Early postlesional visuomotor capacity after V1 damage in the macaque.

Damage to the primary visual cortex (V1) causes field defects in conscious vision, yet stimuli in the affected field may still guide saccades and other visuomotor responses ("blindsight"). Chronic blindsight is well documented, but early postlesional saccade profiles to scotoma stimuli remain poorly characterized. We mapped visuomotor functions in two macaques 3 to 4 weeks after a unilateral V1 lesion using a perimetry-like saccade assay up to 35° eccentricity, with analyses of saccade direction and catch-trial baselines. Targets had either unpredictable onset, as in clinical perimetry, or tem

The default mode network exhibits circuit-specific selective vulnerability despite widespread amyloid burden in an animal model of Alzheimer's disease.

In Alzheimer's Disease (AD) the default mode network (DMN) exhibits selective vulnerability to functional decline and amyloid beta (Aβ) plaques in early AD. DMN vulnerability may simply be the result of Aβ accumulation. Alternately, Aβ may unmask an intrinsic susceptibility of DMN circuits. Should DMN vulnerability persist amidst global Aβ deposition, this would support the latter hypothesis. We tested this using long-term, multi-region recordings of single units in mice characterized by widespread Aβ (APP/PS1). We tracked a neuronal population within the retrosplenial cortex (a DMN region) an

Psilocybin Prolongs the Neurovascular Coupling Response in Mouse Visual Cortex.

Psilocybin has profound therapeutic potential for various mental health disorders, but its mechanisms of action are unknown. Functional MRI studies have reported the effects of psilocybin on brain activity and connectivity; however, these measurements rely on neurovascular coupling to infer neural activity changes and assume that blood flow responses to neural activity are not altered by psilocybin. Using two-photon excited fluorescence imaging in the visual cortex of awake mice to simultaneously measure neural activity and capillary blood flow dynamics, we found that psilocybin administration

Structural MRI alterations in children with form-deprivation amblyopia: a propensity score-matched case-control study.

AIMS: Form-deprivation amblyopia (FDAM) is a severe neurodevelopmental disorder from early visual deprivation. This study uses T1-weighted MRI to explore its neuroanatomical correlates. METHODS: 18 Chinese children with bilateral FDAM and 36 propensity score-matched healthy Chinese controls (1-to-2 matched for age, sex and mean cortical thickness) underwent 3.0T T1-weighted MRI (1×1×1 mm³ resolution). Cortical and subcortical structures were parcellated using FreeSurfer V.7.3.2 with multiatlas (Desikan-Killiany-Tourville, Destrieux, subset Brodmann and Aseg), alongside specialised modules for

Progressive and Short-Interval Changes Observed in the Corticospinal Tract and Corpus Callosum of Patients with Amyotrophic Lateral Sclerosis: A Texture Analysis Study.

BACKGROUND AND PURPOSE: Diffusion MRI measures indicative of white matter integrity have consistently been shown to be altered in the state of the corticospinal tract (CST) and corpus callosum (CC) of patients with amyotrophic lateral sclerosis (ALS). However, diffusion MRI acquisitions are not routinely collected as part of the standard medical imaging of patients with ALS. T1-weighted MRI scans are commonly available in the clinical assessment of most patients with ALS. While visual inspection of these scans reveals little about the cerebral pathology of ALS, analysis of their textural patte

Sterile inflammation in LHON: mitochondrial stress, innate immune signaling, and implications for AAV gene therapy.

Leber hereditary optic neuropathy (LHON) is a mitochondrial disease caused primarily by pathogenic mitochondrial DNA (mtDNA) variants that impair respiratory chain complex I function. Although bioenergetic failure, oxidative stress, and retinal ganglion cell degeneration are central features of disease pathogenesis, these mechanisms do not fully explain the incomplete penetrance, male predominance, and marked clinical heterogeneity observed in LHON. Emerging evidence suggests that mitochondrial dysfunction can also trigger sterile inflammatory responses through the release of mitochondrial dam

Mapping human visual contrast sensitivity and vision loss across the visual field with model-based fMRI.

Peripheral vision is crucial for daily activities and quality of life, yet traditional measures of visual function like visual acuity primarily assess central vision. Visual field tests can evaluate peripheral vision but require extended focus and precise fixation, often challenging for patients with severe sight loss. Functional MRI (fMRI) with population receptive field (pRF) mapping offers a non-invasive way to map scotomas but relies on single contrast levels and accurate fixation. We developed an fMRI-based approach to measure contrast sensitivity across the visual field without requiring

Recurrent Left Abducens Nerve Palsy and Idiopathic Intracranial Hypertension During Pregnancy: A Diagnostic Challenge.

Idiopathic intracranial hypertension (IIH) is characterized by elevated intracranial pressure without an identifiable mass lesion, hydrocephalus, cerebral venous sinus thrombosis, or abnormal cerebrospinal fluid composition. Diagnosis during pregnancy can be challenging because headache and visual symptoms overlap with several neurologic and obstetric disorders. We report a previously healthy 20-year-old primigravida with a body mass index of approximately 33 kg/m² who presented at 17 weeks' gestation with intermittent headache, photophobia, blurred vision, horizontal diplopia, nasal deviation

Uremic Optic Neuropathy as the Initial Manifestation of Advanced Chronic Kidney Disease.

Uremic optic neuropathy (UON) is a rare but potentially reversible complication of advanced kidney failure characterized by optic disc edema and visual decline that improves with dialysis. We report the case of a 45-year-old man with no prior diagnosis of chronic kidney disease who presented with 3 months of progressive bilateral blurry vision, eye pain, and exertional fatigue. Ophthalmologic examination revealed bilateral optic disc edema with reduced visual acuity. Laboratory evaluation showed profound azotemia (blood urea nitrogen 182.9 mg/dL; creatinine 29.5 mg/dL), hyperkalemia, anemia, m

Immediate and Long-Term Effects of Different Montages of Transcranial Direct Current Stimulation Over Primary Visual Cortex in Patients With Chronic Migraine: A Randomized, Double-Blind, Sham-Controlled Trial.

BACKGROUND: We evaluated the immediate and long-term therapeutic efficacy of transcranial direct current stimulation (tDCS) using two distinct montages of anodal and cathodal electrical stimulation over the primary visual cortex compared with a sham tDCS in patients with chronic migraines. METHODS: In this randomized, double-blind, controlled trial, 72 eligible patients with resistant chronic migraine were randomly assigned to one of three groups, receiving anodal or cathodal (2 mA for 20 min), or sham tDCS (0.5 mA for 30 s), over the pain-dominant side of the primary visual cortex. Comparison of clinical features and cerebral glucose metabolism between patients with major depressive disorder and Parkinson's with depression.

BACKGROUND: Whether there are differences in clinical symptoms and brain glucose metabolism between major depressive disorder (MDD) and Parkinson’s comorbid depression (DPD) is unclear. The aims of this study were to investigate the differences in characteristics in clinical and glucose metabolism between MDD and DPD and their internal correlation. METHODS: We enrolled 30 MDD patients, 25 DPD patients, and 20 Parkinson’s patients without depression (PD-ND), all of whom were evaluated by neuropsychological scales and position emission tomography (PET) imaging. We compared the differences in dep

Glutamate Levels in Medial Prefrontal Cortex According to Illness Phase of Never-Medicated Individuals With Psychosis.

IMPORTANCE: Evidence suggests that elevated glutamate levels in the medial prefrontal cortex may be associated with the illness stage, but little is known about whether these effects occur in the absence of antipsychotic treatment. OBJECTIVE: To evaluate differences in glutamate levels in treatment-naive individuals with psychosis on the schizophrenia spectrum at different stages of the illness. DESIGN, SETTING, AND PARTICIPANTS: This cross-sectional study uses proton magnetic resonance spectroscopy in the medial prefrontal cortex of individuals experiencing their first episode of nonaffecti

The contribution and mode of integration of parvocellular and magnocellular information in the course of shape orientation perception.

Visual information reaches the cortex via multiple pathways, most notably the magnocellular (M) and parvocellular (P) streams. How their inputs are integrated remains unclear. We previously documented the case of Davida, a young woman who perceives high-contrast, sharply bounded 2D shapes as rotated or mirror-reversed, while he perception normalizes when stimuli are blurred, low in contrast, or brief. Here, we first review previous findings and propose that her disorder may reflect abnormally strong P-on-M inhibition. We then present three studies supporting this account: two demonstrate abnor

Suicide attempt history, childhood trauma, and functional brain network alterations in major depressive disorder: a resting-state functional connectivity-based multivariate pattern analysis.

A history of suicide attempts is one of the strongest predictors of future suicidal behavior in major depressive disorder (MDD). However, the resting-state functional connectivity (rs-FC) alterations that specifically differentiate MDD patients with a suicidal attempt history (SD) from those without (NSD) remain poorly understood. This study aimed to identify suicide attempt-specific rs-FC alterations using a two-stage multivariate pattern analysis (MVPA) framework-first contrasting the MDD group with the healthy control (HC) group, then directly comparing the SD group with the NSD group-and t

Uniformity in happiness and uniqueness in sadness: Naturalistic emotional representation in major depression.

Humans develop shared concepts of others' emotions to support adaptive social functioning, yet how these concepts are dynamically represented in major depressive disorder (MDD) during naturalistic movie viewing is not yet fully established. Using functional MRI, we examined patients with MDD (n = 55) and healthy controls (HCs; n = 62) as they freely viewed movie clips depicting happy and sad emotions. Neural similarity was quantified with inter-subject correlation at whole-brain, network, and regional levels, and its association with emotional traits was assessed using inter-subject representa

Mapping the cortical architecture of sleep deprivation: insights from fMRI, neurotransmission, and metabolic activity.

BACKGROUND: Sleep deprivation (SD) produces profound cognitive deficits, yet its integrative effects on large-scale cortical organization, metabolic alterations, and cognitive function remain unclear. METHODS: Based on meta-analytic seeds, lesion network mapping was applied to connectome data from 1000 healthy individuals to generate an SD-related cortical weight map (SD-CWM). Thirty participants then completed a 24-hour acute SD protocol, during which resting-state and memory-task-evoked brain activities, untargeted metabolomics, and memory performance were measured under both rested wakeful

Longitudinal changes in neurometabolites in pediatric migraine across child development: a pilot study.

Migraine severely impacts quality of life in children and adolescents and has been identified as the most important cause of neurological disability in youth. There is evidence that an imbalance between brain excitation (glutamate) and inhibition (GABA) is one of the driving factors behind migraine attacks. During adolescence, changes to glutamatergic and GABAergic neurons occur. It is therefore perhaps unsurprising that adolescence is also a transitory time for migraine, with changes in incidence and associated symptoms. However, there is a lack of research into interactions between brain che

GABAergic interneuron subtypes differentially regulate noise correlations that are critical for visual orientation discrimination in mice.

Noise correlations-synchronized firing fluctuations among neurons-critically influence sensory processing, yet their regulatory mechanisms remain unclear. We investigated how V1 inhibitory neuron subtypes regulate noise correlations and visual encoding in mice using optogenetics and electrophysiology. Suppression of parvalbumin-positive (PV+) or somatostatin-positive (SOM+) neurons increased noise correlations, whereas suppression of vasoactive intestinal peptide-positive (VIP+) neurons decreased them, reflecting their distinct inhibitory roles. These changes correlated with altered orientatio
