Left-Right Determination Factor 2 (LEFTY2) Is an Aqueous Humor Biomarker for Exfoliation Glaucoma.

PURPOSE: Exfoliation syndrome (XFS) is characterized by deposition of exfoliation material (XFM) in the anterior segment. XFM contributes to development of exfoliation glaucoma (XFG). Here, we evaluated left-right determination factor 2 (LEFTY2), a TGFβ superfamily protein, as an XFS and XFG biomarker. METHODS: Aqueous humor (AH) and/or blood was collected from patients undergoing cataract surgery. Controls included patients with cataracts (CAT; AH, n = 78; blood, n = 20) or cataracts and primary open-angle glaucoma (CAT/GL; AH, n = 67; blood, n = 20). Affected groups included patients with c

Altered serum biomarkers of oxidative stress and antioxidant capacity in primary open-angle glaucoma.

This study investigated the role of oxidative stress in primary open-angle glaucoma (POAG) by comparing serum levels of ischemia-modified albumin (IMA), malondialdehyde (MDA), and total thiol (TT) between POAG patients and healthy controls. The study included 48 POAG patients and 54 age- and sex-matched healthy controls. Serum concentrations of IMA, MDA, and TT were measured using established assay methods. Statistical analysis revealed significantly higher levels of IMA and MDA and lower levels of TT in POAG patients than controls (p < 0.001). The findings show that patients with POAG tend to

Mitochondria transplantation reduces intraocular pressure by improving mitochondrial function and remodeling trabecular meshwork.

BACKGROUND: Mitochondrial transplantation represents a promising therapeutic strategy for diseases characterized by mitochondrial dysfunction, with mitochondrial dysfunction in trabecular meshwork (TM) recognized as a key pathogenic mechanism in primary open-angle glaucoma (POAG) where impairment drives sustained intraocular pressure (IOP) elevation. OBJECTIVE: This study aimed to develop a novel therapeutic approach through direct mitochondria transplantation to replenish mitochondrial quantity in TM cells. METHODS: The healthy mitochondria were isolated from mouse liver tissues, and uptake b

Pro-Homeostatic Effects of Estrogen on Intraocular Pressure and the Trabecular Meshwork.

PURPOSE: Elevated intraocular pressure (IOP) results from the dysregulation of aqueous humor outflow through the mechanoresponsive trabecular meshwork (TM). Evidence suggests that low estrogen (E2) and high TGFβ2 levels are risk factors for elevated IOP and primary open-angle glaucoma. We sought to examine whether E2 signaling is involved in TM regulation of IOP homeostasis. METHODS: IOP and central cornea thickness were measured in male and female Esr1-/- mice and wild-type littermate controls from 3 to 12 months of age. Primary human TM cells (n = 10, 6 females and 4 males) were treated wit

Ferroptosis regulator NOX1 acts a diagnostic biomarker and mediates disease progression with the transcriptional regulation of STAT3 in glaucoma.

PURPOSE: Primary open-angle glaucoma (POAG) is the leading cause of irreversible blindness. Regrettably, the roles of ferroptosis-related (FR) genes in POAG remain elusive. DESIGN: Five GEO data sets and a series of experimentations in vitro were used for bioinformatic exploration and biological validation. METHODS: Using multiple machine learning algorithms, four critical FR genes in POAG progression were screened. The clinical value and biological function of NOX1 were comprehensively analyzed using bioinformatic approaches. A POAG in vitro model was constructed using H2O2 treatment. NOX1
