Biased 5-HT1A agonist NLX-101 improves executive function under NMDA receptor hypofunction and preferentially enhances cortical ERK2 phosphorylation.

Cognitive inflexibility and working memory deficits are major contributors to functional impairment in neuropsychiatric disorders and remain insufficiently addressed by current treatments. NLX-101, a preferential postsynaptic 5-HT1A receptor biased agonist, engages cortical serotonergic signaling and ERK1/2-related pathways, offering a mechanistically distinct approach to improving prefrontal cortex-dependent cognition. Here, we examined whether NLX-101 counteracts executive function deficits induced by NMDA receptor blockade in BALB/c mice. Working memory was assessed using the 8-arm radial m

Genetic susceptibility and neuroprotective gene therapy of normal tension glaucoma: a review.

Normal-tension glaucoma (NTG) is a distinct form of glaucomatous optic neuropathy characterized by progressive retinal ganglion cell (RGC) degeneration and visual field loss despite intraocular pressure (IOP) remaining within the statistically normal range. Although IOP reduction remains the only established disease-modifying strategy, continued disease progression despite adequate IOP control highlights the importance of IOP-independent mechanisms. Increasing evidence implicates vascular dysregulation, mitochondrial dysfunction, oxidative stress, impaired autophagy, neuroinflammation, axonal

Global Research Trends in Traditional Chinese Medicine for Diabetic Kidney Disease: A 20-Year Bibliometric Analysis.

BACKGROUND AND PURPOSE: The global burden of diabetic kidney disease (DKD) continues to escalate, establishing it as a predominant contributor to chronic kidney failure; nonetheless, effective therapeutic options remain limited. Emerging evidence indicates that traditional chinese medicine (TCM) may offer novel strategies for addressing this condition. However, the existing literature is characterized by considerable heterogeneity in both the specific TCM interventions employed (eg, single herbs, compound formulas, and phytochemical extracts) and the reported therapeutic outcomes across indivi

Multimodal approaches to visual experimental research: Imaging, electrophysiology, and behavior.

Although vision is one of the primary senses in humans, direct mechanistic research in patients is challenging both technically and ethically because of its multiscale complexity-spanning phototransduction, neural processing, and perception. The foundation of visual science is animal models, which offer controlled, systematic assess to every stage of the visual pathway. Animal models' visual phenotyping methods are highly significant for vision-related research. However, the selection of various experimental methods and critical differences between them are frequently disregarded in study desi

C1q dependency of retinal ganglion cell death in blast-mediated traumatic brain injury.

Traumatic brain injury is a widespread health concern that annually affects millions of people worldwide, with blast-mediated TBI (bTBI) being of particular relevance to military personnel and civilians in war zones. Current treatments primarily target symptoms, rather than the underlying neuropathology, and are ultimately limited by the need for an improved etiological understanding of TBI. Here, we utilized a pre-clinical mouse model of bTBI, previously shown to evoke a high susceptibility to retinal ganglion cell (RGC) death, to investigate whether neuroinflammatory pathways associated with

Sociodemographic and Clinical Predictors of Colonoscopy Utilization in Oman: A Call to Action within a Population-Based Cross-Sectional Study.

OBJECTIVE: Colorectal cancer (CRC) is the second leading cause of cancer mortality in Oman, yet structured screening remains absent. Most cases are diagnosed at advanced stages, highlighting the need to understand predictors of colonoscopy uptake to inform national screening strategies. This study aimed to identify sociodemographic, perceptual, and healthcare-related factors associated with colonoscopy utilization-whether for screening or diagnostic purposes among Omani adults. METHODS: A nationwide, cross-sectional online survey was conducted using a structured questionnaire adapted from the

Subtyping OCD based on individual symptom networks: subtype-specific neural dynamics and morphometric similarity linked to molecular profiles.

Obsessive-compulsive disorder (OCD) is clinically heterogeneous, posing challenges for diagnosis and treatment. Individual symptom network-based subtyping could provide the framework for characterizing this heterogeneity. This study developed an individualized symptom network (ISN)-based subtyping framework, leveraging a healthy control dataset (n = 3227) to model the normative symptom network. Based on the prominence of obsessive-compulsive (OC) symptoms within ISN (Expected Influence within ISN, EIISN), three OCD subtypes were identified in the discovery cohort (n = 238). Subtype 1 (39.50%)

Beyond Single Variants: A Pathway-Based Approach to Explore the Genetic Basis of Memory.

Memory plays a crucial role in human cognitive processes and daily functioning. While memory has a genetic basis, identifying the specific genetic factors influencing memory performance has proven challenging. This challenge arises because memory is a complex trait, whose genetic architecture likely comprises an accumulation of many low-effect size common variants. Thus, study sample sizes are easily insufficient, leading to underpowered genetic association analyses. This limitation is especially pronounced in studies that prioritize deep phenotyping over broader recruitment, resulting in smal

Primary and nonprimary auditory cortex connectivity with brain regions involved in cognitive and emotional processing: in mouse and human.

Auditory cortex connectivity extends beyond the processing of acoustic stimuli, playing a crucial role in cognitive and emotional regulation through its interactions with higher-order brain regions. Although the neural mechanisms underlying acoustic information processing along the auditory pathway are well-documented, the connections supporting auditory-related cognitive and emotional processing, particularly in comparative studies between mice and human adults, are not yet fully clarified. In this study, we aim to investigate connections between the auditory cortex and brain regions involved

Functional dysconnectivity in breast cancer patients undergoing hormone therapy.

INTRODUCTION: Breast cancer patients undergoing adjuvant hormone therapy commonly report adverse effects that can lead to lower quality of life and treatment nonadherence. How hormone therapy, independent of other systemic therapies, may impact patient functioning is a relatively new area of research with few neuroimaging studies delineating the effects. Prior nonspecific neuropsychological findings and the multifaceted role of estrogen in the brain suggest potentially diffuse effects of hormone therapy. The current study examined intrinsic neural functional organization and cognitive correlat

Differential GABA dynamics across brain functional networks in autism.

Differences in the ϒ-aminobutyric acid (GABA) system contribute to an excitatory-inhibitory imbalance in autism, particularly affecting sensory processing. However, the brain's broader response to interventions targeting GABA pathways in individuals with autism remains poorly understood. This study tested the hypothesis that GABAergic control of information transfer across large-scale brain functional networks is altered in autism. We conducted a phase-amplitude coupling (PAC) analysis of resting-state EEG signals within and between these networks. Responses were compared after double-blind, r

Dihydrotestosterone and Finasteride Effects on Alcohol Cue-Elicited Brain Activity in Males With Heavy Episodic Drinking.

Preliminary animal and human studies have shown that blood dihydrotestosterone concentrations are increased in males with alcohol use disorder, and 5α-reductase inhibitors, which decrease dihydrotestosterone concentrations, reduce alcohol consumption. To gain mechanistic insight, we studied the effects of reduced dihydrotestosterone concentrations following pharmacological 5α-reductase inhibition on alcohol cue-elicited brain activity and alcohol craving in males with problematic alcohol use. To this end, this randomized, placebo-controlled, crossover challenge experiment investigated associat

Distinct brain pathways link depressive symptoms due to respiratory dysfunction and reduced physical activity in patients with chronic obstructive pulmonary disease: A single-center observational pilot study.

Mental health issues, such as depression, are increasingly recognized as critical comorbidities in chronic obstructive pulmonary disease (COPD). While analyses of individual domains or pairwise relationships among airflow obstruction severity, brain morphological features, and mental health conditions in COPD have provided valuable insights into their direct associations, a more integrative approach may offer additional mechanistic understanding. This study aimed to clarify potential mediators in multiple brain regions between airflow obstruction and depressive tendencies in patients with COPD

Early hypoxia treatment and oxygen-concentration alteration promote the retinal progenitor proliferation and ganglion cell maturation in human retinal organoid development.

BACKGROUND: Retinal organoids (ROs) derived from human embryonic stem cells (hESCs) hold immense potential for modeling retinal development and diseases. However, current differentiation protocols often overlook the physiological hypoxic microenvironment of early embryogenesis, potentially compromising developmental fidelity. In this study, we investigated how staged oxygen modulation-hypoxic priming followed by normoxic transition-optimizes RO development by mimicking in vivo oxygen dynamics. METHODS: The H9 hESC line was differentiated into ROs using a modified serum-free floating culture o

Sex-specific early cognitive changes are linked to global and pathway-specific genetic risk for Alzheimer's disease in at-risk individuals.

Alzheimer's disease (AD) is a progressive neurodegenerative condition in which genetic predisposition plays a key role, yet the sex-specific mechanisms linking genetic risk to early cognitive changes remain unclear. This study examined the impact of polygenic risk scores (PRS) on early cognitive changes in 318 cognitively unimpaired participants from the ALFA+ cohort, a nested longitudinal cohort from the ALFA study (see details in Study Participants Section, Methods). Participants were followed for three years, with assessments across five cognitive domains and a preclinical composite (PACC).

From Regeneration Failure to Functional Restoration: Unlocking the Neuronal-Intrinsic Regenerative Capacity as a Therapeutic Frontier for Optic Neuropathy and Glaucoma.

BACKGROUND: Optic neuropathy encompasses ocular conditions arising from traumatic or nontraumatic damage to optic nerves, causing permanent visual impairment due to retinal ganglion cell (RGC) loss and disrupted axonal connections. Glaucomatous optic neuropathy represents the most prevalent form, affecting over 70 million individuals worldwide and causing blindness in nearly 4 million people. CURRENT LIMITATIONS: Current treatments targeting elevated intraocular pressure can slow disease progression but cannot restore vision once RGC axons are lost, largely due to regeneration failure in surv
