Targeting the self-amplifying loop between ATF6 and SNAI1 to inhibit epithelial‒mesenchymal transition in fibrotic lesions.

Background: The epithelial‒mesenchymal transition (EMT) decisively contributes to human diseases such as organ fibrosis and tumors. However, the molecular mechanism triggering EMT remains unclear. Methods: We elucidated a novel self-amplifying feedback loop involving activating transcription factor 6 (ATF6) and Snail family transcriptional repressor 1 (SNAI1) and assessed its significant role in unfolded protein response (UPR)-dependent EMT. Multiple in vivo and in vitro models were applied, including mouse models of trauma and laser-induced lens injury and human lens epithelial explants from

Plasma N-glycan signature as biomarker for primary open angle glaucoma.

Primary open angle glaucoma (POAG) is a major subtype of glaucoma and a leading cause of irreversible visual impairment and blindness worldwide, but with elusive pathogenesis. Here we aimed to delineate the N-glycan profile in the plasma samples of POAG patients and compare to that of the mild senile cataract subjects. Fasting peripheral whole blood was collected from 44 unrelated Han Chinese POAG patients and 39 mild senile cataract subjects. The plasma samples were isolated and subjected to the N-glycan profiling analysis by high throughput fluorophore-assisted capillary gel electrophoreses

Full-thickness macular hole after uneventful phacoemulsification cataract surgery: Case report and review of literature.

A 61-year-old healthy female patient underwent an uneventful left phacoemulsification procedure under local anesthesia for a left visually significant senile cataract. One month postoperatively, her decimal visual acuity improved from 0.06 preoperatively to 0.5. However, 3 months postoperatively, the best-corrected visual acuity (BCVA) had decreased to 0.3. Fundus examination revealed the development of a full-thickness macular hole (FTMH). Diagnosis was confirmed by optical coherence tomography of the macula, which demonstrated FTMH. The patient subsequently underwent pars plana vitrectomy wi

Plasma caffeine level as a protective factor against age-related eye diseases: a two-sample Mendelian randomisation study.

Traditional studies examining caffeine intake and age-related eye diseases (ARED) have shown inconsistent results, potentially related to variations in caffeine assessment methods. This two-sample Mendelian randomisation study investigated associations between plasma caffeine and four ARED: senile cataract, diabetic retinopathy (DR) and glaucoma and age-related macular degeneration (AMD). Summary data on genetically predicted plasma caffeine came from a genome-wide association study of 9876 European-ancestry participants across six population-based studies. ARED data were extracted from FinnGe

[Amyloidogenesis and neurotrophic dysfunction in age-related macular degeneration in correlation with Alzheimer's disease].

The pathogenesis of diseases associated with amyloid deposition in various organs and tissues has been a concern for researchers and clinicians since their discovery. Particular attention is paid to the relationship between amyloidogenesis and neurotrophic disorders in age-related neurodegenerative pathology. In this context, the amyloid cascade theory and the neurotrophic dysfunction theory remain relevant, as evidenced by the results of numerous studies conducted in recent years. Meanwhile, it has been shown that amyloidosis, being a systemic pathological process, affects ocular tissues and

Modifiable Dietary Habits, Inflammatory Mediation, and Senile Cataract: Unraveling Causality via Mendelian Randomization.

Senile cataract (SC) is a major cause of global vision impairment and blindness, posing a significant public health challenge in aging populations. This study employed Mendelian randomization (MR) using genetic instruments for 83 dietary habits from the UK Biobank GWAS, with SC data from the FinnGen R12 as the outcome. We employed multiple MR methods, including bidirectional MR, replication sample MR, and multivariable MR (MVMR), to identify dietary habits significantly associated with SC. The potential mediating effects of inflammatory biomarkers were further assessed using a two-step MR. Fin

A Comparative Study of Surgically Induced Astigmatism From Temporal and Steep-Axis Clear Corneal Incisions in Phacoemulsification Cataract Surgery.

Background Surgically induced astigmatism (SIA) is a critical determinant of visual outcomes after phacoemulsification cataract surgery. The choice of incision site significantly influences postoperative refractive stability. This study compares temporal clear corneal incision (CCI) and steep-axis CCI in terms of SIA, uncorrected visual acuity (UCVA), and corneal stability. Methods This prospective randomized study included 130 patients with senile cataract and preoperative corneal astigmatism ≤1.5 D (diopters), allocated into two groups: Group A (temporal incision, n = 65) and Group B (steep-

Organic electrolyte gated field effect transistor for detecting alpha synuclein longitudinally in Parkinsonism mouse model.

BACKGROUND: Enthesopathy is an evolving area for applied clinical research. MRI is the gold standard in the diagnosis of elbow joint pathology, but recent reports indicate that ultrasound imaging is more sensitive and accurate than MRI in detecting enthesopathy of the heels and knees. Too many patients are under-diagnosed and/or misdiagnosed because the early pathological changes of enthesitis in the different types of seronegative arthropathies are not detected. OBJECTIVES: This study was undertaken to describe the ultrasound features of elbow enthesitis in patients with seronegative arthrop
