Neurofibrillary tangle deposition in primary idiotypic isocortices using flortaucipir PET in histopathologically confirmed Alzheimer's disease.

Tau deposition in Alzheimer's disease (AD) spreads throughout the brain in a stereotypical pattern captured by the Braak neurofibrillary tangle staging scheme. While involvement of primary isocortex defines the highest stage, it is unclear whether flortaucipir PET uptake differs among primary isocortical regions. We determined relationships between flortaucipir uptake in primary visual, auditory, motor, and somatosensory isocortices and Braak stage in 155 autopsy-confirmed participants with AD. Flortaucipir uptake was observed starting at Braak stage V, with visual and auditory isocortices sho

A topographic protomap in mouse subplate neurons precedes the onset of sensory experience.

Within sensory systems, thalamic projections to layer 4 (L4) neurons establish precise neocortical topography. During development, thalamic axons first synapse onto subplate neurons (SPNs) beneath the cortical plate, however, how thalamic inputs are organized within the subplate is unknown. To examine SPN organization and activity, we generated an inducible Cplx3-CreER mouse driver line to examine SPNs during development. Before sensory onset, we found that Cplx3+ SPNs in the auditory and visual cortices exhibited topographically organized spontaneous activity. Stimulating thalamocortical affe

Does the stria of Gennari integrity reflect acquired visual field defects?

Visual input reaches the primary visual cortex (V1) at the stria of Gennari, a band of highly myelinated fibers at the level of layer IV. Damage to the visual pathways can cause visual field defects (scotomas) that prevent visual input from reaching V1. Here, we evaluate whether the stria of Gennari degrades in the absence of visual input. Using 7 Tesla MRI, we investigate the integrity of the stria of Gennari in case of acquired homonymous visual field defects, in particular hemianopia: the loss of vision in half of the visual field. These visual field defects allow us to compare stria of Gen

Transient disruption of progenitor states and layer 2/3 projection neuron generation in frontal association cortex due to 22q11.2 gene deletion.

The developmental origin of Layer 2/3 projection neuron (PN) pathology in the frontal association cortex due to heterozygous 22q11 gene deletion-the genetic foundation of elevated risk for schizophrenia and related psychiatric disorders in 22q11.2 Deletion Syndrome (22q11DS)-reflects cell state-dependent, temporally restricted vulnerability of transcriptionally diverse subsets of intermediate progenitors and neuroblasts. The molecular and cell biological consequences of these evanescent state-dependent changes include divergent proliferative capacity of the most highly proliferative progenitor

Salient affective scenes compete with concurrent visual responses in the absence of task demands.

Visual attention systems of the human brain resolve competition between concurrent inputs by prioritizing information that is relevant to an observer's goals, motives, and biological states. Research shows that concurrent distractors-especially emotionally salient stimuli-reduce task performance and visual responses to task cues. In many naturalistic viewing situations however, there are multiple competing cues with no obvious demarcation between task and distractor. The present study examined whether viewing two overlapping task-irrelevant stimulus streams produces emotion-dependent competiti

Metabolic and neural responses to ultraprocessed foods: a randomized, controlled, crossover study.

Dietary patterns worldwide have shifted toward increased consumption of ultraprocessed foods (UPFs), which has been linked to higher disease burden. One proposed mechanism underlying both UPF consumption and metabolic disease is altered post-ingestive responses relative to nutritionally similar foods. Here we report the effects of food processing on post-ingestive metabolism and brain response in a randomized, crossover study involving 57 healthy-weight adults who consumed a nutritionally matched UPF or non-UPF meal. We show that despite being nutritionally similar, UPF meals evoke greater ins

Recovering directional brain networks under temporal undersampling: an application to schizophrenia.

Two decades of functional connectivity research have established schizophrenia as a disorder of distributed dysconnectivity, with a robust thalamocortical signature: reduced prefrontal and increased sensory coupling. A fundamental issue is that functional connectivity is undirected, operates at a single slow timescale, and cannot reveal causal direction. Moreover, the mismatch between BOLD sampling speed and neural dynamics can hide edges, fabricate spurious ones, and reverse the apparent orientation of causal relationships. To overcome these limitations, we introduce a general framework for e

Linked multimodal brain alterations in depression with suicidal risk.

Suicidal ideation and behavior (SIB) represent a major clinical challenge in major depressive disorder (MDD), yet the multimodal brain structural signatures associated with SIB and their relationships with cognitive dysfunction remain poorly understood. We acquired multimodal neuroimaging measures, including gray matter volume, cortical thickness, pial area (PA), white matter fractional anisotropy, together with neurocognitive measures from 61 MDD patients with SIB (MDD-SIB), 38 without SIB (MDD-nSIB), and 142 healthy controls (HC). Early posttrauma disruptions within default mode network connectivity predict PTSD symptom trajectories: evidence from a longitudinal mega-analysis.

Posttraumatic stress disorder (PTSD) is a chronic psychiatric disorder associated with functional impairment and health burden. Evidence highlights the role of large-scale neural network disruptions, within and between the Default Mode Network (DMN) and Salience Network (SN), in PTSD pathophysiology, and early alterations in these networks may serve as biomarkers for predicting PTSD symptom trajectories and guiding early interventions. This longitudinal mega-analysis integrated resting-state functional connectivity (rsFC) data from two trauma-exposed cohorts (Tel Aviv and Toledo), collected wi

Adeno-associated virus-mediated restoration of NaV1.1 in targeted brain regions ameliorates epilepsy in symptomatic Dravet syndrome mice.

OBJECTIVE: Dravet syndrome (DS) is a developmental and epileptic encephalopathy primarily caused by haploinsufficiency of the SCN1A gene, which encodes the alpha subunit of NaV1.1 voltage-gated sodium channel. The disease manifests with febrile and spontaneous seizures, developmental delay, cognitive impairment, and increased risk of sudden unexpected death in epilepsy. Most preclinical therapeutic strategies aim to enhance endogenous Scn1a gene expression or deliver a functional copy via perinatal interventions. Although promising, these approaches face challenges in achieving efficient brain

Spatial transcriptomics reveals laminar and cell-type-specific A-to-I RNA editing signatures in the macaque cortex.

ADAR-mediated A-to-I RNA editing is crucial for proper neuronal function. However, its spatial signatures in the cerebral cortex remain largely unexplored. To address this, we developed SRDRE, an analysis framework for identifying RNA editing sites in spatial transcriptomics data. By analyzing over 40 million cells across 142 spatially annotated cortical regions in the adult macaque cortex, we uncovered laminar and cell-type-specific RNA editing landscapes. Notably, neuronal RNA editing levels correlated with cortical hierarchy in the visual and somatosensory systems. Parvalbumin neurons exhib

Subacromial bursitis is associated with short-term response to ultrasound-guided corticosteroid injection in rotator cuff disorders.

OBJECTIVE: Rotator cuff disorders (RCD) are among the most common causes of shoulder pain and functional limitation in clinical practice. Ultrasound (US)-guided subacromial corticosteroid injections are frequently used as a conservative treatment option for symptom relief in these patients. This study aimed to investigate the impact of subacromial bursitis on treatment outcomes following these injections. METHODS: This single-center observational study included 55 adult patients diagnosed with RCD who underwent US-guided subacromial corticosteroid injections. Patients were categorized into tw

Transcriptional signatures associated with cortical similarity network changes in adolescents with major depressive disorder.

BACKGROUND: Adolescent major depressive disorder (MDD) has been associated with cortical structural abnormalities. However, whether these structural changes correlate with gene expression profiles remains unclear. Here, we investigated the cortical similarity network alterations and the associated transcriptional patterns in adolescents with MDD using a novel Morphometric Inverse Divergence (MIND) network model. METHODS: Using structural images from 174 adolescents with MDD and 82 healthy controls (HCs), we constructed individual MIND networks based on multiple morphological features. We comp

Heidenhain Disease: Challenging Diagnosis of a Creutzfeldt-Jakob Variant.

Creutzfeldt-Jakob disease (CJD) is a rare disorder caused by prions, characterized by rapid deterioration of cognitive and neurological functions. The Heidenhain variant is an uncommon form in which visual symptoms are prominent at onset due to involvement of the occipital cortex, leading to a delayed diagnosis. We present the case of a 67-year-old woman who initially exhibited decreased visual acuity, followed by difficulties in reaching for and identifying objects, experiencing optic apraxia, psychomotor slowing, perseverative behavior, and impaired judgment. Brain magnetic resonance imaging

Autism genes converge on three functional programs organized by neuronal subclass, developmental timing, and cortical patterning.

Our companion sequencing study uncovered 253 genes robustly associated with autism spectrum disorder (ASD), yet the biological programs they impact, and the cellular, developmental, and spatial contexts in which they converge, remain unresolved. Here we systematically contextualize genes associated with ASD across neurodevelopment and cortical areas, integrating developmental single-cell and spatial atlases with gene- and isoform co-expression, regulatory, proteomic, and synaptic networks. Genetic burden concentrates within temporally-resolved neuronal subclasses: newborn excitatory neurons, i

Early postlesional visuomotor capacity after V1 damage in the macaque.

Damage to the primary visual cortex (V1) causes field defects in conscious vision, yet stimuli in the affected field may still guide saccades and other visuomotor responses ("blindsight"). Chronic blindsight is well documented, but early postlesional saccade profiles to scotoma stimuli remain poorly characterized. We mapped visuomotor functions in two macaques 3 to 4 weeks after a unilateral V1 lesion using a perimetry-like saccade assay up to 35° eccentricity, with analyses of saccade direction and catch-trial baselines. Targets had either unpredictable onset, as in clinical perimetry, or tem

The default mode network exhibits circuit-specific selective vulnerability despite widespread amyloid burden in an animal model of Alzheimer's disease.

In Alzheimer's Disease (AD) the default mode network (DMN) exhibits selective vulnerability to functional decline and amyloid beta (Aβ) plaques in early AD. DMN vulnerability may simply be the result of Aβ accumulation. Alternately, Aβ may unmask an intrinsic susceptibility of DMN circuits. Should DMN vulnerability persist amidst global Aβ deposition, this would support the latter hypothesis. We tested this using long-term, multi-region recordings of single units in mice characterized by widespread Aβ (APP/PS1). We tracked a neuronal population within the retrosplenial cortex (a DMN region) an

Psilocybin Prolongs the Neurovascular Coupling Response in Mouse Visual Cortex.

Psilocybin has profound therapeutic potential for various mental health disorders, but its mechanisms of action are unknown. Functional MRI studies have reported the effects of psilocybin on brain activity and connectivity; however, these measurements rely on neurovascular coupling to infer neural activity changes and assume that blood flow responses to neural activity are not altered by psilocybin. Using two-photon excited fluorescence imaging in the visual cortex of awake mice to simultaneously measure neural activity and capillary blood flow dynamics, we found that psilocybin administration

Structural MRI alterations in children with form-deprivation amblyopia: a propensity score-matched case-control study.

AIMS: Form-deprivation amblyopia (FDAM) is a severe neurodevelopmental disorder from early visual deprivation. This study uses T1-weighted MRI to explore its neuroanatomical correlates. METHODS: 18 Chinese children with bilateral FDAM and 36 propensity score-matched healthy Chinese controls (1-to-2 matched for age, sex and mean cortical thickness) underwent 3.0T T1-weighted MRI (1×1×1 mm³ resolution). Cortical and subcortical structures were parcellated using FreeSurfer V.7.3.2 with multiatlas (Desikan-Killiany-Tourville, Destrieux, subset Brodmann and Aseg), alongside specialised modules for

Progressive and Short-Interval Changes Observed in the Corticospinal Tract and Corpus Callosum of Patients with Amyotrophic Lateral Sclerosis: A Texture Analysis Study.

BACKGROUND AND PURPOSE: Diffusion MRI measures indicative of white matter integrity have consistently been shown to be altered in the state of the corticospinal tract (CST) and corpus callosum (CC) of patients with amyotrophic lateral sclerosis (ALS). However, diffusion MRI acquisitions are not routinely collected as part of the standard medical imaging of patients with ALS. T1-weighted MRI scans are commonly available in the clinical assessment of most patients with ALS. While visual inspection of these scans reveals little about the cerebral pathology of ALS, analysis of their textural patte
