Exploring the causal link between long-term elevated plasma caffeine levels and age-related eye diseases using Mendelian randomization.

OBJECTIVE: Experimental evidence suggests that caffeine has the potential to reduce the risk of age-related eye diseases (AREDs). However, there is a lack of population-based evidence directly assessing whether long-term caffeine exposure benefits these conditions. In this study, we employ Mendelian randomization (MR) to evaluate the causal effects of long-term elevated plasma caffeine levels (PCL) on AREDs, including age-related macular degeneration (AMD), age-related cataract (ARC), and glaucoma. METHODS AND ANALYSIS: Single-nucleotide polymorphisms strongly associated with PCL and located

Macrophage Migration Inhibitory Factor (MIF)-CD74 Signaling Pathway Mediates Trabecular Meshwork Dysfunction in Glaucoma.

PURPOSE: Macrophage migration inhibitory factor (MIF) is a pleiotropic cytokine implicated in many inflammatory and fibrotic diseases; however, its role in primary open-angle glaucoma (POAG) and trabecular meshwork (TM) dysfunction remains unknown. In this study, we investigated whether MIF-CD74 signaling regulates TM pathobiology through modulation of the transcription factor, Blimp-1, and downstream cytoskeletal reorganization and extracellular matrix (ECM) remodeling. METHOD: Primary human TM cells (HTMC) were exposed to glaucomatous stressors, including TGF-β2, rMIF, or a pro-inflammatory

Chromatin dynamics identifies 78 genes at loci associated with elevated intraocular pressure and primary open-angle glaucoma.

Primary open-angle glaucoma (POAG) is a chronic neurodegenerative disorder, and elevated intraocular pressure (IOP) represents the major, and only modifiable, risk factor for the disease. We modeled increased IOP by treating three primary human trabecular meshwork (TM) cell strains with dexamethasone, then generated a high-resolution map of promoter-centered chromatin contacts and regulatory modules to decipher how the genomic architecture and epigenetic state of disease-associated loci contribute to pathogenesis. We identify dynamic changes in chromatin compartments and looping, cis-regulator

Bioengineered 3D Human Trabecular Meshwork Models for Outflow Physiology and Glaucoma Research.

Primary open angle glaucoma (POAG) is one of the leading causes of irreversible blindness and is associated with dysfunction of the trabecular meshwork (TM), a three-dimensional (3D) structure that regulates aqueous humor outflow and, consequently, intraocular pressure (IOP). IOP is the only modifiable factor for glaucoma. Outflow facility is the inverse of aqueous humor outflow resistance caused by the presence of the TM and adjacent tissues, and reflects the TM's central role in IOP control, representing the most physiologically relevant measure of human trabecular meshwork (HTM) function. Variant-to-gene mapping identifies ARHGEF12 as a primary open-angle glaucoma effector gene operating within retinal ganglion cells.

Primary open-angle glaucoma (POAG), a leading cause of irreversible blindness, has a strong genetic basis. The Primary Open-Angle African Ancestry Glaucoma Genetics study previously identified 46 risk loci. To pinpoint causal variants and their corresponding effector genes, we analyzed gene expression, chromatin accessibility, and conformation in two ocular cell-types: trabecular meshwork cells (hTMCs) and retinal ganglion cells derived from induced pluripotent stem cells (hiPSC-RGCs). We identified 24 candidate genes in hTMCs and 56 in hiPSC-RGCs. The ARHGEF12 gene was selected for further va

Association of neovascular glaucoma with increased mortality and cardiovascular risk: a multicenter propensity-matched study.

Neovascular glaucoma (NVG) is a severe, vision-threatening secondary glaucoma associated with ocular ischemia and systemic vascular disease. Whether NVG also serves as a marker of systemic vascular risk remains unclear. In this retrospective, multicenter cohort study, adults with NVG were identified from a global electronic health record network and 1:1 propensity score matched to controls, accounting for demographics, comorbidities, medications, and healthcare utilization. Follow-up was up to 10 years. Primary outcomes were all-cause mortality, myocardial infarction, stroke, cardiac arrest, a

Anterior segment, static and dynamic pupillography changes in patients with primary open-angle glaucoma using different types of topical antiglaucomatous agents.

INTRODUCTION: Glaucoma is a major cause of avoidable blindness and the most common type is primary open angle glaucoma (POAG). Topical antiglaucomatous agents are the mainstay of treatment. There are many studies on the ocular effects of antiglaucomatous agents and these results may contradict each other. The aim of our study was to investigate the effects of topical antiglaucomatous agents on the anterior segment and pupil in patients with POAG. MATERIALS AND METHODS: The right eye of the patients using dorzolamide+timolol, brimonidine, latanoprost and the right eye of the control group were

Intraocular pressure induced blood retinal barrier compromise in mouse models and human glaucoma.

The integrity of the blood-retinal barrier remains largely unexplored in glaucoma. Here we show that elevated intraocular pressure partially compromises the blood-retinal barrier in three distinct mouse models of glaucoma. This compromise induces subtle leakage without bleeding or detectable disruption of endothelial cell junctions and precedes neurodegeneration. Leakage occurs from peripheral veins in the retinal ganglion cell layer with a concomitant loss of the transcytosis inhibitor MFSD2A. RNA-sequencing, protein analyses and electron microscopy support enhanced transcytosis as the mechan
