Presence of lower limb joint arthritis moderates the interrelationship between vision impairment and fear of falling avoidance behavior in older adults: A cross-sectional study.

Falling is a common concern for older adults. Previous studies showed that mobility performance is significantly lower in those with age-related eye diseases or lower limb arthritis, contributing to their increased fall risk. However, it is currently unknown how the co-presence of these 2 age-related sensorimotor impairments interacts to affect fear of falling (FOF) and associated activity-avoidance behaviors. A total of 200 participants recruited from an outpatient regional medical center (127 women, 73 men, age = 65.5 ± 8.4 years) participated in this study. The presence of age-related, unco

Systemic Loss of Chromosome Y Associates With Age-Related Ophthalmic Diseases in Middle-Aged Men: A Longitudinal Multi-Omic Study.

Maintaining health during global population aging represents a critical challenge, where mosaic loss of chromosome Y (LOY) in the hematopoietic system surfaces as a prevalent somatic mutation in aging men. However, its longitudinal impact on sensory organ degeneration remains insufficiently characterized. Analyzing 223,051 male participants from the UK Biobank, we demonstrated that LOY uniquely and independently predicted an increased risk of incident age-related macular degeneration (AMD) (Hazard Ratio = 0.611, 95% Confidence Interval: 0.397-0.942). Restricted cubic splines identified a criti

Age- and sex-dependent retinal degeneration in peripherin-2 mutant mice, a model of central areolar choroidal dystrophy.

Central areolar choroidal dystrophy (CACD) is a progressive macular dystrophy without treatment. Although PRPH2 mutations are the most common cause of disease, the mechanisms driving their phenotypic variability remain poorly understood. Here, a comprehensive characterization of the pathophysiological consequences of p.Arg195Leu mutation in PRPH2 was carried out in a mouse model (Prph2KI/WT). For evaluating the retinal degeneration, this study combines the analysis of electroretinographic responses together with a complete study of bulk RNA-seq transcriptomics and the key cellular and molecula

Nano formulations of quercetin for ocular delivery.

Quercetin (QUE), a bioactive flavonoid with potent antioxidant, anti‑inflammatory, and neuroprotective properties, has emerged as a promising therapeutic candidate for age‑related macular degeneration, diabetic retinopathy, cataracts, glaucoma, dry eye disease, and allergic conjunctivitis. Its therapeutic effects are primarily mediated through modulation of key molecular pathways, including direct scavenging of reactive oxygen species (ROS), activation of Nrf2‑dependent antioxidant responses, inhibition of NF‑κB and MAPK signaling, and downregulation of pro‑inflammatory cytokines and angiogeni

Application of nanozyme-based drug delivery systems in the treatment of ROS-related ophthalmic diseases.

ROS-related ophthalmic diseases span the ocular surface as well as the anterior and posterior segments of the eye. Dry eye disease (DED), glaucoma, diabetic retinopathy (DR) and age-related macular degeneration (AMD) have distinct anatomical targets but share common pathophysiological features, including oxidative stress, inflammation, vascular dysfunction, epithelial injury or neurodegeneration. Conventional ocular drug delivery and therapeutic modalities are hindered by severe limitations, including low bioavailability in the posterior segment of the eye and the requirement for frequent inva

Examination of Alzheimer Disease and Antidementia Medications' Effect on Age-Related Macular Degeneration.

PURPOSE: To clarify the prevalence of age-related macular degeneration (AMD) in Alzheimer disease and assess whether antidementia medications modify this relationship. METHODS: A retrospective cohort study was performed using electronic health record data (TriNetX) of adults aged 65 years and older with 10 or more years of ophthalmology follow-up. Patients with Alzheimer disease were compared with controls to estimate unadjusted and propensity score-adjusted prevalence odds ratios (ORs) for any AMD and progression to advanced AMD. Within the Alzheimer disease cohort, associations between dura

Comparative Evaluation of EfficientNet-B3, Vision Transformers, and RETFound for Seven-Category Retinopathy Fundus Image Classification.

Background/Objectives: Retinal fundus photography is the most widely available means of screening for sight-threatening disease, but it is difficult to read: several conditions leave overlapping signs on the same photograph, the features that separate them are often subtle and confined to small regions of the retina, and a single image carries no systemic context, so an automated reader must resolve categories on which experienced clinicians themselves agree only moderately. Methods: We compared five deep learning architectures-EfficientNet-B3, DenseNet-121, ViT-Base/16, Inception-V3, and RETF

RNA base-modification and A-to-I editing landscapes of oxidatively stressed human retinal pigment epithelium under Quantum Molecular Resonance electrotherapy and blood-derived secretome.

Chemical modifications of RNA bases and adenosine-to-inosine (A-to-I) editing are regulatory layers essentially unexplored in the retinal pigment epithelium (RPE), whose oxidative dysfunction underlies age-related macular degeneration. We mapped both layers in ARPE-19 cells exposed to tert-butyl hydroperoxide under two non-pharmacological interventions, Quantum Molecular Resonance (QMR) electrotherapy and a patient blood-derived secretome, across 56 libraries spanning four arms, three time points and matched stressed and stress-free conditions (GEO: GSE328981). [Subretinal fibrosis in neovascular age-related macular degeneration: risk factors, mechanisms of development, and current options for prevention and treatment].

The results of numerous randomized clinical trials and clinical practice have demonstrated the effect of angiogenesis inhibitors (anti-VEGF agents) on the activity and growth of the neovascular complex in neovascular age-related macular degeneration (nAMD). At the same time, a reduction in the risk of macular fibrosis has been demonstrated, with its incidence decreasing from 39-100% in the "pre-antiangiogenic era" to 20.4-61.4% during anti-VEGF therapy. The main factors determining the risk of subretinal fibrosis are disease activity, timing of treatment initiation, treatment regimen and regul

Metabolic and Immunometabolic Neuroprotection in Glaucoma: Mitochondrial Dysfunction, Neuroinflammation, and Emerging Therapeutic Strategies.

Glaucoma is increasingly recognized as a multifactorial neurodegenerative optic neuropathy in which intraocular pressure (IOP) is one of several stress amplifiers rather than a sufficient etiological explanation. Vascular dysregulation, mitochondrial and metabolic vulnerability, oxidative stress, excitotoxicity, glial dysregulation, and persistent neuroimmune activation can converge on retinal ganglion cell (RGC) and optic nerve injury. This broader framework is particularly relevant to normal-tension glaucoma (NTG), which represents a large proportion of primary open-angle glaucoma in Asian p

Retinal Ganglion Cell Quantification for Evaluating Neurodegeneration in Mouse Models of Multiple Sclerosis.

Quantification of retinal ganglion cell (RGC) loss provides a sensitive and reproducible measure of neurodegeneration in mouse models of neurological  disease and injury. In experimental autoimmune encephalomyelitis (EAE), a widely used mouse model of multiple sclerosis, inflammation of the optic nerve can lead to optic neuritis, axonal injury, and progressive RGC loss. Therefore, quantification of RGC survival provides a valuable readout of inflammation-mediated neurodegeneration and can be used to evaluate disease severity and neuroprotective interventions. Here, we describe a protocol for r

Acquired Corneal Subepithelial Hypertrophy: First Ultrastructural Characterization by Transmission Electron Microscopy.

Acquired corneal subepithelial hypertrophy (ACSH) is a rare corneal condition whose ultrastructural features have not previously been characterized. We report the first detailed ultrastructural analysis of ACSH in a 57-year-old man with a history of penetrating keratoplasty for corneal trauma followed by Descemet stripping automated endothelial keratoplasty who presented with progressive visual decline due to a superficial corneal opacity. Clinical evaluation included slitlamp biomicroscopy, anterior segment optical coherence tomography, corneal topography, and pachymetry, which localized the

Therapeutic Window of AAV-SIRT1 Gene Therapy in Mouse Models of Optic Nerve Degeneration.

PURPOSE: Optic neuropathies involve progressive loss of retinal ganglion cells (RGCs) and their axons, leading to irreversible vision deficits. While pretreatment with drugs or gene therapy that upregulate sirtuin 1 (SIRT1), an NAD⁺-dependent deacetylase, has shown neuroprotective effects, its effectiveness when administered after disease onset remains uncertain. We investigated whether adeno-associated virus (AAV)-mediated SIRT1 overexpression in RGCs can provide post-onset neuroprotection in preclinical models of optic neuropathy. METHODS: RGC degeneration was induced in C57BL/6 mice (8-10/

What is the patient experience of receiving lifestyle advice regarding age-related macular degeneration (AMD) in secondary eye care? A multimethods study.

OBJECTIVES: Age-related macular degeneration (AMD) is a progressive condition which can lead to irreversible sight impairment. While there is no cure for early or intermediate AMD, modification of lifestyle risk factors (smoking, dietary changes and vitamin supplementation) can slow down disease progression. Although professional guidelines recommend that clinicians provide lifestyle advice to patients, evidence suggests that this may not be provided consistently. This study aimed to investigate the experiences of people with AMD receiving lifestyle modification advice in secondary eye care to

Wavelength and polarization optimization through Stokes parameters for second harmonic generation microscopy in corneal tissue.

Second harmonic generation (SHG) microscopy has experienced a research expansion from fundamental collagen studies in tendon and skin to a growing body of work on ocular tissues, where the label-free and high-resolution aspects have offered unique possibilities. Nevertheless, polarization states and excitation wavelength have not been investigated for corneal samples. Here, we present a comprehensive investigation of SHG microscopy applied to fibrillar collagen from murine corneal tissue. Emphasis is given to polarization control, cross-polarization effects, and excitation wavelength optimizat

Intrafamilial phenotypic variation in four Palestinian siblings with CFAP418-related ciliopathy: a familial case series.

BACKGROUND: Biallelic CFAP418 variants cause retinal degeneration with variable extraocular findings. The principal contribution of this familial case series is a comparison of four affected siblings carrying the same previously reported splice-donor variant, with explicit separation of original clinical records from recalled history and historical summaries. CASE PRESENTATION: Four Palestinian siblings from a consanguineous family had homozygosity for CFAP418 NM_177965.3:c.155 + 1G > A reported in a 2018 family research letter; a 2019 clinical laboratory report confirmed the variant in the o

Dynamic frailty trajectories and risk of age-related macular degeneration: a prospective cohort study in UK Biobank.

Age-related macular degeneration (AMD) is a leading cause of irreversible vision loss in the aging population. Although frailty has been recognized as a potential risk factor, previous studies relying on static assessments cannot capture its dynamic nature. This study aims to prospectively evaluate the association between dynamic frailty trajectories and incident AMD. The baseline cohort included 462,573 UK Biobank participants who were free of AMD and had frailty phenotype (FP) data at enrollment. Among them, 53,059 with at least one follow-up FP assessment comprised the trajectory cohort. Pa

The Eye-Brain Axis in High Myopia: From Retinal Deformation to Cortical Network Reorganization.

High myopia is a major cause of irreversible visual impairment and is increasingly recognized as a condition involving interactions between ocular remodeling and central neural systems. This scoping review synthesizes current evidence on structural, functional, genetic, and regulatory links between the eye and brain in high myopia. We systematically searched PubMed and Web of Science from database inception to August 2026, identifying 54 eligible primary studies. The available evidence indicates that high myopia is associated with retinal and optic nerve abnormalities, altered white matter mic
