A Comparative Analysis of Different MRI Sequences Dedicated for Invivo Detection of Mineralization in a Single Large Cohort of Progressive Supranuclear Palsy and Healthy Controls.

Iron plays an important role in normal brain metabolism. Increase in brain iron is a part of the normal brain development, but an excessive deposition of brain iron may be a risk factor for degenerative diseases. At the clinical setting, there are two major issues of controversy. One is that of interpreting the varying degree of mineralization, which occurs with normal aging, which is physiological from pathological mineralization. The second issue is interpretating and drawing a single conclusion, as to whether mineralization is occurring or not from the various MRI sequences available. Vario

Metabolic stress and early cell death in photoreceptor precursor cells following retinal transplantation.

BACKGROUND: Progressive photoreceptor loss in retinal degenerative diseases leads to irreversible vision impairment. Transplantation of human embryonic or induced pluripotent stem cell-derived photoreceptor precursor cells (PRPCs) offers potential for vision restoration. However, substantial early donor cell loss remains a major challenge. This study aims to elucidate the mechanisms underlying early PRPC loss and to evaluate host retinal responses to transplantation. METHODS: PRPCs derived from human embryonic stem cells (hESC)-based retinal organoids were subretinally transplanted into both

Protein kinase G inhibition preserves photoreceptor viability and function in a new mouse model for autosomal dominant retinitis pigmentosa.

Retinitis Pigmentosa (RP) is the most common inherited retinal degeneration, characterized by an initial loss of rod photoreceptor cells. Photoreceptor cell death has been associated with high levels of cyclic guanosine-3', 5'- monophosphate (cGMP) in animal models of autosomal recessive RP (ARRP) and autosomal dominant RP (ADRP). cGMP analogues inhibiting protein kinase G (PKG) have been found to prevent rod degeneration in ARRP disease models, but their effects on ADRP are unknown. Here, we used the recently generated rhodopsin-mutant RhoI255d/+ ADRP mouse model to study cGMP-signaling and t

Inactivation of cellular retinol-binding protein 1 protects against bis-retinoid accumulation and light-induced retinal degeneration in mice.

Delayed clearance of all-trans-retinal (at-RAL) in photoreceptors is linked to prevalent retinal diseases such as Stargardt disease, rod-cone dystrophies, and age-related macular degeneration. Pharmacological modulation of retinoid metabolism in the eye presents a promising therapeutic strategy, with cellular retinol-binding protein 1 (RBP1) emerging as a potential target. However, it lacks genetic validation as a therapeutic target in an animal model of human disease. Thus, we investigated the effect of the Rbp1 gene inactivation on the phenotype of the Abca4-/-/Rdh8-/- model of Stargardt dis

Optogenetic tools and their applications for therapeutic intervention in end-stage inherited retinal diseases.

Retinal degenerative diseases pose significant therapeutic challenges; however, optogenetic vision restoration offers a promising strategy that is independent of mutations and disease progression. Optogenetic tools consisting of photosensitive membrane proteins can be ectopically expressed in retinal cells, effectively converting them into artificial photoreceptors. This review outlines the development and application of optogenetic tools to restore light sensitivity in degenerated retinas. A thorough understanding of retinal cell morphology, pathological remodeling, and functional loss is cri

Shining light on photoreceptors: A minireview on the development and clinical applications of optoretinography.

Retinal diseases often result in photoreceptor dysfunction and cell death, leading to progressive vision impairment and eventual blindness. Clinical management can benefit from assessing photoreceptor mediated visual function, such as for progression monitoring. Meanwhile, therapeutic development requires accurate and reliable vision end points. The emerging optoretinography (ORG) technologies promise noninvasive, objective, and highly sensitive markers of photoreceptor function. ORG is an umbrella term that covers several imaging modalities that use intrinsic optical signal, i.e., without dye

Development and validation of machine learning for pathological changes in high myopia patients with tessellated fundus: a cross-sectional study.

PURPOSE: Pathological myopia causes irreversible damage, yet early changes in high myopia patients with tessellated fundus are often overlooked. The identification of such patients relies on OCT and fundus imaging, but early detection is difficult. Therefore, we developed and validated machine learning models using lifestyle and clinical parameters. METHODS: Cross-sectional data were collected from high myopia patients with tessellated fundus at the First Affiliated Hospital of Guangxi Medical University (May 2023-July 2024). Data were split (7:3) into training and test sets. Five-fold cross-

Glaucoma diagnosis and treatment in the era of precision medicine.

Glaucoma is a chronic, degenerative eye disease for which personalized diagnostic and therapeutic biomarkers are still lacking. Precision medicine offers a promising strategy to improve glaucoma management, and pharmacogenomics may play a key role in optimizing treatment response, minimizing side effects, and enhancing adherence. Advances in genetics have introduced potential tools such as polygenic risk scores (PRS) for adult-onset glaucoma and CRISPR/Cas9 for early-onset monogenic forms. Although still in its early stages, pharmacogenetic research in glaucoma has identified several genetic v

Deceleration Capacity as a Marker of Autonomic Cardiac Modulation in Prodromal and Manifest Parkinson's Disease, Multiple System Atrophy, and Progressive Supranuclear Palsy.

BACKGROUND: Degenerative parkinsonian syndromes, including the alpha-synucleinopathies (aSYN) Parkinson's disease (PD), and multiple system atrophy (MSA), and the tauopathy progressive supranuclear palsy (PSP), are characterized by motor and non-motor symptoms. The later subsume autonomic dysfunction, which may appear early or progress with the disease. Cardiac dysfunction varies by syndrome and can also occur in isolated REM sleep behavior disorder (iRBD), a prodromal stage of aSYN. Overlapping motor features make early differentiation challenging. Heart rate variability (HRV) analysis is a n

Enhancing Cerebellar Findings with Positional Manoeuvres.

The positional characteristics of vertigo are often attributed to a peripheral origin, which may lead to diagnostic errors. Downbeat nystagmus (DBN), a marker of cerebellar dysfunction, can be imperceptible in primary gaze and only become evident after positional manoeuvres, which tend to enhance an otherwise subtle nystagmus. We present two elderly patients with progressive cerebellar ataxia and DBN that was barely present in primary gaze but clearly enhanced in the supine position. Both exhibited additional central oculomotor signs and bilateral vestibulopathy, without significant structural
