariable MR (MVMR) was employed to adjust for key comorbidities (e.g., hypertension, type 2 diabetes), while enrichment analyses explored biological mechanisms. RESULT

Association of accelerometer-monitored physical activity with incident cataract: epidemiological and genetic evidence.

PURPOSE: To investigate the association and potential causal relationship between accelerometer-assessed physical activity (PA) and the risk of senile cataract (SC). METHODS: The study population was derived from the UK Biobank database. Triaxial accelerometer data from wearable devices, collected over a continuous 7-day wear period (accounting for potential seasonal variation), were used to define PA at light (LPA), moderate (MPA), and vigorous (VPA) intensities. Multivariable Cox proportional hazards models were used to examine the association between PA intensity-specific domains and SC in

Quality of ICD-10 Diagnoses for Cataract in the Danish National Patient Register.

PURPOSE: To assess the quality of International Classification of Diseases (ICD)-10 diagnoses for cataract in the Danish National Patient Register, which remains unexplored. METHODS: We extracted 100 consecutive patients registered with a diagnosis of cataract (50 with H25 [senile cataract] and 50 with H26 [other cataract]). Positive predictive values (PPVs) were calculated as the proportion of diagnoses that were confirmed after manual medical record review. PPVs were calculated both overall and in prespecified subgroups. RESULTS: The median patient age was 76 years, and most were females (

Cataract GWAS meta-analysis and multi-omics Mendelian randomisation inform disease mechanisms and candidate target prioritisation.

BACKGROUND: Cataract is a highly prevalent cause of visual impairment and blindness worldwide. Although genome-wide association studies (GWAS) have mapped multiple cataract risk loci, further work is required towards translating these signals and integrating multi-omics evidence for preventing or delaying age-related cataract progression. METHODS: A senile cataract GWAS was first conducted in All of Us, followed by ancestry-specific GWAS meta-analyses in European-ancestry (161,812 cases and 1,099,738 controls) and East Asian ancestry (66,679 cases and 521,995 controls). To prioritise candidat

Causal association between cataract and psychiatric disorders: A bidirectional 2-sample Mendelian randomization study.

This study aimed to investigate the genetic causality between cataract (senile and other types) and psychiatric disorders using Mendelian randomization (MR). We performed a bidirectional 2-sample MR analysis using genome-wide association studies summary data. Primary analysis employed inverse-variance weighted, MR-Egger, weighted median, and weighted mode methods. Sensitivity analyses included Cochran Q, MR-pleiotropy residual sum and outlier, MR-Egger intercept, and leave-one-out tests to assess heterogeneity and pleiotropy. Forward MR analysis revealed a suggestive causal effect of other cat

[Expression of Concern] Anti‑metastatic and differential effects on protein expression of epigallocatechin‑3‑gallate in HCCLM6 hepatocellular carcinoma cells.

INTRODUCTION: Available work from North America indicates that same-sex attracted (SSA) individuals enjoy aspects of rural life but nonetheless report encountering homophobia and experiencing isolation from SSA networks. The experience of prejudice and social isolation are often associated with psycho-social distress among the general population of same-sex attracted individuals. Little is known of how SSA women experience life in rural areas of Australia and how this influences their psycho-social wellbeing. METHODS: This was a small-scale qualitative study using guided interviews to explore

Mechanism of activation of an ancestral TEC kinase by PIP3.

BACKGROUND: The introduction of second-generation antipsychotics (SGAs) over the past 2 decades generated considerable optimism that better antipsychotic treatments for schizophrenia and bipolar disorder were possible. SGAs offer several tolerability benefits over first-generation antipsychotics (FGAs), particularly with respect to extrapyramidal symptoms. However, SGAs can induce serious metabolic dysregulations, especially in drug-naive, first-episode, and child and adolescent populations, with olanzapine and clozapine having the highest propensity to cause these abnormalities. In this conte

Response to "Unmeasured confounding by mental health and chronic pain: a cautionary note on the causal interpretation of nonrestorative sleep and healthcare costs".

Morphological and biophysical techniques described in this study have shown that membrane derangement occurs in human cataractous lenses. The data suggest that these disruptions were globules, vacuoles, multilamellar membranes and clusters of highly undulating membranes. Deleterious structural damage of the lens fibre cell plasma membranes serve as the primary light-scattering centres that cause the observed lens opacity. Nuclear cataract, a major cause of loss of lens transparency in the aging human, has been thought to be associated with oxidative damage, particularly at the site of the nucl

PBRM1 knockdown is associated with NRF2‑dependent ALDOA upregulation and metabolic reprogramming in HCCC‑9810 intrahepatic cholangiocarcinoma cells.

IMPORTANCE: The coronavirus disease 2019 (COVID-19) was the third leading cause of mortality in the United States for three years in a row. The genetic contributions to disease severity remain unclear and many previously identified single nucleotide polymorphisms (SNPs) have not been replicated nor linked with functional significance. OBJECTIVE: To identify SNPs associated with mortality among hospitalized COVID-19 patients supplemented by expression quantitative trait loci (eQTL) evidence to infer plausible functional mechanisms related to COVID-19 severity. DESIGN: A quality-controlled gen

Harnessing Artificial Intelligence to Restore Standards in Medical and Dental Education in Pakistan.

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l

Effect of participatory ergonomics program on reducing ergonomic risk factors among the aging workers on ginger peelers in Wat Puranawas community, Thailand.

Iridocyclitis and the use of glucocorticoid medication have been widely studied as susceptibility factors for cataracts. However, the causal relationship between them remains unclear. This study aimed to investigate the causal relationship between the development of iridocyclitis and the genetic liability of glucocorticoid medication use on the risk of senile cataracts occurrence by performing Two-sample Mendelian randomization (MR) analyses. Instrumental variables (IVs) significantly associated with exposure factors (P < 5 × 10-8) were identified using published genome-wide association data f

A decade of intracapsular cataract extraction: Clinical profile and visual outcomes at Hospital Melaka, Malaysia.

INTRODUCTION: Cataract is a leading cause of visual impairment globally and in Malaysia, with surgery being the definitive treatment. While intracapsular cataract extraction (ICCE) is now less commonly performed due to advancements in surgical techniques, it remains essential in specific cases, such as those with weak zonular support. This study aims to evaluate the incidence, clinical profiles, and visual outcomes of patients who underwent ICCE, with or without intraocular lens (IOL) implantation, at Hospital Melaka, Malaysia, over the past decade. MATERIALS AND METHODS: A retrospective revi
