Geographic variation in the distribution of hard tick relapsing fever spirochete (Borrelia miyamotoi) (Spirochaetales: Spirochaetaceae) genotypes in host-seeking Ixodes scapularis (Acari: Ixodidae) nymphs and adults in the eastern United States.

Borrelia miyamotoi Fukunaga (Spirochaetales: Spirochaetaceae), the etiological agent of hard tick relapsing fever, has been detected at low prevalence in Ixodes spp. (Acari: Ixodidae) ticks from the Northern Hemisphere. Genetically distinct populations of B. miyamotoi have been described across continents and among Ixodes tick species distributed within continents. Within the eastern United States, a previous study identified two B. miyamotoi genotypes infecting Ixodes scapularis Say (Acari: Ixodidae). Here, we expand upon that work by developing a new multiplex PCR amplicon sequencing assay t

Short-Term Stability of the Fecal Microbiota in Wild Cavia tschudii During Early Captivity Transition.

This study evaluated the short-term response of the fecal microbiota of wild Cavia tschudii during its transition from natural conditions to captivity under a standardized alfalfa-based diet. Fecal samples were collected over a 21-day period and analyzed using 16S rRNA gene sequencing to assess microbial diversity, community structure, and taxonomic composition. Rarefaction analysis indicated sufficient sequencing coverage; however, samples were normalized to 500 reads per sample, and results should be interpreted cautiously. No significant changes were detected in alpha diversity, beta divers

Monospecific antibodies reveal reduction of granulins in frontotemporal dementia caused by GRN mutations.

PURPOSE: Primary open-angle glaucoma (POAG) is a common ocular disease, associated with abnormalities in aqueous humor circulation and an increase in intraocular pressure (IOP), leading to progressive optical neuropathy and loss of vision. POAG pathogenesis includes alterations of the structural properties of the sclera, especially in the optic nerve head area, contributing to the degeneration of the retinal ganglion cells. Abnormal sclera biomechanics hinder adequate compensation of IOP fluctuations, thus aggravating POAG progression. The proteomic basis of biomechanical disorders in glaucoma

Assessment of Lyme Seroconversion Among US Military Personnel in Honduras.

INTRODUCTION: Lyme disease is caused by Borrelia burgdorferi sensu lato that is transmitted through the bite of infectious ticks. Within the US active duty military component, Lyme is the most frequently reported vector-borne disease. There have not been reports on Lyme disease prevalence in Central America, but reports of travelers who contracted rickettsiosis after their trip to Honduras suggest a need for an increased tick-borne disease surveillance, including Lyme disease. The aim of this study is to determine the prevalence of Lyme disease in US military personnel deployed to Honduras. M

Hygromycin A Treatment of Borrelia burgdorferi-Infected Peromyscus leucopus Suggests Potential as a Reservoir-Targeted Antibiotic.

Lyme disease spirochetes are maintained in natural reservoirs before spilling over into human populations. Targeting these reservoirs with vaccinations or antibiotics could impact the Borrelia burgdorferi enzootic cycle and reduce the risk of human Lyme disease. In this work we report that the narrow-spectrum antibiotic hygromycin A is sufficient to disrupt B burgdorferi transmission from the main eastern US reservoir, Peromyscus leucopus, to ticks. Additionally, hygromycin A-containing baits can clear B burgdorferi from P leucopus. These studies lay the foundation for the use of hygromycin A

Leveraging Lyme Disease Surveillance to Simulate Powassan Virus Prevalence.

Powassan virus (POWV), a tick-borne flavivirus, poses a public health risk with an estimated case fatality rate of approximately 10%. Its limited surveillance results in underreporting. Using Lyme disease data as a proxy, we developed an exploratory simulation study that estimated 4649 POWV cases in the United States from 2018 to 2022, as opposed to 156 reported cases. We further estimated mortality rates (0.2%-1%), which were lower than reported rates (4.8%-21%). These discrepancies suggest that POWV cases go undetected, likely due to asymptomatic and mild to moderate presentations. Our findi

Monoclonal antibody CIS43LS sets the bar for long-acting malaria protection.

BACKGROUND: Lyme borreliosis (LB) is one of the most commonly diagnosed tick-borne diseases globally, caused by Borrelia spirochetes. Despite advances in laboratory diagnostics, both overdiagnosis and underdiagnosis of LB remain significant issues. Nonspecific symptoms such as chronic fatigue or musculoskeletal complaints are often misattributed to LB, creating controversy and delaying appropriate diagnosis and treatment. OBJECTIVE: To analyze clinical data and diagnostic results of patients referred to the Department of Infectious Diseases and Neuroinfections, Medical University of Bialystok

The BosR Is Back!

BosR is a novel nucleic acid-binding protein in the ferric uptake regulator (FUR) family that regulates gene expression in the Lyme disease spirochete Borrelia (Borreliella) burgdorferi. This issue of Molecular Microbiology contains a comprehensive transcriptomic study that keenly defines the regulatory swath of BosR in the vertebrate host of B. burgdorferi. Despite homology to Fur-like and PurR-like orthologs, BosR has traditionally been linked to regulation of RpoS, the alternative sigma factor that controls the regulon required for establishing a vertebrate infection. However, BosR regulate

Targeting of interaction between BB0323-BB0238 informs new paradigms in Lyme disease therapeutics.

Borrelia burgdorferi, one of the most prevalent tick-borne pathogens, can cause a complex and multisystem illness called Lyme disease, where there has been an unmet need for novel therapeutic or preventive strategies. We previously identified an essential protein-protein interaction (PPI) event in B. burgdorferi involving two unique proteins, BB0323 and BB0238; herein, we show that this PPI is indispensable for long-term borrelial survival in mammals and explore its potential as a novel target for small molecule therapeutics. Using X-ray crystallography, we solved the structure of the BB0238-B

Lyme disease as a rare trigger for autoimmune hemolytic anemia.

INTRODUCTION: Lyme disease, caused by Borrelia burgdorferi, is a zoonotic infection affecting the skin, nervous system, joints, and heart. Diagnosis relies on clinical history, symptoms, and a two steps serologic test confirmed by Western Blot. Although it frequently affects other systems, data on the haematological involvement spectrum of Lyme disease appears to be limited to case reports, and there are few studies that clearly demonstrate its relationship with haemolytic anaemia. This case highlights Lyme disease presenting with autoimmune hemolytic anemia (AIHA) and thrombocytopenia. CASE:

Disentangling complex disease ecology networks: Using structural equation modelling to quantify the direct and indirect effects of deer on Lyme borreliosis hazard.

Quantifying the strengths of interactions in complex vector-borne disease ecological systems is challenging. Yet, overcoming this challenge is fundamental for understanding the ecological mechanisms shaping disease hazard. Here we quantified the strengths of the hypothesised direct and indirect mechanistic pathways through which deer affect ticks and one of the Lyme borreliosis pathogens, Borrelia afzelii, by conducting a combined analysis of three previously published datasets from 39 sites across Scotland. Structural equation modelling revealed that, as predicted, deer had a strong positive

Unravelling the genomic landscape of Canadian Borrelia burgdorferi: a comparison across global strains.

The main agent of Lyme borreliosis (LB) in North America, the bacteria Borrelia burgdorferi, is spreading in Canada following the northward expansion of its primary tick vectors, Ixodes scapularis and Ixodes pacificus. Despite the importance of this pathogen for human health, the precise geographical origin and genome structure of Canadian B. burgdorferi strains remain to be determined, and no complete genome sequence from this region is available. The complex genome structure of Borrelia species makes their assembly challenging, but the latest long-read sequencing technologies and bioinformat

Lyme Disease: More Than a Bullseye Rash.

Lyme disease, much like the nymphs and adult ticks that transmit the disease, can be tricky and at times hard to identify. When the disease presents with hallmark symptoms, such as the erythema migrans rash with central clearing, diagnosis and treatment are often straightforward. However, especially when practicing in an endemic area, health care providers can be presented with a wide range of clinical presentations and symptoms, which can lead to variable testing and treatment recommendations based on the stage of the disease the patient presents in. Therefore, a thorough understanding of Lym

Global risk assessment of Lyme borreliosis transmission.

We analyzed the geographic risk of Lyme borreliosis taking into account the biogeography of tick vectors and wild carrier hosts, together with environmental and anthropogenic factors. Four pathogeographical scenarios were set in order to represent the contribution of vectors and hosts in the spatial zoonotic risk. For that propose, we built distribution models based on the occurrence of Lyme borreliosis cases in humans and ixodid vectors. Besides Ixodes species, we considered other ixodid ticks with potential to be vectors. These models were combined through fuzzy logic operators, according to

Comparative genomics of Borrelia lusitaniae.

Human Lyme disease is a frequent tick-borne human disease that is caused by several species in the Borrelia burgdorferi sensu lato (BBSL) clade of the bacterial spirochete genus Borrelia. Although Borrelia lusitaniae does not appear to be a major cause of this disease, it has been isolated from human patients. This Borrelia species is unusual in that its vertebrate reservoir includes reptiles in Europe and North Africa. We describe here the complete genome sequences of three B. lusitaniae isolates, PotiB2T (the species type strain) and PotiB3 that represent a Southern Portugal-North African li

Running into the Lyme-light: a retrospective cross-sectional study of tick bites and Lyme disease prevalence, incidence, and prevention in hill runners, Scotland, UK.

BACKGROUND: Lyme disease (LD) incidence is increasing globally, driven by changes to habitat and human-vector interactions. Effective prevention relies on local and individual-level understanding of risk and ecosystem interactions. Despite frequent exposure to tick habitats, little is known about tick bites and LD risk among hill, trail, and mountain runners. This study aimed to evaluate the period prevalence of tick bites and incidence of LD in hill runners in Scotland, UK and examine preventive behaviours. METHODS: A cross-sectional retrospective survey of Scottish hill runners was conducted

Borrelia burgdorferi s.l. and its genetic variability in five European countries.

The Borrelia burgdorferi s.l. complex is known for its complex genome and significant genetic variability. We used multilocus sequence typing (MLST) to study the genetic variability of Borrelia in five European countries: Slovakia, Croatia, Ukraine, Finland and Bulgaria. We compared the prevalence, genospecies spectrum and genetic variability of Borrelia in 4013 questing Ixodes ricinus ticks. The highest prevalence of Borrelia was found in Slovakia (26.7 %) and Ukraine (22 %), while in Finland and Bulgaria the prevalence was found to be 12.4 % and 7.2 %, respectively. Genospecies diversity was

Peptidoglycan architecture dictates protein interactions, tissue tropism, and arthritis in the Lyme disease spirochete Borrelia burgdorferi.

Lyme disease is a vector-borne illness transmitted by infected Ixodes spp. ticks. Dissemination of the Lyme spirochete-Borrelia burgdorferi-from the tick bite site results in a bi-phasic infection; the latter phase can cause severe musculoskeletal disease including arthritis. Lyme arthritis is an inflammatory disorder and maladaptive immune response to B. burgdorferi infection and its cellular products. One such product, which has been implicated as a key mediator of Lyme arthritis, is peptidoglycan. Peptidoglycan (PG) is a near ubiquitous feature of the bacterial cell envelope, but several ch
