One Health genomic surveillance of ticks found on community-dwelling non-human 'farm' animals to identify microbes associated with tick borne illnesses in Eastern Ontario.

Companion animals, such as dogs and cats, and wildlife are recognized as sentinel species in One Health surveillance due to periodic exposure to ticks and infection by tick-borne pathogens (TBPs). Surveillance of this type is a strategy that captures a broad range of genetic, immunological, and behavioral factors that affect tick and disease dynamics. In contrast, non-human animals (hereafter referred to as 'animals') that are traditionally farmed for their meat and other products have been largely overlooked as sentinel species. Community-dwelling farm sanctuary residents, such as cattle, pig

Atypical Lyme Neuroborreliosis Presenting With Ventriculitis and Cerebritis: A Case Report.

Lyme neuroborreliosis is a neurological manifestation of infection by Borrelia burgdorferi sensu lato and most commonly presents with lymphocytic meningitis, cranial neuropathy, or painful radiculitis. Central nervous system parenchymal involvement is uncommon, and ventriculitis is rarely reported. We describe a man in his late 70s who presented with acute confusion, dysarthria, and left-sided weakness following several days of severe lower-limb pain and progressive mobility impairment. His neurological status rapidly deteriorated, with secondary seizures requiring invasive mechanical ventilat

Barriers to entry: A participatory science study of tick identification, monitoring, and physical barriers to reduce tick bites on horses.

Equine tick-borne diseases pose increasing challenges due to climate change and expanding tick geographical ranges. With no vaccines available for Lyme disease or equine anaplasmosis in horses, tick bite prevention remains the primary control method. This two-year study evaluated tick species composition on horses in Pennsylvania and assessed the effectiveness of physical barriers for tick prevention through community-based participatory research. The study engaged horse owners as research partners. Year 1 (2022) established baseline tick surveillance across 45 participants with 60 horses, con

Rapid Motility Arrest of the Lyme Disease Spirochete, Borrelia burgdorferi, by Vaccine-Induced OspA Antibodies.

Outer surface protein A (OspA)-based Lyme disease vaccines elicit antibodies that inhibit transmission of infectious Borrelia burgdorferi spirochetes from ticks to humans, although the mechanism by which this occurs is unclear. Here we demonstrate using high-resolution, single-cell fluorescence video microscopy that B. burgdorferi motility is arrested within minutes by human OspA-specific monoclonal antibodies and polyclonal OspA antisera from mice immunized with a candidate OspA mRNA lipid nanoparticle vaccine.

When Clades Collide: Genomic Admixture in Blacklegged Ticks (Ixodes scapularis) From the Great Plains.

Ixodes scapularis ticks transmit a number of pathogens important to human health, including Borrelia burgdorferi sensu stricto, the causative agent of Lyme disease. While I. scapularis is found across the Eastern United States, Lyme disease transmission is largely limited to the Northeast and Upper Midwest and is nearly absent in the South. This indicates that differences in northern versus southern clades of I. scapularis are associated with differences in Lyme disease transmission risk. Ixodes scapularis is undergoing range expansion, including into the Great Plains region of the United Stat

The Impact of Infographics and Numerical Framing on Health Risk Perceptions and Behaviors: A Study of Ticks and Lyme Disease.

This study explores how infographic exposure and numerical framing affect perceptions of ticks and Lyme disease. Using an experimental approach, 1,001 participants viewed messages with or without infographics containing numerical data presented either as percentages or absolute values. Infographic exposure boosted susceptibility and behavioral intentions. Numerical framing influenced severity, with percentages perceived as less severe. An interaction between infographic exposure and numerical framing reduced susceptibility. Higher arithmetic aptitude lessened the effects of message variation o

Utility of combined detection of IgG/IgM/IgA vs. separate IgG and IgM antibodies to Anaplasma phagocytophilum in culture and PCR-confirmed patients with human granulocytic anaplasmosis.

Sera from 26 patients from New York State with culture and PCR confirmed human granulocytic anaplasmosis (HGA) were used to assess the performance of three different immunofluorescence assays (IFA), combined IgG/IgA/IgM and separate IgM and IgG to detect acute and/or convalescent antibodies to Anaplasma phagocytophilum. Convalescent samples were obtained within 6 weeks of the acute sample. An IFA titer ≥ 80 was considered positive. Overall, 22 acute and 21 convalescent samples were tested by all 3 IFA assays. On the acute phase serum sample, 14/22 (63.6%) were positive using the combined IgG/Ig

Host diversity and landscape structure drive genospecies-specific Lyme disease risk across Eurasia.

Lyme disease poses an escalating global health challenge partly driven by climate change and land-use transformation, while mechanistic understanding of transmission remains limited. We analyzed prevalence records from 43 Eurasian countries to map pathogen biogeography and applied theory-driven structural equation modeling (SEM) to evaluate hypothesized direct, indirect, and cascading pathways linking climate, land cover, landscape structure, and host biodiversity to pathogen prevalence. Genospecies exhibited distinct biogeographic patterns shaped by vector-host specificity, with Ixodes ricinu

Multiplex single-tier LymeSeek test improves sensitivity during the diagnostic gray zone of early Lyme disease.

Lyme disease, caused by the spirochete Borrelia burgdorferi, is the most common vector-borne disease in the U.S. and a growing public health concern due to the clinically challenging nature of the disease. Prompt antibiotic treatment early in the infection results in the best patient outcomes, while delayed treatment increases the risk of developing debilitating chronic symptoms. Because standard-of-care diagnostic tests (standardized and modified two-tier tests) have low sensitivity during the first few weeks of infection, current recommendations for diagnosing early Lyme disease rely heavily

Endosome restricted human TLR1 polymorphism highlights a role for MyD88 in Pam3CSK4 detection.

Toll-like receptor 2/1 (TLR2/1) heterodimers detect triacylated lipoproteins. Although TLR2/1 complexes localize to both the plasma membrane and endosome, the functional contributions of compartment-specific signaling are not completely defined. A common human TLR1 polymorphism (rs5743618), at which one allele substitutes isoleucine for serine adjacent to the transmembrane domain, restricts TLR1 intracellularly and is associated with an increased risk of antibiotic-refractory Lyme arthritis. This mis-localization provides a model to examine how receptor positioning influences downstream signal

Is 28 days of doxycycline still justified for canine Lyme arthropathy? A call for a randomized controlled trial.

Canine Lyme borreliosis is commonly treated with a 28-day course of doxycycline, a regimen based on historical precedent rather than evidence. Most dogs exposed to Borrelia burgdorferi remain clinically normal despite seroconversion; of those that do develop clinical Lyme disease, the most common presentation is acute or recurrent shifting-leg lameness consistent with Lyme arthropathy. This Viewpoint proposes there is clinical equipoise to evaluate shorter durations of 10 days of doxycycline in dogs with uncomplicated Lyme arthropathy and outlines a prospective parallel-group randomized contro

Comparative evaluation of viability assays and functional analysis of bbi29 in Borrelia burgdorferi.

UNLABELLED: In vitro viability assays are essential for studying the survival of Borrelia burgdorferi after exposure to stressful conditions. In contrast to the cumbersome and tedious manual microscopic counting method, fluorescence-based assays are more rapid and convenient and thus can be used in a high-throughput format. In the SYBR Green I/propidium iodide (PI) assay, spirochetes are labeled with different fluorescent dyes, and fluorescence ratios are then compared to calculate the ratio between live and dead spirochetes. Stained spirochetes are also manually counted under a fluorescence m

Efficacy of a low-dose fipronil bait in controlling blacklegged tick (Ixodes scapularis) larvae parasitizing white-footed mice (Peromyscus leucopus) under field conditions in the northeastern United States.

BACKGROUND: Lyme disease, caused primarily by Borrelia burgdorferi sensu stricto, remains the most common vector-borne disease in the United States despite control efforts. Peromyscus leucopus (white-footed mouse) is a key natural pathogen reservoir and Ixodes scapularis ticks, commonly infesting this rodent, serve as vectors for spirochetes causing human Lyme disease. Laboratory and simulated field studies demonstrated substantial efficacy of a 0.005% fipronil bait in systemically controlling I. scapularis parasitizing treated P. leucopus. To evaluate fipronil bait efficacy under natural cond

Prevalence of Lyme disease in patients with peripheral facial palsy in the province of Quebec: a prospective multicenter Canadian study.

OBJECTIVES: Lyme disease is a recognized cause of peripheral facial palsy, but its estimated prevalence in affected patients varies widely, ranging from 2 to 65%. There are no clear guidelines to assist clinicians in the empiric management of undifferentiated peripheral facial palsy in regions where Lyme disease is prevalent. This prospective multicenter study aimed to determine the prevalence of Lyme disease-associated facial palsy in patients presenting with peripheral facial palsy in four emergency departments in the most endemic regions of Quebec, Canada (Estrie and Montérégie). METHODS:

Metabolic regulation of the carbonyl stress pathway and methylglyoxal in Lyme disease and relapsing fever spirochetes.

The carbonyl stress pathway in bacteria is mediated by the enigmatic metabolite methylglyoxal and is thought to be activated in response to cellular carbohydrate and phosphate imbalance. Methylglyoxal, produced from dihydroxyacetone phosphate by methylglyoxal synthase, covalently modifies nucleophilic groups of amino acid residues, such as lysine, arginine and cysteine, to yield advanced glycation end products. In this work, we have identified several environmental signals, including glycerol and peptides, which activate carbonyl stress in the Lyme disease spirochete Borrelia (Borreliella) bur

Evaluation of capture sequencing on the Oxford Nanopore MinION as a stand-alone molecular test for tick-borne diseases.

Molecular diagnostics of tick-borne diseases (TBD) require the capacity to target a broad range of microbial pathogens circulating in ticks, along with the high sensitivity needed for detection of low burden agents such as Borrelia burgdorferi. Next generation sequencing (NGS) can have broad applicability for TBD. However, insufficient sensitivity and large infrastructure requirements limit the utility of this method for diagnostics. The application of capture-based NGS onto the Oxford Nanopore instruments presents an alternative approach that can provide high sensitivity coupled with smaller
