Mapping ticks (Acari: Argasidae, Ixodidae) and informing local public action: Insights from the United Kingdom Tick Surveillance Scheme (2021-2024).

The Tick Surveillance Scheme (TSS) is a citizen science initiative which relies on public and professional submissions to monitor tick species distribution, seasonal exposure, and host associations across the United Kingdom (UK). During the period 2021-2024, 3182 tick records were received and 27 tick species were detected. The data confirm that Ixodes ricinus, the primary vector for Lyme disease and Tick-borne encephalitis, remains the most common tick in the UK. The TSS continues to detect imported tick species, underscoring the risk of novel pathogens entering the country, with new species

Doxycycline-Induced Gastrointestinal Injury for Potential Lyme Disease Exposure.

Doxycycline is an antibiotic frequently used to treat tick-borne diseases. It is known to cause esophageal injury, although gastric injury is less often described. Few cases have been reported of gastrointestinal bleeding secondary to biopsy-proven doxycycline use. In the setting of widespread doxycycline use, it is important to recognize this drug-specific pattern of injury to guide appropriate patient management. This case demonstrates doxycycline-induced gastrointestinal injury and highlights characteristic histological findings.

Unravelling the genomic landscape of Canadian Borrelia burgdorferi: a comparison across global strains.

The main agent of Lyme borreliosis (LB) in North America, the bacteria Borrelia burgdorferi, is spreading in Canada following the northward expansion of its primary tick vectors, Ixodes scapularis and Ixodes pacificus. Despite the importance of this pathogen for human health, the precise geographical origin and genome structure of Canadian B. burgdorferi strains remain to be determined, and no complete genome sequence from this region is available. The complex genome structure of Borrelia species makes their assembly challenging, but the latest long-read sequencing technologies and bioinformat

Lyme Disease: More Than a Bullseye Rash.

Lyme disease, much like the nymphs and adult ticks that transmit the disease, can be tricky and at times hard to identify. When the disease presents with hallmark symptoms, such as the erythema migrans rash with central clearing, diagnosis and treatment are often straightforward. However, especially when practicing in an endemic area, health care providers can be presented with a wide range of clinical presentations and symptoms, which can lead to variable testing and treatment recommendations based on the stage of the disease the patient presents in. Therefore, a thorough understanding of Lym

Global risk assessment of Lyme borreliosis transmission.

We analyzed the geographic risk of Lyme borreliosis taking into account the biogeography of tick vectors and wild carrier hosts, together with environmental and anthropogenic factors. Four pathogeographical scenarios were set in order to represent the contribution of vectors and hosts in the spatial zoonotic risk. For that propose, we built distribution models based on the occurrence of Lyme borreliosis cases in humans and ixodid vectors. Besides Ixodes species, we considered other ixodid ticks with potential to be vectors. These models were combined through fuzzy logic operators, according to

Comparative genomics of Borrelia lusitaniae.

Human Lyme disease is a frequent tick-borne human disease that is caused by several species in the Borrelia burgdorferi sensu lato (BBSL) clade of the bacterial spirochete genus Borrelia. Although Borrelia lusitaniae does not appear to be a major cause of this disease, it has been isolated from human patients. This Borrelia species is unusual in that its vertebrate reservoir includes reptiles in Europe and North Africa. We describe here the complete genome sequences of three B. lusitaniae isolates, PotiB2T (the species type strain) and PotiB3 that represent a Southern Portugal-North African li

Running into the Lyme-light: a retrospective cross-sectional study of tick bites and Lyme disease prevalence, incidence, and prevention in hill runners, Scotland, UK.

BACKGROUND: Lyme disease (LD) incidence is increasing globally, driven by changes to habitat and human-vector interactions. Effective prevention relies on local and individual-level understanding of risk and ecosystem interactions. Despite frequent exposure to tick habitats, little is known about tick bites and LD risk among hill, trail, and mountain runners. This study aimed to evaluate the period prevalence of tick bites and incidence of LD in hill runners in Scotland, UK and examine preventive behaviours. METHODS: A cross-sectional retrospective survey of Scottish hill runners was conducted

Lyme Disease and Health Care Costs.

IMPORTANCE: Lyme disease (LD) is the most common vector-borne illness in the US. Given the increasing prevalence and expanding geographic bounds of LD, in-depth, up-to-date understanding of costs associated with an LD diagnosis, including patient out-of-pocket (OOP) costs, is needed. OBJECTIVE: To assess health care costs in a broad US population diagnosed with LD, overall and stratified by localized disease vs disseminated disease. DESIGN, SETTING, AND PARTICIPANTS: This retrospective cohort study identified patients from Optum's deidentified Market Clarity Data (Optum Market Clarity) betwe

Late Lyme neuroborreliosis presenting with tremor and gait impairment.

A 60-year-old woman presented with a 38-month history of progressive tremor and gait impairment. Examination was consistent with a myeloradiculopathy, cerebrospinal fluid (CSF) analysis was inflammatory and imaging showed meningeal and white matter disease. Revisiting the history identified long periods spent gardening in the months before symptom onset with significant tick exposure. Lyme testing was strikingly positive in serum and CSF: a diagnosis of late Lyme neuroborreliosis was confirmed. There was clinical and radiological response to appropriate antimicrobial treatment with ongoing imp

Develop a durable, memory-driven, CspZ-targeting Lyme disease vaccine by rationale adjuvant selection.

Rational adjuvant selection is a systematic approach based on adjuvant-mediated immunomodulation to identify safe vaccine regimens that enhance protective immunity. Transmitted through ticks and caused by the bacterium Borrelia burgdorferi (Bb), Lyme disease (LD) is the most common vector-borne disease in the Northern hemisphere. There are no effective vaccines, making it suitable for testing the concept of rational adjuvant selection. Here, we formulated our previously developed and effective LD vaccine antigen, CspZ-YAC187S, with different adjuvants suitable for human use; we analyzed the im

Borrelia burgdorferi s.l. and its genetic variability in five European countries.

The Borrelia burgdorferi s.l. complex is known for its complex genome and significant genetic variability. We used multilocus sequence typing (MLST) to study the genetic variability of Borrelia in five European countries: Slovakia, Croatia, Ukraine, Finland and Bulgaria. We compared the prevalence, genospecies spectrum and genetic variability of Borrelia in 4013 questing Ixodes ricinus ticks. The highest prevalence of Borrelia was found in Slovakia (26.7 %) and Ukraine (22 %), while in Finland and Bulgaria the prevalence was found to be 12.4 % and 7.2 %, respectively. Genospecies diversity was

Skin Tone Representation of Early Lyme Disease in Medical Education Resources: Gaps and Implications for Equity.

BACKGROUND: Cutaneous Lyme disease presents differently in light versus dark skin, and delayed diagnosis can increase outcome severity. Insufficient exposure to manifestations of Lyme disease in dark skin during medical training may contribute to health inequities due to late or missed diagnoses. It remains unclear how Lyme disease, specifically, is represented in commonly used medical training materials. To inform curricula updates, we identified primary educational resources used for teaching dermatology at top-tier U.S. medical schools and assessed the representation of erythema migrans on

Gut microbiota landscape of Haemaphysalis longicornis under Borrelia burgdorferi infection.

This study aimed to explore the reason for Haemaphysalis longicornis restricting Borrelia burgdorferi colonization and transmission from the perspective of gut microbiota, and to investigate the impact of different infection statuses on the Haemaphysalis longicornis microbiota and its potential role in pathogen transmission. Pathogen-free Haemaphysalis longicornis ticks and IFNAR1-/- mice were used to establish infection models. Ticks fed on pathogen-infected mice, and their midguts were analyzed at day 4 and 10 post-feeding. DNA was extracted from the midguts, and the V3-V4 region of bacteria

Peptidoglycan architecture dictates protein interactions, tissue tropism, and arthritis in the Lyme disease spirochete Borrelia burgdorferi.

Lyme disease is a vector-borne illness transmitted by infected Ixodes spp. ticks. Dissemination of the Lyme spirochete-Borrelia burgdorferi-from the tick bite site results in a bi-phasic infection; the latter phase can cause severe musculoskeletal disease including arthritis. Lyme arthritis is an inflammatory disorder and maladaptive immune response to B. burgdorferi infection and its cellular products. One such product, which has been implicated as a key mediator of Lyme arthritis, is peptidoglycan. Peptidoglycan (PG) is a near ubiquitous feature of the bacterial cell envelope, but several ch

Xenodiagnosis to search for Borrelia burgdorferi after antibiotic treatment of Lyme disease: a prospective cohort study.

BACKGROUND: Some patients report non-specific symptoms after antibiotic therapy for Lyme disease (LD), raising questions about ongoing infection, despite no compelling evidence. We investigated whether xenodiagnosis could detect Borrelia burgdorferi in such patients, and if positive results correlated with symptoms. METHODS: Participants were adults who completed antibiotic treatment for LD 3-12 months earlier (post-therapy, n=40) or had persistent symptoms for ≥ 12 months after treatment, (post-treatment LD symptoms [PTLDS], n=20). Controls included one patient with erythema migrans (EM), on

Borrelia burgdorferi-Induced Neuroinflammation in Lyme Disease: A Potential Driver of Alzheimer's Disease Pathology?

Emerging evidence suggests that chronic infections may contribute to neurodegenerative diseases such as Alzheimer's disease (AD). One such infection is caused by Borrelia burgdorferi sensu lato (Bbsl), the spirochete complex responsible for Lyme disease, which can invade the central nervous system (CNS) and trigger Lyme neuroborreliosis (LNB). Bbsl infection is associated with persistent neuroinflammatory responses and immune evasion mechanisms, which may contribute to long-term neurological sequelae in a subset of patients. Neuroinflammation is increasingly recognized as a contributing factor

Discovery of an Adaptive Neuroimmune Response Driving Itch and Fast Tick Removal with Implications for Preventing Pathogen Transmission.

Acquired tick resistance (ATR) is well characterized in tick-exposed animals, compromising tick fitness through antibody-mediated activation of basophils. Yet, anti-tick vaccines inducing ATR have had limited success. Here, we describe a neuroimmune event preceding ATR that leads to rapid host-mediated tick removal. Tick-sensitized guinea pigs mechanically remove ticks within 3-6 h via an acquired neuroimmune-induced itch response that correlates with increased dermal expression of itch-associated genes and skin infiltration by CD3+T cells and Iba-1+ macrophages, independent of IgG and IgE ant
