The Eye-Brain Axis in High Myopia: From Retinal Deformation to Cortical Network Reorganization.

High myopia is a major cause of irreversible visual impairment and is increasingly recognized as a condition involving interactions between ocular remodeling and central neural systems. This scoping review synthesizes current evidence on structural, functional, genetic, and regulatory links between the eye and brain in high myopia. We systematically searched PubMed and Web of Science from database inception to August 2026, identifying 54 eligible primary studies. The available evidence indicates that high myopia is associated with retinal and optic nerve abnormalities, altered white matter mic

Kidney Transplantation From a Heterozygous CLDN19 Carrier in a Child With FHHNC: A 2-Year Follow-Up.

BACKGROUND: Familial hypomagnesemia with hypercalciuria and nephrocalcinosis is a rare autosomal recessive tubulopathy caused by mutations in CLDN16 or CLDN19, leading to progressive chronic kidney disease. While kidney transplantation is the definitive treatment, the safety of using living-related heterozygous carrier donors remains debated due to potential subclinical tubular dysfunction. We report the 2-year follow-up of a pediatric patient with FHHNC and a CLDN19 mutation who received a graft from his heterozygous carrier mother. CASE REPORT: A male patient presented at 3 months of age wi

Genotype-phenotype variability in TRMT10A deficiency: divergent developmental outcomes associated with biallelic loss-of-function variants.

Loss-of-function variants in TRMT10A are known to cause a rare autosomal recessive disorder primarily characterized by microcephaly, short stature, and impaired glucose metabolism. However, the extent of phenotypic variability and the factors influencing clinical heterogeneity remain poorly understood. Here, we describe two individuals harboring distinct biallelic loss-of-function variants in TRMT10A who exhibited divergent developmental phenotypes. The first patient, a 34-year-old man with a homozygous early frameshift variant, presented with preserved cognitive function and previously unrepo

The growing burden of myopia in Europe: evidence from population-based cohorts.

Myopia is a childhood-onset condition driven by excessive axial ocular elongation, associated with increased risk of sight-threatening complications and irreversible visual impairment. Despite rising global prevalence, epidemiological data on long-term trends in Europe remain limited. This study examines temporal trends in myopia prevalence and axial length (AL) across European adult birth cohorts. We conducted a comparative birth-cohort analysis using harmonised data from 28 population-based studies (n = 81,828) across nine countries participating in the European Eye Epidemiology (E3) Consort

BMI moderates the association between physical fitness and spherical equivalent in Chinese college students: a cross-sectional study.

BACKGROUND: Myopia has become a global public health crisis, with young adults representing a priority population for prevention. While physical fitness is recognized as a protective factor, whether this protection is uniformly distributed across different BMI subgroups remains unclear. This study examined the moderating role of BMI in the relationship between physical fitness and spherical equivalent (SE) among Chinese university students. METHODS: A cross-sectional study was conducted with 300 college students (100 normal weight, 100 overweight, 100 obese) from Zhejiang Province, China. Phy

The key mechanisms and intervention strategies of cell-type-specific mitochondrial metabolism in ophthalmic diseases.

The eye is one of the most metabolically active organs, and mitochondria serve as the central hub for cellular energy metabolism by providing adenosine triphosphate through oxidative phosphorylation. Disruption of mitochondrial metabolism-including insufficient adenosine triphosphate synthesis, excessive production of reactive oxygen species, and dysregulated quality control-can trigger various ocular diseases. Common blinding diseases such as myopia, diabetic retinopathy, and glaucoma are closely associated with mitochondrial dysfunction in specific cell types. This review delineates the mech

Hyperopic reserve, early myopia risk and screening strategies in children: A multi-regional analysis from the CREAM-KIDS consortium.

Reduced hyperopic reserve (HR) in early childhood is a strong predictor of future myopia. A harmonized dataset from the CREAM-Kids Consortium was utilized to assess regional differences in HR. Among 7-year olds, mean (SD) HR was 1.44 (1.17) D in Europe, 1.32 (0.80) D in Australia, 0.78 (0.89) D in Hong Kong, and 0.59 (0.66) D in Singapore (p < 0.001). The annualized decline in HR was -0.12 D/year among European-Australian followed from age 6.73 (0.64) to 11.64 (1.49), and -0.35 D/year among Asian followed from age 7.71 (0.77) to 10.71 (0.77). In European-Australian aged 6-7, the sensitivity an

Routine baseline clinical data do not reliably predict poor first-year myopia control with low-dose atropine: a machine-learning benchmarking study.

PURPOSE: The aim of this study was to determine whether routinely recorded baseline clinical factors, alone or combined with machine-learning predictive modeling, can provide individual risk stratification for myopia control by identifying children who will progress more than 0.25 diopters (D) during their first year of low-dose atropine treatment. METHODS: From a 555-child source cohort treated initially with atropine 0.01% or 0.02%, 468 children had qualifying 9- to 15-month follow-up; 427 children started atropine monotherapy without a baseline optical adjunct. A child was classified as po

Amine-responsive Rhodopsin-like GPCRs in ocular pathology: mechanistic associations, candidate biomarkers, and therapeutic potential.

Rhodopsin-like G protein-coupled receptors (GPCRs) play a pivotal role in ophthalmic pathologies, and recent studies have substantially advanced our understanding of their underlying molecular mechanisms. Amine-responsive Rhodopsin-like GPCRs (ARh-GPCRs), encompassing adrenoceptors, dopamine receptors, muscarinic acetylcholine receptors, histamine receptors, serotonin receptors, melatonin receptors, and trace amine-associated receptors, are involved in a variety of ocular pathological processes and have emerged as key therapeutic targets. Elucidating their functional roles, alongside those of

Interocular symmetry of ocular rigidity and related ocular parameters in isometropia.

PURPOSE: Driven by scleral stiffness, ocular rigidity (OR) is emerging as a critical factor in refractive development. Although a common assumption is that OR is symmetric in a healthy individual's eyes, empirical data remains scarce. This study directly tests this assumption by quantifying intereye OR differences in healthy, isometropic subjects. METHODS: Isometropia was defined as intereye differences in spherical equivalent refraction (SER) of <0.50 diopters (D) and axial length (AL) of <0.15 mm. OR was measured using a validated method based on video-rate optical coherence tomography imag

Establishment of a TGF-β-Induced Rodent Myopia Model Using Subconjunctival Injections.

Myopia is a major global public health concern characterized by excessive axial elongation and scleral extracellular matrix (ECM) remodeling. While experimental models are essential for investigating these mechanisms, traditional optical defocus methods in small rodents present technical limitations. Here, a standardizable pharmacological myopia model was established and evaluated via repeated subconjunctival injections of transforming growth factor-beta (TGF-β). To achieve targeted delivery, a dual-handed surgical stabilization technique was developed to evert the upper eyelid, allowing a 33-
