Cardiovascular disease risk prediction in multi-ethnic Asian populations: evidence from two population-based cohorts in Singapore.

BACKGROUND: The rising burden of cardiovascular diseases (CVD) in Asia requires risk assessment tools tailored to Asian populations. Therefore, we recalibrated the ACC/AHA Pooled Cohort Equations for non-Hispanic Whites (PCE-W) and compared its performance in predicting 10-year CVD risk with two other established CVD prediction models that have been recently recalibrated for Asian populations. METHODS: We used data from the Singapore Multi-Ethnic Cohort (MEC1) and the Singapore Epidemiology of Eye Diseases (SEED) cohort comprising ethnic Chinese, Indian, and Malay participants. The PCE-W was

Hyperactive microtubule binding of RP1L1 R45W underlies retinal degeneration and is suppressed by glycerol.

Photoreceptors rely on microtubule (MT)-based transport within the connecting cilium to maintain cellular homeostasis. Mutations in RP1L1, a retina-specific doublecortin (DC) domain protein, cause inherited retinal disorders including occult macular dystrophy (OMD), yet the underlying mechanisms remain poorly defined. Here, we show that the RP1L1 R45W variant, prevalent in East Asian individuals with OMD, confers a toxic gain-of-function phenotype characterized by abnormally strong MT binding. Live-cell imaging revealed an approximately twofold increase in MT association relative to wild-type

Single-cell profiling of trabecular meshwork identifies mitochondrial dysfunction in a glaucoma model that is protected by vitamin B3 treatment.

Since the trabecular meshwork (TM) is central to intraocular pressure (IOP) regulation and glaucoma, a deeper understanding of its genomic landscape is needed. We present a multimodal, single-cell resolution analysis of mouse limbal cells (includes TM). In total, we sequenced 9,394 wild-type TM cell transcriptomes. We discovered three TM cell subtypes with characteristic signature genes validated by immunofluorescence on tissue sections and whole-mounts. The subtypes are robust, being detected in datasets for two diverse mouse strains and in independent data from two institutions. Results show

Synergistic therapy for diabetic retinopathy via dual blockade of VEGF-A and TNF-α with a bispecific nanobody.

INTRODUCTION: Anti-vascular endothelial growth factor (VEGF) therapy fails in approximately 30% of diabetic retinopathy (DR) patients, indicating the limitations of solely targeting VEGF-A. This study aimed to design a bispecific nanobody, Nb-TV, targeting both tumor necrosis factor-α (TNF-α) and VEGF-A to achieve dual anti-inflammatory and anti-angiogenic effects for potential DR treatment. METHODS: The inhibitory effects of Nb-TV were evaluated in vitro using adult retinal pigment epithelium-19 (ARPE-19) cells and human umbilical vein endothelial cells (HUVECs). Key assays included analysis

First-order basis of second-order temporal loss in amblyopia.

BACKGROUND: Amblyopia is a neurodevelopmental visual disorder characterized by monocular visual impairment due to abnormal binocular experience in early childhood, with first-order (luminance-defined) and second-order (contrast-defined) processing deficits. While first-order temporal perception deficits in amblyopia are well-documented, the nature of second-order temporal deficits remains unclear, particularly whether they stem from intrinsic extrastriate dysfunction or secondary effects of first-order spatial impairments. This study investigated temporal perception deficits in anisometropic a

Astaxanthin protects retinal Müller cells against high glucose-induced oxidative stress through the sirtuin 1/AMPK/FOXO1 pathway.

Diabetic retinopathy (DR) is characterized by microvascular damage in the retina due to hyperglycemia-induced oxidative stress. The pivotal role of Müller glial cells in DR pathogenesis has gained increasing recognition. Sirtuin 1 (SIRT1), a nicotinamide adenosine dinucleotide (NAD+)-dependent deacetylase, plays a crucial role in DR by preventing mitochondrial damage and apoptosis. Astaxanthin has protective effects against various diseases with its antioxidant and anti-inflammatory potency, but its interaction with SIRT1 in DR has not been explored. We hypothesized that astaxanthin alleviates

Cataracts in Atomic-bomb Survivors More than 70 Years after Radiation Exposure.

Previous ophthalmological studies among atomic-bomb survivors and other radiation-exposed populations have shown a significant association between radiation exposure and cataract, particularly posterior subcapsular cataract (PSC). However, reports of radiation effects on nuclear and cortical cataract are inconsistent. In the current study, we reexamined the issue of radiation effects on cataractogenesis using modern methods and well-defined diagnostic criteria. A questionnaire on ophthalmologic diseases was obtained from 2,439 atomic bomb survivors who were exposed at age 15 or younger and par
