Old molecule, new scrutiny: Benzoyl peroxide in dermatology.

INTRODUCTION: Benzoyl peroxide (BPO) is a widely available over-the-counter topical medication approved in the 1960s for acne vulgaris treatment. NICE guidelines recommend BPO as a monotherapy or in combination with topical retinoids or antibiotics. However, concerns persist regarding optimal concentration and long-term safety, particularly following recent controversies surrounding benzene formation. AIM: To systematically assess evidence on the indications, efficacy, and safety of topical BPO across dermatological conditions. METHODOLOGY: A literature search following the PRISMA-P guidelin

An enhanced grey wolf optimizer-based Facebook artificial intelligence similarity search for Alzheimer's disease diagnosis.

Early diagnosis of Alzheimer's disease (AD) and its prodromal stage, mild cognitive impairment (MCI), requires efficient computational frameworks capable of handling high-dimensional and large-scale 18F-FDG-PET data. To address challenges in feature redundancy and computational efficiency, we propose a unified framework integrating an enhanced Grey Wolf Optimizer (CDL-GWO) with approximate nearest neighbor search based on Facebook AI Similarity Search (FAISS). The proposed CDL-GWO incorporates Logistic chaotic initialization to improve search space coverage, dual random projection to enhance p

Distinct migration behaviors of monocyte-derived versus resident langerhans cells drive the pathogenesis of imiquimod-induced skin inflammation.

BACKGROUND: Langerhans cells (LCs) consist of embryonically derived resident LCs (rLCs) and monocyte-derived LCs (moLCs), yet how distinct LC subsets migrate and function during psoriatic skin inflammation remains incompletely understood. OBJECTIVE: To characterize the migratory behavior and immunological roles of rLCs and moLCs in imiquimod (IMQ)-induced skin inflammation. METHODS: We examined LC subset dynamics in an IMQ-induced murine dermatitis model using flow cytometry and immunofluorescence. To dissect subset-specific functions, moLCs were selectively ablated in Cd207fl/flLyz2Cre cond

Exercise-Induced Acid-Base, Electrolyte, and Metabolic Responses in Colombian Creole Horses Across Different Gait Modalities.

Exercise physiology in Colombian Creole Horses remains poorly characterized despite the breed's extensive use in athletic activities. The objective of this study was to evaluate exercise-induced changes in blood biochemical and gas parameters and to determine whether these responses differed according to sex, geographic origin, or gait modality. An analytical observational study with repeated measures was conducted in 76 athletic CCHs. Blood samples were obtained from the jugular vein at rest and immediately after exercise to measure acid-base indicators (pH, carbon dioxide pressure, bicarbona

Joint association of dietary fatty acid patterns identified by Gaussian graphical models and cognitive impairment status with all-cause mortality in U.S. adults: A prospective cohort study.

BACKGROUND: The association between dietary fatty acid patterns (DFAPs) and cognitive function jointly affecting long-term survival remains poorly understood. Network-based methods addressing conditional dependencies among fatty acids may reveal novel patterns, yet the joint association of DFAPs derived from Gaussian Graphical Models (GGMs) and cognitive status with all-cause mortality (ACM) risk has not been examined in nationally representative U.S. adults. METHODS: GGMs were applied to 24-hour recall data for 19 fatty acids from 2446 adults aged ≥18 years in the NHANES 2011-2014 cycles. Ps

Dissecting Alzheimer's proteomic landscape through NULISA profiling of brain cell-specific extracellular vesicles.

INTRODUCTION: Blood-based biomarkers are essential for early detection, monitoring, and therapeutic development in Alzheimer's disease (AD) and related dementia (ADRD), but current assays lack brain cell specificity and sensitivity to low-abundant proteins. METHOD: We isolated the following brain cell-derived small extracellular vesicles (sEV) from the plasma of individuals with normal cognition (CN), mild cognitive impairment (MCI), or ADRD: neurons (NDE), astrocytes (ADE), microglia (MDE), oligodendrocytes (ODE), pericytes (PDE), and endothelial cells (EDE). Using NULISAseq, we profiled 122

De novo variants in NPTN cause a neurodevelopmental disorder with autism and neuroplastin-PMCA hypofunction.

BACKGROUND: NPTN encodes human neuroplastin (hNp), a transmembrane immunoglobulin (Ig)-superfamily glycoprotein and a subunit of the plasma membrane calcium (Ca2+)-ATPases (PMCA). The critical importance of hNp and its associations with PMCA in the human brain remains unknown. METHODS: Here, we describe de novo NPTN variants in individuals with autism and mild-to-severe DD/ID and evaluate their effects using animal models and in silico, molecular, and cellular approaches. RESULTS: Four individuals present variants affecting the two hNp isoforms, hNp55 and hNp65. Other four variants affect on

Othello syndrome as an early manifestation of Alzheimer's disease: an anatomoclinical case study.

Othello syndrome (OS) is characterized by pathological jealousy toward one's spouse, usually marked by delusions of infidelity. We report the first case of OS in Brazil, similar to the case described by Alzheimer in 1906, in a 54-year-old right-handed patient diagnosed with Alzheimer's disease (AD). The diagnostic workup included a clinical history and a positive family history of dementia, the Clinical Dementia Rating (CDR) scale, the Edinburgh Handedness Inventory, and the Mini-Mental State Examination (MMSE). Electroencephalography revealed slow waves predominantly in the bilateral temporo-

Liddle syndrome caused by SCNN1B mutation presenting with severe early-onset hypertension and hypokalemia in an adolescent: a case report.

BACKGROUND: Liddle syndrome is a rare autosomal dominant form of monogenic hypertension caused by gain-of-function mutations in genes encoding the epithelial sodium channel (ENaC). The disease is characterized by early-onset hypertension, hypokalemia, metabolic alkalosis, and suppression of the renin-angiotensin-aldosterone system (RAAS). Early diagnosis is essential because targeted therapy with ENaC inhibitors can effectively control blood pressure and prevent end-organ damage, whereas mineralocorticoid receptor antagonists are ineffective. CASE PRESENTATION: We report a 15-year-old Chinese

Anti-SRP immune-mediated necrotizing myopathy from childhood to adulthood: A case series highlighting rituximab-responsive disease.

ObjectivesAnti-signal recognition particle (SRP) myositis is a rare subset of immune-mediated necrotizing myopathy (IMNM). It is characterized by proximal muscle weakness, markedly elevated serum creatine kinase (CK) levels, and poor response to conventional therapies. Evidence for optimal management remains limited, particularly regarding the long-term outcomes of rituximab treatment.MethodWe retrospectively reviewed anti-SRP myositis patients who received rituximab at Seoul National University Hospital. Diagnosis was based on clinical features, elevated CK levels, and positivity for anti-SRP

Severe Postoperative Hypernatremia in an Adolescent Following Sleeve Gastrectomy.

A 14-year-old boy presented 6 weeks after out-of-country sleeve gastrectomy with profound hypernatremic dehydration (serum sodium 195 mmol/L), stage 3 acute kidney injury, and severe rhabdomyolysis. The patient had no structured postoperative follow-up and no primary care pediatrician and relied on sports drinks and intermittent intravenous fluids at a medical spa for hydration. Despite extreme hypernatremia, neurologic findings were relatively mild, consistent with gradual onset. Judicious rehydration and correction of serum sodium in the pediatric intensive care unit resulted in full recover

Enzymatic preparation and functional development of agaro-oligosaccharides: Advances in structure-function relationships and application challenges.

Agaro-oligosaccharides (AGOS), derived from agar depolymerization, are marine-derived functional carbohydrates with diverse structures and promising bioactivities, including antioxidant, anti-inflammatory, and metabolic regulatory effects. Enzymatic degradation is an attractive strategy for AGOS production due to its mild conditions, high selectivity, and ability to preserve functional groups. However, the rational preparation of structurally defined AGOS remains challenging because product profiles are determined by complex interactions among agarase catalytic architecture, substrate recognit

Efficacy and Safety of Low-Molecular-Weight Collagen Peptide GT for Improving Hair Condition in Individuals with Damaged Hair: A Randomized, Double-Blind, Placebo-Controlled Trial.

Addressing hair damage is essential for maintaining overall hair and scalp health. Previous studies have suggested that low-molecular-weight collagen peptides may support hair follicle biology through regulation of extracellular matrix homeostasis and cellular signaling pathways. In addition, recent preclinical studies have demonstrated that low-molecular-weight fish collagen peptides promote hair regrowth through activation of proliferative signaling and suppression of inhibitory pathways. However, their clinical efficacy in improving hair damage has not been clearly established. In this stud

Cytogenetics of Shwachman Diamond syndrome: 27 years report of the Italian cohort and review of the literature.

Shwachman-Diamond syndrome (SDS1) is an autosomal recessive disorder principally characterized by exocrine pancreatic insufficiency, a mild to severe bone marrow failure, peripheral blood cytopenias and an increased risk of developing myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML). Since 1995 several cytogenetic studies were performed mainly in relation to the myelodysplastic/myeloproliferative risk, and recurrent chromosome changes in bone marrow (BM) were discovered. In this paper we extensively reviewed the cytogenetic studies in SDS patients, and the related significance h

Crystal precipitates of local anaesthetic and adjuvant mixtures can be visualised and graded using ultrahigh-frequency ultrasound imaging.

BACKGROUND: The use of mixtures of local anaesthetics and/or adjuvants is a common practice in regional anaesthesia, aimed at optimising onset time and prolonging block duration. However, several previous in vitro studies have demonstrated that mixing local anaesthetics may lead to crystallisation. A major barrier to understanding their potential physiological effects has been the lack of imaging techniques capable of detecting crystal formations beyond conventional microscopy. METHODS: In this in vitro study, ropivacaine 0.75%, lidocaine 2%, chloroprocaine 2% and mepivacaine 2%, as well as s
