Ferroptosis in retinal neurodegeneration: mechanistic vulnerability, therapeutic targeting, and translational barriers.

Progressive retinal neurodegeneration remains a major therapeutic challenge in ophthalmology because structural and functional loss may continue despite control of the primary insult, including intraocular pressure reduction in glaucoma, metabolic management in diabetic retinopathy, or suppression of acute inflammatory activity. Ferroptosis, an iron-dependent form of regulated cell death driven by phospholipid peroxidation and insufficient antioxidant buffering, has emerged as a potential mechanism linking metabolic stress, ischemic injury, excitotoxicity, and neuroinflammatory signaling to su

Assessment of microexon-associated junction usage in Parkinson's disease and spinocerebellar Ataxia type 3.

BACKGROUND: Microexons are short exonic sequences of 3-30 nucleotides that undergo alternative splicing and have been implicated in several neurodevelopmental disorders. However, their contribution to neurodegenerative diseases remains insufficiently characterized. METHODS: We analyzed microexon-associated junction usage in the frontal cortex of Parkinson's disease (PD) patients and in the cerebellum of Spinocerebellar Ataxia Type 3 (SCA3) patients using deep RNA-Sequencing of human postmortem brain tissue and corresponding SH-SY5Y cellular models. Microexon-associated junction usage was quan

Quantification of the neuropathology of alcohol use disorder using tissue microarrays.

Alcohol use disorder (AUD) is characterized by an inability to stop consuming alcohol. Neuroimaging studies of patients with AUD show mild grey and white matter atrophy, while pathological studies suggest that atrophy is restricted to the white matter. The effects on individual brain cells are largely unknown. Mild neuronal loss has been described in the prefrontal cortex but this has not been consistent. Studies quantifying oligodendrocytes, astrocytes, and microglia are rare. These knowledge gaps impede therapeutic advancements. Here, we piloted the use of tissue microarrays, immunohistochem

Retinal nerve fiber layer and ganglion cell layer thinning as biomarkers of cognitive decline in multiple sclerosis: an 18-month prospective cohort study.

BACKGROUND: Retinal neurodegeneration measured by optical coherence tomography (OCT) has emerged as a candidate biomarker of central nervous system (CNS) damage in multiple sclerosis (MS). However, its relationship with cognitive dysfunction - a clinically relevant and underappreciated dimension of disease burden - remains poorly characterized. OBJECTIVE: To evaluate cross-sectional and longitudinal associations between retinal nerve fiber layer (RNFL) and ganglion cell layer (GCL) thickness and cognitive performance in MS patients without a history of optic neuritis. METHODS: A prospective

Reduced nerve growth factor (NGF) mediates arsenic-induced mitochondrial dynamics imbalance and neuronal damage both in vivo and in vitro.

Arsenic exposure is known to cause cognitive deficits, although the underlying mechanisms are yet to be explored. In this study, we investigated the role of nerve growth factor (NGF), a neuroprotective factor, in arsenic-induced cognitive impairment. In mouse models exposed to 25 and 50 mg/L sodium arsenite (NaAsO2), we observed neuronal damage accompanied by the downregulation of NGF, decreased phosphorylation of phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT), reduced phosphorylation of the mitochondrial fission protein dynamin-related protein 1 (Drp1), and downregulation of the mi

Neural tracking of surprisal and semantic distance in naturalistic movie viewing.

Understanding speech requires listeners to integrate incoming input with prior linguistic and thematic knowledge to access meaning, a task greatly aided by prediction. Surprisal and related phenomena (e.g., next word prediction) tend to be associated with broad activation of language regions during listening. A major challenge for interpreting the neural correlates of surprisal is that lexical expectancies are constrained both by syntactic and semantic information. In contrast, semantic distance yields more of a pure metric of relatedness between one language constituent (e.g., word, phrase, n

Therapeutic Applications of rTMS in Migraine Prophylaxis.

Migraine is one of the main causes of neurological disability, especially in young adults. Recent developments in preventive migraine therapies have improved outcomes for many patients, while highlighting the need for additional therapeutic options for those who remain refractory to pharmacological prophylaxis. Repetitive Transcranial Magnetic Stimulation (rTMS) has emerged as a promising neuromodulatory tool. We searched published literature for articles describing the efficacy of different rTMS protocols in the treatment of migraine. The literature search was conducted using PubMed and Scopu

Global Research Trends in Traditional Chinese Medicine for Diabetic Kidney Disease: A 20-Year Bibliometric Analysis.

BACKGROUND AND PURPOSE: The global burden of diabetic kidney disease (DKD) continues to escalate, establishing it as a predominant contributor to chronic kidney failure; nonetheless, effective therapeutic options remain limited. Emerging evidence indicates that traditional chinese medicine (TCM) may offer novel strategies for addressing this condition. However, the existing literature is characterized by considerable heterogeneity in both the specific TCM interventions employed (eg, single herbs, compound formulas, and phytochemical extracts) and the reported therapeutic outcomes across indivi

IL-17A Induces Chondrocyte Senescence via the STAT3-OPA1-mtROS Axis in Temporomandibular Joint Osteoarthritis.

OBJECTIVES: To elucidate the pathogenic mechanisms underlying interleukin-17A (IL-17A) induced chondrocyte senescence in TMJ-OA and provide preclinical evidence supporting IL-17A as a novel therapeutic target for TMJ-OA. METHODS: We established a TMJ-OA mouse model using unilateral anterior crossbite (UAC) to investigate IL-17A expression and chondrocyte senescence. We subsequently cultured ATDC5 mouse chondrocytes with IL-17A in vitro to assess cellular senescence and further explored mitochondrial-related mechanisms. Finally, we evaluated the inhibitory effect of Secukinumab (an anti-IL-17A

The Heterogeneity of the Neurovascular Unit in the Optic Nerve Head During Glaucomatous Neurodegeneration Based on Single-Cell RNA Sequencing.

Glaucoma is a multifactorial neurodegenerative disease, and the dysfunction of the neurovascular unit (NVU) in optic nerve head (ONH) has been gradually recognized to contribute to the neurodegeneration progression, yet its heterogeneity features remain poorly understood. Here, we established rat models of high intraocular pressure (HIOP) and lipopolysaccharide-induced ocular inflammation (LPS), and performed single-cell RNA sequencing (scRNA-seq) of ONH-centered tissues to characterize the cellular and molecular characteristics of NVU and its main cell components. Distinct alterations in the

Multilayer MEG source modelling enables depth-resolved laminar inference in humans.

Neural dynamics at the laminar level are critical for cortical computation. However, in humans, non-invasive methods to probe such dynamics have been limited to coarse distinctions between deep and superficial layers. Here, we present a multilayer magnetoencephalography source reconstruction framework and evaluate the conditions under which depth-resolved laminar inference may be feasible. Using simulations, we systematically assess the limits of magnetoencephalography depth resolution, showing that laminar discrimination depends on sufficiently high signal-to-noise ratio, precise co-registrat

Predictors of visual outcomes and recurrence after pars plana vitrectomy for uveitis: A 3-Year longitudinal cohort study.

Uveitis is a chronic relapsing inflammatory disease that often requires long-term systemic immunomodulatory (IMD) therapy. Pars plana vitrectomy (PPV) is increasingly used as an adjunctive treatment for selected uveitis cases; however, robust long-term evidence remains limited. This retrospective cohort study evaluated 3-year outcomes in 159 uveitic eyes undergoing PPV and a reference group of 22 eyes receiving medical therapy alone to assess visual, anatomical, and systemic treatment burden changes. Following PPV, best-corrected visual acuity (BCVA) improved significantly and remained stable
