PURPOSE: Primary open-angle glaucoma (POAG) is a common ocular disease, associated with abnormalities in aqueous humor circulation and an increase in intraocular pressure (IOP), leading to progressive optical neuropathy and loss of vision. POAG pathogenesis includes alterations of the structural properties of the sclera, especially in the optic nerve head area, contributing to the degeneration of the retinal ganglion cells. Abnormal sclera biomechanics hinder adequate compensation of IOP fluctuations, thus aggravating POAG progression. The proteomic basis of biomechanical disorders in glaucoma

BACKGROUND: Spondylolisthesis is a common cause of lower back pain, often accompanied by spinal stenosis, disc degeneration, and altered biomechanics. Reduction is achieved anteriorly through anterior lumbar interbody fusion (ALIF) or posteriorly through pedicle screws/rods. ALIF allows for favorable biomechanics for fusion but has limited reduction capability in high-grade slips. Anterior reduction allows for increased slip reduction without relying on spacer impaction but has only been explored in grade I spondylolistheses. The objective of this study was to evaluate the reduction capacity o

INTRODUCTION: Lyme disease is caused by Borrelia burgdorferi sensu lato that is transmitted through the bite of infectious ticks. Within the US active duty military component, Lyme is the most frequently reported vector-borne disease. There have not been reports on Lyme disease prevalence in Central America, but reports of travelers who contracted rickettsiosis after their trip to Honduras suggest a need for an increased tick-borne disease surveillance, including Lyme disease. The aim of this study is to determine the prevalence of Lyme disease in US military personnel deployed to Honduras.

Lyme disease spirochetes are maintained in natural reservoirs before spilling over into human populations. Targeting these reservoirs with vaccinations or antibiotics could impact the Borrelia burgdorferi enzootic cycle and reduce the risk of human Lyme disease. In this work we report that the narrow-spectrum antibiotic hygromycin A is sufficient to disrupt B burgdorferi transmission from the main eastern US reservoir, Peromyscus leucopus, to ticks. Additionally, hygromycin A-containing baits can clear B burgdorferi from P leucopus. These studies lay the foundation for the use of hygromycin A

Powassan virus (POWV), a tick-borne flavivirus, poses a public health risk with an estimated case fatality rate of approximately 10%. Its limited surveillance results in underreporting. Using Lyme disease data as a proxy, we developed an exploratory simulation study that estimated 4649 POWV cases in the United States from 2018 to 2022, as opposed to 156 reported cases. We further estimated mortality rates (0.2%-1%), which were lower than reported rates (4.8%-21%). These discrepancies suggest that POWV cases go undetected, likely due to asymptomatic and mild to moderate presentations. Our findi

BACKGROUND: Lyme borreliosis (LB) is one of the most commonly diagnosed tick-borne diseases globally, caused by Borrelia spirochetes. Despite advances in laboratory diagnostics, both overdiagnosis and underdiagnosis of LB remain significant issues. Nonspecific symptoms such as chronic fatigue or musculoskeletal complaints are often misattributed to LB, creating controversy and delaying appropriate diagnosis and treatment.
OBJECTIVE: To analyze clinical data and diagnostic results of patients referred to the Department of Infectious Diseases and Neuroinfections, Medical University of Bialystok

CRISPR/Cas systems have the potential to revolutionize DNA detection of vector-borne pathogens with highly specific and user-friendly assays. One such system, named DNA Endonuclease Targeted CRISPR Trans Reporter (DETECTR), uses a guide RNA (gRNA) and Cas enzyme to bind to and cut DNA targets. Following cutting, Cas12a exhibits non-specific collateral cleavage of single-stranded DNA (ssDNA). A ssDNA reporter in the reaction allows the trans-cleavage activity to be harnessed as an amplified output signal upon recognition of the target by the Cas12a/gRNA complex. We developed a DETECTR assay to

Dozens of over-the-counter tests offer to evaluate users for an array of contagious illnesses, including sexually transmitted infections, acute respiratory infections like coronavirus disease 2019 (COVID-19) and influenza, and complicated chronic infections like Lyme disease, tuberculosis, and human immunodeficiency virus (HIV). Diagnosing any of these conditions in a home setting raises both logistical and ethical issues that may prevent home testing from becoming the public health benefit its advocates envision. Over the last century, early home tests such as the first urine glucose screenin

The blacklegged tick (Ixodes scapularis) is a primary vector of medical concern in the eastern United States, capable of infesting a wide range of hosts. The first established population in Ohio was detected on rural residential properties in Coshocton County and has since expanded across the state. To assess current conditions, these index sites were revisited, and questing ticks were collected between 2019 and 2021, with small mammals sampled in 2021. Ticks and host tissues were screened for Borrelia burgdorferi sensu lato, Borrelia burgdorferi sensu stricto, Borrelia mayonii, and Anaplasma

BACKGROUND: Regional blood donor screening for Babesia has been conducted in the United States since 2019, using highly sensitive and specific nucleic acid testing (NAT). Currently, there are no recommendations regarding the management of asymptomatic blood donors who test positive for Babesia.
METHODS: A multidisciplinary expert panel was convened to develop guidance for the management of asymptomatic Babesia-infected blood donors. Additionally, a survey was distributed through the Infectious Diseases Society of America (IDSA) Emerging Infections Network (EIN) to evaluate how a geographicall

BosR is a novel nucleic acid-binding protein in the ferric uptake regulator (FUR) family that regulates gene expression in the Lyme disease spirochete Borrelia (Borreliella) burgdorferi. This issue of Molecular Microbiology contains a comprehensive transcriptomic study that keenly defines the regulatory swath of BosR in the vertebrate host of B. burgdorferi. Despite homology to Fur-like and PurR-like orthologs, BosR has traditionally been linked to regulation of RpoS, the alternative sigma factor that controls the regulon required for establishing a vertebrate infection. However, BosR regulate

Borrelia burgdorferi, one of the most prevalent tick-borne pathogens, can cause a complex and multisystem illness called Lyme disease, where there has been an unmet need for novel therapeutic or preventive strategies. We previously identified an essential protein-protein interaction (PPI) event in B. burgdorferi involving two unique proteins, BB0323 and BB0238; herein, we show that this PPI is indispensable for long-term borrelial survival in mammals and explore its potential as a novel target for small molecule therapeutics. Using X-ray crystallography, we solved the structure of the BB0238-B

BACKGROUND: Ticks are obligate blood-feeding parasites associated with a huge diversity of diseases globally. The hard tick Ixodes ricinus is the key vector of Lyme borreliosis and tick-borne encephalitis in Western Eurasia. Ixodes ticks have large and repetitive genomes that are not yet well characterized.
RESULTS: Here we generate two high-quality I. ricinus genome assemblies, with haploid genome assembly sizes of approximately 2.15 Gbp. We find transposable elements comprise at least 69% of the two I. ricinus genome assemblies, amongst the highest proportions found in animals. The transpos

INTRODUCTION: Lyme disease, caused by Borrelia burgdorferi, is a zoonotic infection affecting the skin, nervous system, joints, and heart. Diagnosis relies on clinical history, symptoms, and a two steps serologic test confirmed by Western Blot. Although it frequently affects other systems, data on the haematological involvement spectrum of Lyme disease appears to be limited to case reports, and there are few studies that clearly demonstrate its relationship with haemolytic anaemia. This case highlights Lyme disease presenting with autoimmune hemolytic anemia (AIHA) and thrombocytopenia. CASE:

Quantifying the strengths of interactions in complex vector-borne disease ecological systems is challenging. Yet, overcoming this challenge is fundamental for understanding the ecological mechanisms shaping disease hazard. Here we quantified the strengths of the hypothesised direct and indirect mechanistic pathways through which deer affect ticks and one of the Lyme borreliosis pathogens, Borrelia afzelii, by conducting a combined analysis of three previously published datasets from 39 sites across Scotland. Structural equation modelling revealed that, as predicted, deer had a strong positive

In recent decades, increased incidences of tick-borne diseases, including Lyme disease (Borrelia burgdorferi), Anaplasmosis (Anaplasma phagocytophilum), and Babesiosis (Babesia microti) have occurred in urban areas. The prevalence of the pathogens causing these diseases relies on a complex set of interactions between the tick vector (Ixodes scapularis) and its prominent hosts, white-tailed deer (Odocoileus virginianus) and white-footed mice (Peromyscus leucopus). Additionally, predators of small mammal hosts, such as red fox (vulpes) may influence host-tick interactions. While urbanization alt
