An NR2F1-dependent retinoic acid network controls retinal specialization in mice and reveals foveal hypoplasia in patients with BBSOAS.

The molecular programs that establish specialized retinal regions during development are essential for high-acuity vision, yet how their disruption contributes to human visual disorders remains poorly understood. Bosch-Boonstra-Schaaf optic atrophy syndrome (BBSOAS), caused by pathogenic variants in NR2F1 and characterized by visual impairment, provides an opportunity to investigate these mechanisms. Using single-cell RNA sequencing of three complementary Nr2f1 mouse models, including two carrying patient-specific mutations, we identified a shared Nr2f1 -dependent transcriptional program enric

OPA1 controls mitochondrial dysfunction-driven liver fibrosis in MASLD.

Progressive hepatic fibrosis is the principal determinant of morbidity and mortality in metabolic dysfunction-associated steatotic liver disease and steatohepatitis (MASLD/MASH). Mitochondrial dysfunction is a hallmark of MASH, and the release of mitochondrial damage-associated molecular patterns (mito-DAMPs) from injured hepatocytes can promote fibrosis. However, how mitochondrial dynamics and quality control shape the fibrotic response in MASLD/MASH remains unclear. Here, through large-scale genomic analyses of mitochondrial genes governing mitophagy, fusion and fission in human MASLD, with

Repurposing the gastroprotective agent geranylgeranylacetone for atherosclerosis: a novel role of the HSP22-Nrf2-mitochondrial axis.

Mitochondrial dysfunction is a pivotal driver of vascular endothelial injury and atherogenesis. This study aimed to evaluate the therapeutic potential of geranylgeranylacetone (GGA), a clinically established gastroprotective agent, in atherosclerosis and to explore the underlying molecular mechanisms. Apolipoprotein E-deficient (ApoE-/-) mice with genetic heat shock protein 22 (HSP22) overexpression or deficiency were used to investigate the vascular effects of HSP22 in vivo. In vitro, human umbilical vein endothelial cells (HUVECs) were challenged with oxidized low-density lipoprotein (Ox-LDL

Mitochondrial rewiring supports survival of triple negative breast cancer cells after ionizing radiation.

Triple negative breast cancer (TNBC) is an aggressive disease with limited therapeutic options. Conventional treatments include neoadjuvant chemo-immunotherapy followed by surgical resection and may include further adjuvant immunotherapy and/or radiotherapy of the tumor bed and lymph nodes. Nonetheless, TNBC patients with residual disease have rapid metastatic recurrence. While the roles of metabolic and mitochondrial adaptations in chemotherapeutic resistance have been the subject of many studies, their importance in the context of ionizing radiation (IR) therapy remains poorly understood. Chlorogenic Acid Derivative B11 Inhibits Glioma Proliferation by Targeting MST4 to Regulate Oxidative Stress.

This study focuses on the mechanism of action of chlorogenic acid derivative B11 on glioma. Although chlorogenic acid exhibits certain anti-tumor activity, it has limitations such as low bioavailability, broad action targets, and insufficient specificity, which restrict its practical application in tumor treatment. Therefore, chlorogenic acid derivative B11 was obtained through structural modification for investigation. Using cell experiments, animal experiments, and molecular techniques, it was found that in cell experiments, the CCK-8 (Cell Counting Kit-8) assay confirmed that B11 has a stro

A novel NDUFS1 missense variant associated with optic atrophy and mitochondrial dysfunction.

This study identified two compound heterozygous variants in the NDUFS1 gene in a patient presenting with optic atrophy including the known variant c.64C>T p.(R22X) and a novel variant c.404G>A p.(G135D). The Ndufs1G135D/- mouse model exhibited reduced Ndufs1 expression, impaired optic nerve function and thinning of the ganglion cell complex/retinal thickness ratio. Structural and interaction analyses indicated the G135D variant located within the Fe-S cluster domain of NDUFS1, disrupted its interaction with NDUFV1, leading to protein instability and aberrant mitochondrial localization. Sterile inflammation in LHON: mitochondrial stress, innate immune signaling, and implications for AAV gene therapy.

Leber hereditary optic neuropathy (LHON) is a mitochondrial disease caused primarily by pathogenic mitochondrial DNA (mtDNA) variants that impair respiratory chain complex I function. Although bioenergetic failure, oxidative stress, and retinal ganglion cell degeneration are central features of disease pathogenesis, these mechanisms do not fully explain the incomplete penetrance, male predominance, and marked clinical heterogeneity observed in LHON. Emerging evidence suggests that mitochondrial dysfunction can also trigger sterile inflammatory responses through the release of mitochondrial dam

Reduced nerve growth factor (NGF) mediates arsenic-induced mitochondrial dynamics imbalance and neuronal damage both in vivo and in vitro.

Arsenic exposure is known to cause cognitive deficits, although the underlying mechanisms are yet to be explored. In this study, we investigated the role of nerve growth factor (NGF), a neuroprotective factor, in arsenic-induced cognitive impairment. In mouse models exposed to 25 and 50 mg/L sodium arsenite (NaAsO2), we observed neuronal damage accompanied by the downregulation of NGF, decreased phosphorylation of phosphatidylinositol 3-kinase/protein kinase B (PI3K/AKT), reduced phosphorylation of the mitochondrial fission protein dynamin-related protein 1 (Drp1), and downregulation of the mi

Optical Coherence Tomography-Based Differentiation of Nonarteritic Anterior Ischemic Optic Neuropathy and Central Retinal Artery Occlusion: Longitudinal Analysis of Inner Retinal Layer Thinning.

BACKGROUND: Nonarteritic anterior ischemic optic neuropathy (NAION) and central retinal artery occlusion (CRAO) are both causes of acute vision loss. When patients present within the first several weeks of symptom onset, clinical signs such as optic disc or inner retinal edema often allow differentiation. However, as time progresses and edema resolves, both diseases lead to optic atrophy and retinal thinning, sometimes making them difficult to distinguish. Clinical experience suggests that CRAO results in more significant thinning of the inner retinal layers, particularly the ganglion cell lay

[The "Triple-hit" pathogenic mechanism of Leber hereditary optic neuropathy].

Leber hereditary optic neuropathy (LHON) is a classic mitochondrial disorder primarily affecting retinal ganglion cells and leading to irreversible visual loss. Although primary mitochondrial DNA (mtDNA) mutations, such as m.11778G>A, m.14484T>C, and m.3460G>A, provide the genetic basis of LHON, they do not fully explain its incomplete penetrance, male predominance, or clinical heterogeneity. Based on familial genetic analyses and functional studies, this review proposes a "triple hit" pathogenic mechanism for LHON. The first hit consists of primary mtDNA mutations, mainly affecting respirator

Unveiling the diversity, ecology, and biotechnological potential of culturable marine yeasts in Western Mediterranean coastal ecosystems.

Coastal ecosystems are dynamic environments characterized by strong seasonal variability in physicochemical parameters and biological communities. In the western Mediterranean, the Region of Murcia (southeastern Spain) is characterized by notable biodiversity and ecological heterogeneity, encompassing diverse habitats, including rocky shores, sandy beaches, seagrass meadows, and the Mar Menor, one of Europe's largest hypersaline lagoons. Within these systems, marine yeasts play critical ecological roles in organic matter recycling and produce metabolites of biotechnological and medical interes

Myelin oligodendrocyte glycoprotein-antibody disease (MOGAD) with leukodystrophy-like presentation.

Myelin oligodendrocyte glycoprotein-antibody disease (MOGAD) is an immune-mediated demyelinating disorder, distinct from MS, which typically affects the optic nerve, spinal cord, brain and/or brainstem. Diffuse white matter involvement that resembles a leukodystrophy has been reported only in children. We present a woman aged 29 years with new right optic neuritis on a background of previously unexplained, longstanding, bilateral, reduced visual acuity. MR scan of the brain and orbits showed confluent, bilateral, posterior predominant white matter changes reminiscent of a leukodystrophy, along

Diabetes and optic atrophy in a young adult: consider Wolfram syndrome.

A 22-year-old woman had an 8-year history of progressive bilateral vision loss and of diabetes mellitus. Her mother had diabetes and two first cousins had severe congenital deafness. On examination, her visual acuities were 6/36 bilaterally, with absent colour vision and gross optic disc pallor. Her HbA1c was 94 mmol/mol (20-42). Visual evoked potentials were bilaterally symmetrically prolonged with P100 140 ms. Optical coherence tomography showed significant thinning of the peripapillary retinal nerve fibre layer. MR scan of brain and orbits showed small optic nerves and chiasm. Genetic testi

Recessive variants in mitochondrial complex I nuclear subunits are an underrated cause of optic atrophy.

Our understanding of the genetic landscape of inherited optic neuropathies (ION) has grown significantly over the past decades, and it is now known to involve many genes found in both the nuclear and mitochondrial genomes, exhibiting all possible inheritance patterns. Furthermore, pathogenic variants in nuclear genes encoding mitochondrial respiratory complex I (CI) subunits have been identified in some cases of ION, in addition to the more common severe presentation of CI deficiencies, which usually have an early onset. We conducted next-generation sequencing screening of CI genes to identify

"Inverse" Foster Kennedy Syndrome: A Case Report.

INTRODUCTION: Foster Kennedy syndrome classically occurs in frontal space-occupying lesions (SOL). It describes the appearance of optic atrophy in one eye and optic nerve swelling in the other. Optic atrophy occurs due to direct compression from lesion (on the ipsilateral side) and optic nerve head swelling occurs due to increased intracranial tension due to mass effect (on the contralateral side). CASE REPORT: We report a case of a 37-year-old female presenting with a right SOL, yet left optic nerve atrophy and right optic nerve head swelling; inverse Foster Kennedy syndrome. CONCLUSION: In
