Comparative analysis of human γD-crystallin aggregation under physiological and low pH conditions.

Cataract, a major cause of visual impairment worldwide, is the opacification of the eye's crystalline lens due to aggregation of the crystallin proteins. The research reported here is aimed at investigating the aggregating behavior of γ-crystallin proteins in various incubation conditions. Thioflavin T binding assay, circular dichroism spectroscopy, 1-anilinonaphthalene-8-sulfonic acid fluorescence spectroscopy, intrinsic (tryptophan) fluorescence spectroscopy, light scattering, and electron microscopy were used for structural characterization. Molecular dynamics simulations and bioinformatics

Non-oxidative modification of lens crystallins by kynurenine: a novel post-translational protein modification with possible relevance to ageing and cataract.

In humans, the crystallin proteins of the ocular lens become yellow-coloured and fluorescent with ageing. With the development of senile nuclear cataract, the crystallins become brown and additional fluorophores are formed. The mechanism underlying crystallin colouration is not known but may involve interaction with kynurenine-derived UV filter compounds. We have recently identified a sulphur-linked glutathionyl-3-hydroxykynurenine glucoside adduct in the lens and speculated that kynurenine may also form adducts with GSH and possibly with nucleophilic amino acids of the crystallins (e.g. Cys).

Glutathione: a vital lens antioxidant.

The reducing compound glutathione (GSH) exists in an unusually high concentration in the lens where it functions as an essential antioxidant vital for maintenance of the tissue's transparency. In conjunction with an active glutathione redox cycle located in the lens epithelium and superficial cortex, GSH detoxifies potentially damaging oxidants such as H2O2 and dehydroascorbic acid. Recent studies have indicated an important hydroxyl radical-scavenging function for GSH in lens epithelial cells, independent of the cells' ability to detoxify H2O2. Depletion of GSH or inhibition of the redox cycl

Age-related changes in the kinetics of water transport in normal human lenses.

Magnetic resonance microscopy (MRM) has been used to study the kinetics of water transport in human eye lenses. Fresh lenses obtained from the Queensland Eye Bank were incubated at 34.5 degrees C in artificial aqueous humour (AAH) containing nutrients and metabolites similar to those that are present in vivo. MR images were acquired over approximately a 20 hr period following replacement of H(2)O based AAH with deuterium oxide (D(2)O) based AAH. NMR signal intensity from the lenses decreased with time corresponding to a decrease in concentration of H(2)O within the lenses. A statistically sign

Regulation of indoleamine 2,3-dioxygenase, the first enzyme in UV filter biosynthesis in the human lens. Relevance for senile nuclear cataract.

3-Hydroxykynurenine (3OHKyn), the precursor of UV filters in human lens, is highly autooxidizable, generates H2O2, and binds to lens proteins, yielding a tanned/yellow product resembling senile nuclear cataractous materials. Thus, if 3OHkyn can be shown to be the causative agent in cataract, it may be possible to prevent the disease by lowering the level of 3OHKyn. To this end, indoleamine 2,3-dioxygenase, the first enzyme in UV filter synthesis, was studied using lens epithelial cell lines. The results indicated that the IDO expression is mediated by IFN-gamma. Immuno-suppressants which inhib

Oxidation products of 3-hydroxykynurenine bind to lens proteins: relevance for nuclear cataract.

3-Hydroxykynurenine (3OHKyn), present as a human lens UV filter, has also been implicated as a carcinogen and neurotoxin. It has been suggested that oxidation of 3OHKyn is involved in each of these effects. In the presence of oxygen, 3OHKyn has been found to react with bovine crystallins, to give brown-coloured products (Stutchbury and Truscott, 1993). In this study the roles of UV-light, pH, glutathione and oxygen were examined, with the objective of determining how these factors may affect the binding of 3OHKyn to crystallins under the conditions found within the lens itself. The presence of

An unusual coloured compound in digests of senile nuclear cataract lens proteins.

An unusual pink compound has been isolated from the proteolytic digests of senile nuclear cataract proteins by adsorption onto Sephadex G-10. The tiny amounts obtained precluded definitive structural elucidation, however TLC comparisons show that it resembles triphenodioxazine-1,8-dicarboxylic acid, a pink heterocyclic chemical formed by oxidative condensation of 3-hydroxyanthranilic acid.

Development of fluorescence and cross-links in eye lens crystallin by interaction with lipid peroxy radicals.

Senile nuclear cataract formation is associated with the accumulation of fluorescent insoluble proteins. alpha-Crystallin from bovine eye lens was treated with the lipid peroxy radicals generated by interaction of linoleic acid 13-monohydroperoxide or phosphatidylcholine hydroperoxide with methemoglobin. A blue non-tryptophan fluorescence composed of at least two kinds of fluorophores, stable and unstable on borohydride treatment, was produced. One of the stable fluorophores was identified as dityrosine by comparison of its retention times in amino acid analysis and HPLC with those of authenti

Decolouration of the lens pigment in senile nuclear cataract.

Human cataract lens proteins can be bleached by exposure to sodium borohydride (NaBH4), sodium cyanoborohydride (NaCNBH3), or hydrogen peroxide (H2O2). The decolouration resulting from these treatments could be monitored by a change in absorbance at 350 nm. At pH 12 the magnitude of the absorbance change increased in proportion with the severity of the nuclear cataract in the case of NaBH4 and H2O2 treatments, but not in the case of NaCNBH3 treatment. The rate of change in absorbance at 350 nm following exposure to the different reagents was used to evaluate three model systems for senile nucl

[Trace element contents in the transparent lens and the senile cataract lens].

The contents of trace elements Zn, Mn, Cu and Fe in 38 senile cataract lenses and 37 transparent lenses were determined by atomic absorption spectrophotometry. It was found that the contents of the elements in senile cortical cataract lenses were markedly different from those in transparent lenses. Comparing transparent lenses with senile nuclear cataract lenses, there was only difference in the content of Mn. It was therefore inferred that the occurrence of senile cataract was directly related to the deficiency of trace elements.

The reaction of proteins with 3-hydroxyanthranilic acid as a possible model for senile nuclear cataract in man.

Proteins, including lens proteins, were incubated in the presence of 3-hydroxyanthranilic acid (30 HA) under oxidizing conditions. Samples were monitored for alterations in color, fluorescence, sulfhydryl content, lysine availability, methionine content, tryptophan content and protein size. Incubation of proteins with 30 HA produced rapid brown coloration and a correspondingly rapid decrease in sulfhydryl content. Alpha-, beta- and gamma-crystallins were all found to react with 30 HA. An increase in protein fluorescence (excitation 340/emission 425 nm) accompanied the color development. No sig

The state of sulphydryl groups in proteins isolated from normal and cataractous human lenses.

Individual crystallins, urea-soluble and urea-insoluble proteins were isolated from the nucleus and cortex of types I-IV cataractous lenses and normal lenses. The levels of protein sulphydryls (P-SH), disulphides (S-S), as well as surface (F-SH) and buried (S-SH) in these proteins were determined by reaction with 5, 5'-dithiotris- (2-nitrobenzoic acid) or performic acid oxidation followed by amino acid analysis. During nuclear colour development there is a progressive decrease in the sulphydryl content of the crystallins. In the nuclei of advanced cataractous lenses, the P-SH decreases to 10%

Selenium-induced cataract--a correlation of dry mass content and light scattering.

Selenium-induced cataracts in young rats were used to compare light scattering and dry mass concentration along the lens axis. Selenium-treated rats and control rats were examined 1, 2, 4 and 6 days after subcutaneous injection of selenium or 0.9% NaCl. The development of nuclear cataract was followed with a Scheimpflug slit-lamp camera. Light-scattering was determined by densitometry of the negatives taken by the Scheimpflug camera. Dry mass concentration was determined by quantitative microradiography of the extracted lenses. Increasing light scatter was seen from day 2 to day 6. The dry mas
