Unveiling the diversity, ecology, and biotechnological potential of culturable marine yeasts in Western Mediterranean coastal ecosystems.

Coastal ecosystems are dynamic environments characterized by strong seasonal variability in physicochemical parameters and biological communities. In the western Mediterranean, the Region of Murcia (southeastern Spain) is characterized by notable biodiversity and ecological heterogeneity, encompassing diverse habitats, including rocky shores, sandy beaches, seagrass meadows, and the Mar Menor, one of Europe's largest hypersaline lagoons. Within these systems, marine yeasts play critical ecological roles in organic matter recycling and produce metabolites of biotechnological and medical interes

Myelin oligodendrocyte glycoprotein-antibody disease (MOGAD) with leukodystrophy-like presentation.

Myelin oligodendrocyte glycoprotein-antibody disease (MOGAD) is an immune-mediated demyelinating disorder, distinct from MS, which typically affects the optic nerve, spinal cord, brain and/or brainstem. Diffuse white matter involvement that resembles a leukodystrophy has been reported only in children. We present a woman aged 29 years with new right optic neuritis on a background of previously unexplained, longstanding, bilateral, reduced visual acuity. MR scan of the brain and orbits showed confluent, bilateral, posterior predominant white matter changes reminiscent of a leukodystrophy, along

Diabetes and optic atrophy in a young adult: consider Wolfram syndrome.

A 22-year-old woman had an 8-year history of progressive bilateral vision loss and of diabetes mellitus. Her mother had diabetes and two first cousins had severe congenital deafness. On examination, her visual acuities were 6/36 bilaterally, with absent colour vision and gross optic disc pallor. Her HbA1c was 94 mmol/mol (20-42). Visual evoked potentials were bilaterally symmetrically prolonged with P100 140 ms. Optical coherence tomography showed significant thinning of the peripapillary retinal nerve fibre layer. MR scan of brain and orbits showed small optic nerves and chiasm. Genetic testi

Recessive variants in mitochondrial complex I nuclear subunits are an underrated cause of optic atrophy.

Our understanding of the genetic landscape of inherited optic neuropathies (ION) has grown significantly over the past decades, and it is now known to involve many genes found in both the nuclear and mitochondrial genomes, exhibiting all possible inheritance patterns. Furthermore, pathogenic variants in nuclear genes encoding mitochondrial respiratory complex I (CI) subunits have been identified in some cases of ION, in addition to the more common severe presentation of CI deficiencies, which usually have an early onset. We conducted next-generation sequencing screening of CI genes to identify

"Inverse" Foster Kennedy Syndrome: A Case Report.

INTRODUCTION: Foster Kennedy syndrome classically occurs in frontal space-occupying lesions (SOL). It describes the appearance of optic atrophy in one eye and optic nerve swelling in the other. Optic atrophy occurs due to direct compression from lesion (on the ipsilateral side) and optic nerve head swelling occurs due to increased intracranial tension due to mass effect (on the contralateral side). CASE REPORT: We report a case of a 37-year-old female presenting with a right SOL, yet left optic nerve atrophy and right optic nerve head swelling; inverse Foster Kennedy syndrome. CONCLUSION: In

Beta Zone and Gamma Zone Parapapillary Atrophy in Nonmyopic Eyes.

PRCIS: Not only β-zone but also ɣ-zone atrophy is observed in nonmyopic eyes. The presence of ɣ-zone atrophy reflects optic nerve head canal offset during eyeball expansion, and its directionality is associated with the offset direction. PURPOSE: To evaluate the locations of β-zone and ɣ-zone parapapillary atrophy (PPA) in nonmyopic eyes. METHODS: A total of 151 nonmyopic eyes with an axial length (AXL) of <24.0 mm were included in this cross-sectional study. From the Bruch membrane opening (BMO), the angular locations of the maximal width of β-zone PPA (without retinal pigment epitheliu

Mitochondrial acetyl-CoA reprogramming by the SIRT3-ACSS2-OPA1 axis confers resistance to ferroptosis in Parkinson's disease.

Mitochondrial dysfunction and ferroptosis have emerged as pivotal contributors to dopaminergic (DA) neuron degeneration in Parkinson's disease (PD). Here, a previously unrecognized SIRT3-ACSS2-OPA1 axis that couples mitochondrial acetyl-CoA (Ac-CoA) metabolism to ferroptosis resistance is identified. Analysis of public human substantia nigra datasets reveals marked reduction in SIRT3 expression, which is further confirmed in 6-OHDA-induced PD models. To establish translational significance, analyses of serum and peripheral blood mononuclear cells (PBMCs) from PD patient cohort demonstrates dec

Combined effects of high-intensity interval training in the form of swimming exercise and rapamycin-sensitive mTORC1 inhibition on mitochondrial adaptations in mouse skeletal muscles.

High-intensity interval training (HIIT) is an effective strategy for enhancing exercise performance by stimulating skeletal muscle mitochondria and increasing their oxidative metabolic capacity. HIIT is also known to stimulate the mechanistic target of rapamycin complex 1 (mTORC1), a key regulatory factor in muscle protein anabolism; however, the role of mTORC1 in HIIT-induced mitochondrial adaptations remains unclear. We hypothesized that mTORC1 modulates mitochondrial adaptation induced by exercise training and investigated the effects of in vivo administration of rapamycin (RAPA), a canonic

Far-infrared irradiation restores mitochondrial dynamics to ameliorate ischemic stroke.

Far-infrared irradiation exhibits promise in chronic diseases, and its role in ischemic stroke specifically in modulating mitochondrial dynamics remains unknown. This study explored the neuroprotective effects of far-infrared irradiation using a rat middle cerebral artery occlusion model and oxygen-glucose deprivation-injured neuronal cells. In middle cerebral artery occlusion rats, daily 30-minute far-infrared irradiation treatment reduced infarct volume, alleviated cerebral edema, and improved neurological function. Proteomic analysis identified far-infrared irradiation-mediated upregulation

Lack of genetic evidence for a role of SLC25A46 in alpha-synucleinopathies.

BACKGROUND: The SLC25A46 gene encodes a mitochondrial carrier protein previously implicated in neuropathy and optic atrophy. Biallelic variants in SLC25A46 have been described in patients with Parkinson's disease (PD) with optic atrophy, but the evidence supporting a role in PD remains limited. OBJECTIVE: To assess whether SLC25A46 variants contribute to PD, REM sleep behavior disorder (RBD), or Dementia with Lewy Bodies (DLB). METHODS: We examined common variants using four representative PD genome-wide association studies (GWAS) and an RBD GWAS and applied Summary-data-based Mendelian Rand

Bridging tradition and modernity: mitochondrial dynamics as a Traditional Chinese Medicine therapeutic target in cardiovascular disease.

BACKGROUND: A pathological link exists between mitochondrial fission/fusion imbalance and cardiovascular disease (CVD). Traditional Chinese Medicine (TCM), based on concepts such as "Qi stagnation and blood stasis" and "Yin-Yang imbalance," helps balance mitochondrial function through the combined effects of multiple components, providing a comprehensive treatment approach for CVD. AIMS OF THE STUDY: To methodically clarify the molecular processes by which TCM formulations, extracts, and bioactive compounds target mitochondrial dynamics to intervene in CVD over the past five years, highlighti

Optic neuropathy arising from the synergy between YARS2 and mitochondrial COX1 mutations.

Leber hereditary optic neuropathy (LHON) is a paradigm for mitochondrial retinopathy. Here, we investigate the mechanism underlying the interaction between nuclear modifier and mtDNA mutation(s) that manifests optic neuropathy in vivo to develop an effective therapeutic approach for this disease, using mouse models bearing LHON-linked Yars2G186V or COIV421A mutation alone and double mutations. Yars2G186V alters mitochondrial translation and assembly and activities of complex I, III, and IV, while COIV421A reduces complex IV activity. However, a single Yars2G186V or COIV421A mutation causes mil
