GPX4 promotes optic nerve regeneration and retinal ganglion cell neuroprotection.

Glaucoma is an optic neuropathy characterized as progressive degeneration of retinal ganglion cells (RGCs) and their axons in the optic nerve (ON). Recently, lipid peroxidation and ferroptosis have been linked to experimental glaucoma, suggesting a new therapeutic strategy. Here, we report that glutathione peroxidase 4 (GPX4), a key regulator of lipid peroxidation, is upregulated in surviving and regenerating RGCs in two mouse optic neuropathy models, traumatic ON crush and glaucoma. To explore the potential neuroprotective role of GPX4, we test adeno-associated virus-mediated RGC-specific ove

A direct method for imaging gradient levels of retinal hypoxia in a model of retinopathy of prematurity (ROP).

BACKGROUND: Retinal hypoxia may contribute to the development of preretinal neovascularization (NV) in patients with retinopathy of prematurity (ROP). We hypothesized that levels of NV may be associated with levels of retinal hypoxia. Imaging retinal hypoxia could be an important diagnostic tool to predict levels of disease severity in ROP patients. In this study, we have investigated a direct method for imaging gradient levels of retinal hypoxia using a model of ROP. We believe that this discovery will help understand the ROP pathogenesis in premature infants. METHODS: The rat 50/10 oxygen-in

Beyond IOP: Neuroprotection and neuroregeneration as important therapeutic strategies for glaucoma.

Glaucoma is the leading cause of irreversible blindness characterized by the progressive death of retinal ganglion cells (RGCs) and damage to the optic nerve. Although the main goal of current treatment approaches is to reduce intraocular pressure (IOP), many patients' disease progression persists even after IOP returns to normal, highlighting the urgent need for effective neuroprotective strategies. The pathogenesis of glaucomatous neurodegeneration is thoroughly reviewed in this study, with special attention given to the roles of inflammation, excitotoxicity, mitochondrial dysfunction, oxida

Effects of a Red-Ginger-Based Multi-Nutrient Supplement on Optic Nerve Head Blood Flow in Open-Angle Glaucoma.

Objectives: Glaucoma is an age-related neurodegenerative disease, characterized by retinal ganglion cell loss and progressive visual field deterioration. Beyond intraocular pressure (IOP), vascular and metabolic dysregulation contributes to optic nerve head (ONH) ischemia and neuronal vulnerability. Nutritional factors with antioxidative and vasodilatory properties may help preserve ocular perfusion. This study investigated the acute and subacute effects of a single dose of a dietary supplement containing red ginger extract (Zingiber officinale var. rubra), lutein, and vitamin B6 on ONH blood

Development and pilot evaluation of a binocular virtual reality Headset-Based pupillometer for quantitative assessment of pupillary light reflexes.

Relative afferent pupillary defect (RAPD) is a key sign of optic neuropathies but is traditionally assessed subjectively. Virtual reality (VR) headsets with integrated binocular eye tracking enable simultaneous measurement under standardized illumination. We developed and conducted a pilot evaluation of a VR headset–based pupillometer that objectively quantifies RAPD using advanced pupil-tracking and automated blink-correction algorithms. This proof-of-concept study enrolled 17 patients with unilateral optic neuropathy (glaucomatous, traumatic, ischemic, or inflammatory) and 30 healthy control

PLGA nanoparticles restore acidic pH and degradative function to compromised lysosomes with Cy3-labeling providing enhanced tracking to lysosomes.

Lysosomal dysfunction and elevated lysosomal pH are hallmark features of age-related neurodegenerative diseases including age-related macular degeneration (AMD), Alzheimer's disease (AD), and Parkinson's disease (PD). Restoring lysosomal acidity is important for maintaining enzymatic degradation, preventing protein aggregation, and reducing cellular waste accumulation in degenerating tissues. Acidic nanoparticles represent a promising therapeutic strategy to normalize lysosomal pH; however, accurate monitoring of their delivery, retention, and dosage is critical for rigorous evaluation. ELP1 Gene Augmentation Restores Visual Function in a Mouse Model of Familial Dysautonomia.

Familial dysautonomia (FD) is an autosomal recessive sensory and autonomic neurodevelopmental and degenerative disorder characterized by complex neurological phenotypes. One of its most debilitating features is progressive optic neuropathy, which leads to severe visual impairment in FD patients by the third decade of life. Although several preclinical approaches have shown partial rescue of retinal ganglion cell (RGC) degeneration through increasing Elongator acetyltransferase complex subunit 1 (ELP1) expression in the retina, currently no treatments exist to prevent vision loss in FD. In this

Baicalin attenuates HIF-1α-driven histone lactylation and neutrophil extracellular trap (NET) formation in autoimmune uveitis.

Uveitis is a group of vision-threatening intraocular inflammatory disorders that can affect the uveal tract, retina, optic nerve, and vitreous, leading to irreversible structural damage and vision loss. Neutrophil extracellular trap (NET) formation is a key driver of acute uveitis (AU) pathology, but the immunometabolic mechanisms governing NETosis remain unclear. Here, we identified hypoxia-inducible factor-1α (HIF-1α) as a central regulator that links neutrophil metabolism to NET formation. DIA-based lactyl-proteomic profiling revealed widespread nuclear protein lactylation, with histone H3

Classifying retinal images via vascular-optic disc cross-segmentation and attentive feature selection.

The prevalence of retinal disorders is rising at an alarming rate, posing a significant risk of irreversible blindness without timely intervention. Recent advancements in artificial intelligence (AI) have enabled the development of automated screening systems for retinal image analysis. However, building robust models presents challenges due to inconsistencies in image acquisition, varying resolutions, and imbalanced class distributions. To address these issues, this research introduces a novel abnormality-aware attentive feature selection approach for classifying retinal images into three cat

Preoperative Neuroimaging Markers, Clinical Severity Measures, and Shunt Characteristics for Predicting Shunt Revision in Idiopathic Intracranial Hypertension: An Explainable Machine Learning Study.

BACKGROUND AND PURPOSE: Surgical shunt placement is a common treatment for idiopathic intracranial hypertension (IIH) but is hampered by high revision rates. Prior predictive models for shunt revision in IIH have overlooked disease-specific neuroimaging markers. We developed an explainable machine learning (ML) model to identify the strongest predictors of shunt revision across neuroimaging markers, clinical severity variables, and shunt-specific factors. The primary objective was to assess the contribution of IIH-related neuroimaging markers within this multimodal predictive framework. MATER

C1q dependency of retinal ganglion cell death in blast-mediated traumatic brain injury.

Traumatic brain injury is a widespread health concern that annually affects millions of people worldwide, with blast-mediated TBI (bTBI) being of particular relevance to military personnel and civilians in war zones. Current treatments primarily target symptoms, rather than the underlying neuropathology, and are ultimately limited by the need for an improved etiological understanding of TBI. Here, we utilized a pre-clinical mouse model of bTBI, previously shown to evoke a high susceptibility to retinal ganglion cell (RGC) death, to investigate whether neuroinflammatory pathways associated with

Dual-functional nanoparticle formulations for simultaneous intraocular pressure reduction and neuroprotection in glaucoma: a review.

Glaucoma is a leading cause of irreversible blindness, driven by elevated intraocular pressure (IOP), progressive retinal ganglion cell (RGC) loss, and optic nerve degeneration. Current therapies rely on lowering IOP, which slows but does not halt disease progression. Dual-functional nanoparticle (NP) formulations represent a promising approach to simultaneously address these therapeutic targets. By improving ocular drug penetration, sustaining release, and enabling co-delivery of diverse agents, nanocarriers can achieve prolonged IOP reduction while directly preserving RGC and optic nerve aga
