Retinitis Pigmentosa and erythrocytic microcytosis related to hypomorphic variants in the TRNT1 gene.

INTRODUCTION: The tRNA Nucleotidyl Transferase 1 (TRNT1) gene encodes an enzyme that adds a trinucleotide sequence (CCA) to the 3'-end of transfer RNA. Biallelic pathogenic TRNT1 variants are associated with a multisystem disorder featuring sideroblastic anaemia, B-cell immunodeficiency, periodic fevers and developmental delay. More recently, hypomorphic TRNT1 variants have been linked to a milder phenotype involving retinitis pigmentosa (RP) and erythrocytic microcytosis. METHODS: We report a novel case of TRNT1-related RP associated with erythrocytic microcytosis. Multimodal retinal imaging

Rodent Models of Glaucoma: Pathogenic Mechanisms, Phenotyping Strategies, and Translational Relevance.

Glaucoma comprises a heterogeneous group of optic neuropathies characterized by progressive degeneration of retinal ganglion cells (RGCs) and their axons. Rodent models have been widely used to investigate disease mechanisms, identify neuroprotective targets and evaluate emerging therapeutic strategies because they are cost-effective, genetically tractable and experimentally controllable. However, mice and rats differ substantially from humans in anterior-segment anatomy, trabecular meshwork architecture, Schlemm's canal development, lamina cribrosa structure, retinal ganglion cell composition

Recurrent Left Abducens Nerve Palsy and Idiopathic Intracranial Hypertension During Pregnancy: A Diagnostic Challenge.

Idiopathic intracranial hypertension (IIH) is characterized by elevated intracranial pressure without an identifiable mass lesion, hydrocephalus, cerebral venous sinus thrombosis, or abnormal cerebrospinal fluid composition. Diagnosis during pregnancy can be challenging because headache and visual symptoms overlap with several neurologic and obstetric disorders. We report a previously healthy 20-year-old primigravida with a body mass index of approximately 33 kg/m² who presented at 17 weeks' gestation with intermittent headache, photophobia, blurred vision, horizontal diplopia, nasal deviation

Restorative and regenerative therapies for vision loss.

Reversing blindness and restoring functional vision remains a central challenge in biomedical engineering. Advances in ocular prosthetics have enabled limited recovery of light perception, motion detection, and coarse object recognition through anterior segment devices such as keratoprostheses and through posterior segment interfaces including retinal, suprachoroidal, optic nerve, and cortical implants. Parallel progress in organ-on-a-chip systems and retinal organoids is providing increasingly sophisticated platforms for disease modelling and therapeutic development. Despite these advances, i

Presentation and management of tislelizumab induced toxic optic neuropathy.

Immune checkpoint inhibitors (ICIs) have revolutionised the management of advanced malignancies, but can cause immune-related adverse events (irAEs), including vision-threatening complications such as optic neuropathy. Ocular irAEs have been described with several programmed death-1/programmed death-ligand 1 and cytotoxic T lymphocyte-associated antigen-4 inhibitors, but ocular toxicities of tislelizumab have so far been limited to Vogt-Koyanagi-Harada (VKH)-like uveitis, panuveitis and retinal vasculitis. We describe a woman with metastatic lung adenocarcinoma who was treated with second-line

Case Report: Intracranial hypertension occurring in association with subacute thyroiditis.

Subacute thyroiditis (SAT) is a self-limited inflammatory thyroid disorder that typically produces a transient thyrotoxic phase. Intracranial hypertension (IH) is a rare but potentially sight-threatening complication of thyrotoxicosis, reported almost exclusively in hyperthyroidism, with only a single previous report linking it to SAT. We report the case of a 29-year-old woman who developed IH during the thyrotoxic phase of subacute thyroiditis, temporally related to dental manipulation. She presented with headache, nausea, vomiting, and fever, followed by a syncopal episode; clinical, laborat

Ferroptosis in retinal neurodegeneration: mechanistic vulnerability, therapeutic targeting, and translational barriers.

Progressive retinal neurodegeneration remains a major therapeutic challenge in ophthalmology because structural and functional loss may continue despite control of the primary insult, including intraocular pressure reduction in glaucoma, metabolic management in diabetic retinopathy, or suppression of acute inflammatory activity. Ferroptosis, an iron-dependent form of regulated cell death driven by phospholipid peroxidation and insufficient antioxidant buffering, has emerged as a potential mechanism linking metabolic stress, ischemic injury, excitotoxicity, and neuroinflammatory signaling to su

Retinal nerve fiber layer and ganglion cell layer thinning as biomarkers of cognitive decline in multiple sclerosis: an 18-month prospective cohort study.

BACKGROUND: Retinal neurodegeneration measured by optical coherence tomography (OCT) has emerged as a candidate biomarker of central nervous system (CNS) damage in multiple sclerosis (MS). However, its relationship with cognitive dysfunction - a clinically relevant and underappreciated dimension of disease burden - remains poorly characterized. OBJECTIVE: To evaluate cross-sectional and longitudinal associations between retinal nerve fiber layer (RNFL) and ganglion cell layer (GCL) thickness and cognitive performance in MS patients without a history of optic neuritis. METHODS: A prospective

Reduced nerve growth factor (NGF) mediates arsenic-induced mitochondrial dynamics imbalance and neuronal damage both in vivo and in vitro.

Arsenic exposure is known to cause cognitive deficits, although the underlying mechanisms are yet to be explored. In this study, we investigated the role of nerve growth factor (NGF), a neuroprotective factor, in arsenic-induced cognitive impairment. In mouse models exposed to 25 and 50 mg/L sodium arsenite (NaAsO2), we observed neuronal damage accompanied by the downregulation of NGF, decreased phosphorylation of phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT), reduced phosphorylation of the mitochondrial fission protein dynamin-related protein 1 (Drp1), and downregulation of the mi

The Heterogeneity of the Neurovascular Unit in the Optic Nerve Head During Glaucomatous Neurodegeneration Based on Single-Cell RNA Sequencing.

Glaucoma is a multifactorial neurodegenerative disease, and the dysfunction of the neurovascular unit (NVU) in optic nerve head (ONH) has been gradually recognized to contribute to the neurodegeneration progression, yet its heterogeneity features remain poorly understood. Here, we established rat models of high intraocular pressure (HIOP) and lipopolysaccharide-induced ocular inflammation (LPS), and performed single-cell RNA sequencing (scRNA-seq) of ONH-centered tissues to characterize the cellular and molecular characteristics of NVU and its main cell components. Distinct alterations in the

MRI Characteristics of Myelin Oligodendrocyte Glycoprotein Antibody-Associated Disease: A Review.

IMPORTANCE: Myelin oligodendrocyte glycoprotein antibody-associated disease (MOGAD) is a demyelinating disease distinct from multiple sclerosis (MS) and aquaporin-4 antibody-positive neuromyelitis optica spectrum disorder (AQP4+NMOSD). Magnetic resonance imaging (MRI) is central to confirming attacks, defining lesion distribution, and excluding alternative diagnoses and is a key component of the 2023 MOGAD diagnostic criteria. However, its imaging spectrum and lesion evolution remain underrecognized in practice, and variability in imaging parameters can contribute to delayed diagnosis and subo

Expression of miR-802 in glaucoma and its diagnostic significance.

INTRODUCTION AND OBJECTIVE: Glaucoma ranks as the second leading cause of blindness globally. The key factors leading to glaucoma are increased intraocular pressure and retinal ganglion cell apoptosis. Glaucoma is an irreversible disease that causes blindness in the eye, characterized by progressive optic nerve damage and visual field defects. The expression of miR-802 is abnormal in the plasma of glaucoma patients, but its function and mechanism remain unclear. The aim of the study is to investigate the expression of miR-802 in glaucoma and retinal ganglion cells, and the effect of miR-802 on
