Use of a mouse model for the isolation of Borrelia puertoricensis from soft ticks.

The isolation of tick-borne relapsing fever (TBRF) spirochetes has proven to be a useful tool to understand their distribution in geographic areas where the tick vectors inhabit. However, their isolation and culture are not easy and in general an animal model is needed to achieve this task. Here, argasid ticks were collected from a neighborhood in Ciudad Caucel, and they were identified as Ornithodoros (Alectorobius) puertoricensis. To determine whether these were infected with TBRF bacteria they were fed with healthy mice but only a low burden of spirochetes was observed. An immunosuppressed

Unmasking Borrelia species: A comprehensive review of their presence in Iran.

Borrelia species are the causative agents of Lyme disease (LD) and tick-borne relapsing fever (TBRF) in humans and animals. These pathogens are transmitted through hard and soft ticks. The increasing tick population, influenced by climate change, underscores the urgent need for enhanced research on tick-borne diseases. Iran, situated in southwestern Asia, boasts a diverse climate that supports a wide range of tick species and their vertebrate hosts. While TBRF is endemic to Iran, recent reports suggest the presence of LD in the country as well. Understanding the various Borrelia species, their

Antigenic variation is caused by long plasmid segment conversion in a hard tick-borne relapsing fever Borrelia miyamotoi.

Borrelia miyamotoi is a hard tick-borne spirochete genetically related to relapsing fever Borrelia and the etiological agent of an emerging infectious disease in humans. Like relapsing fever Borrelia, B. miyamotoi carries clusters of gene cassettes encoding variable major proteins (Vmps) on multiple linear plasmids and shows antigenic variation in mammalian hosts by switching the expression vmp gene cassette. However, it remains unknown how the switch occurs in B. miyamotoi. Here we determined the whole genome sequences of Japanese B. miyamotoi strains to identify the repertoire and arrangemen

[West African tick-borne relapsing fever, a neglected disease of major public health importance in Senegal].

INTRODUCTION: Tick-borne relapsing fever (TBRF) is a zoonotic disease caused by various species of Borrelia that are transmitted to humans worldwide through the bite of soft ticks belonging to the genus Ornithodoros. In Senegal, as in other endemic regions of West Africa, cases of TBRF caused by B. crocidurae and transmitted by Ornithodoros sonrai ticks are underdiagnosed. The disease is not well known among doctors, nurses, or the general population. In some regions, TBRF is the leading cause of vector-borne fever, surpassing malaria. MATERIALS AND METHODS: This narrative and systematic revi

Experimental transmission of the relapsing fever spirochete Borrelia persica in its tick vector Ornithodoros tholozani by transstadial, transovarial, and hyperparasitism routes with description of dynamics within the tick host.

BACKGROUND: Borrelia persica causes tick-borne relapsing fever, a potentially fatal human and animal disease, in the Middle East and Central Asia. The transmission of B. persica in Ornithodoros tholozani ticks has been reported to be transovarial, but little is known about the pathogen's lifecycle in its vector and other pathways of transmission. METHODS: To further understand the transmission of B. persica, colony-bred O. tholozani ticks were fed on blood in an animal-free system, followed during three lifecycles, and infected with cultured B. persica. Spirochetes were detected, and bacteria

Guidelines for tick-borne diseases (excluding Lyme borreliosis): diagnosis and management.

Other tick-borne microorganisms can cause symptoms that differ from those of Lyme borreliosis, often with a more acute course. Fever is the primary clinical warning sign, which may be accompanied by various other symptoms-cutaneous, neuromeningeal, and lymphatic-that help guide diagnosis. A clinical approach makes it relatively straightforward to distinguish Lyme borreliosis from other tick-borne diseases, with the common factor being the tick as the vector. Biological abnormalities (anaemia, thrombocytopenia, cytolysis, etc.) are often present in tick-borne diseases, unlike in Lyme borreliosi

Experimental transmission of a novel relapsing fever group Borrelia harbored by Ornithodoros octodontus (Ixodida: Argasidae) in Chile.

Tick-borne relapsing fever spirochetes of genus Borrelia thrive in enzootic cycles involving Ornithodoros spp. (Argasidae) mainly, and rodents. The isolation of these spirochetes usually involves a murine model in which ticks are fed and the spirochetes detected in blood several days later. Such an experiment also demonstrates that a given species of tick is competent in the transmission of the bacteria. Here, soft ticks Ornithodoros octodontus were collected in Northern Chile with the objective to experimentally determine its capacity to transmit a Borrelia sp. detected in a previous study. T

Isolation and Genetic Characterization of the Human Relapsing Fever Spirochete Borrelia persica from a Dog with Improved Cultivation Techniques.

Borrelia persica is the causative agent of tick-borne relapsing fever in Israel and is prevalent in the Eastern Mediterranean basin and parts of Asia. Infection with B. persica causes severe illness and potentially life-threatening complications in humans and companion animals. Isolation and in vitro cultivation of B. persica in culture medium is difficult and there are only two previous isolates of this spirochete. Here, we describe the first cultivation of B. persica from an ill dog. Isolation from the dog's blood was performed with the Pettenkofer-LMU Bp medium and spirochetes with vibrant

Systematic investigation of the Borrelia miyamotoi spirochetes in ticks, wildlife and domestic animal hosts in Yunnan province, Southwest China.

BACKGROUND: Borrelia miyamotoi is a spirochete species transmitted via hard ticks. Following its discovery in Japan, this pathogen has been detected around the world, and is increasingly confirmed as a human pathogen causing febrile disease, namely relapsing fever. Its presence has been confirmed in the Northeast China. However, there is little information regarding the presence of B. miyamotoi and other hard-tick-borne relapsing fever spirochetes in southern China including Yunnan province, where tick and animal species are abundant and many people both inhabit and visit for recreation. Late-stage borreliosis and substance abuse.

BACKGROUND: Infectious diseases can contribute to substance abuse. Here, a fatal case of borreliosis and substance abuse is reported. This patient had a history of multiple tick bites and increasing multisystem symptoms, yet diagnosis and treatment were delayed. He experimented with multiple substances including phencyclidine (PCP), an N-methyl-d-aspartate (NMDA) receptor antagonist that opposes NMDA agonism caused by Borrelia infection. During PCP withdrawal, he committed one homicide, two assaults, and suicide. METHODS: Brain tissue was obtained from autopsy and stained for microglial activ

Exploring the transcriptome of immature stages of Ornithodoros hermsi, the soft-tick vector of tick-borne relapsing fever.

Blood-feeding behavior has independently evolved in arthropods multiple times. Unlike hard ticks, soft ticks employ a rapid-feeding strategy for hematophagy, and there are comparatively limited studies on the transcriptomes of these organisms. This study investigates the soft tick Ornithodoros hermsi, conducting histopathological examinations at bitten skin sites and tick whole-body transcriptomic analyses across various developmental and feeding stages, including larvae, 1st-nymphal, and 2nd-nymphal stages. The results revealed the ability of O. hermsi to induce skin hemorrhage at the bite si

Tick-Borne Relapsing Fever Infection from a Bunker, a Case Report on a "One Health" Approach.

Background: Tick-borne relapsing fever (TBRF) caused by Borrelia persica is an endemic disease in Israel and highly prevalent in military personnel. Prevention among the Israel Defense Force soldiers is based on increased awareness mainly in hyperendemic areas and selective postexposure prophylaxis with doxycycline. In this study, we report the presence of a suspected outbreak of TBRF in four soldiers who spent 30 h inside a deserted bunker. Materials and Methods: Clinical data on TBRF suspected cases were retrieved from clinical records, soft ticks were collected using carbon dioxide (CO2) tr

Development and validation of systems for genetic manipulation of the Old World tick-borne relapsing fever spirochete, Borrelia duttonii.

Relapsing fever (RF), a vector-borne disease caused by Borrelia spp., is characterized by recurring febrile episodes due to repeated bouts of bacteremia. RF spirochetes can be geographically and phylogenetically divided into two distinct groups; Old World RF Borrelia (found in Africa, Asia, and Europe) and New World RF Borrelia (found in the Americas). While RF is a rarely reported disease in the Americas, RF is prevalent in endemic parts of Africa. Despite phylogenetic differences between Old World and New World RF Borrelia and higher incidence of disease associated with Old World RF spiroche

Production, purification, and quality assessment of borrelial proteins CspZ from Borrelia burgdorferi and FhbA from Borrelia hermsii.

Borrelia, spirochetes transmitted by ticks, are the etiological agents of numerous multisystemic diseases, such as Lyme borreliosis (LB) and tick-borne relapsing fever (TBRF). This study focuses on two surface proteins from two Borrelia subspecies involved in these diseases: CspZ, expressed by Borrelia burgdorferi sensu stricto (also named BbCRASP-2 for complement regulator-acquiring surface protein 2), and the factor H binding A (FhbA), expressed by Borrelia hermsii. Numerous subspecies of Borrelia, including these latter, are able to evade the immune defenses of a variety of potential verteb

Toxicity of Trixis vauthieri DC essential oil on Lutzomyia longipalpis (Diptera, Psychodidae, Phlebotominae), a proven vector of visceral leishmaniasis in Brazil.

BACKGROUND OBJECTIVES: In Brazil, one of the visceral leishmaniasis control measures in urban environments is the elimination of Lutzomyia longipalpis, which occurs through the application of pyrethroid insecticides with residual action in homes and outbuildings. Due to the loss of sensitivity of this vector to these insecticides, the search for more efficient insecticide compounds against L. longipalpis has been intensified. The objective of this work was to evaluate the toxicity of Trixis vauthieri essential oil on adult sandflies of the species L. longipalpis, and identify the phytochemical
