Non-invasive ovulation tracking enables genetic engineering in wild rodents.

Many non-model rodent species are inaccessible to genetic engineering due to our limited understanding of their reproductive biology. Here, we present a low-cost, camera-based estrous-tracking technology that enables transgenesis in the white-footed mouse Peromyscus leucopus, a key reservoir for Lyme disease. We demonstrate the efficient generation of pregnant and pseudopregnant mice via timed ovulation, provide protocols for embryo generation, cultivation, microinjection, and transplantation as well as an accurate developmental timeline, and report the first engineered Peromyscus. The same te

Single-cell immunophenotyping identifies CD8+GZMK+IFNG+ T cells as a key immune population in cutaneous Lyme disease.

The skin lesion erythema migrans (EM) is the first clinical sign of Lyme disease, an infection due to the tick-transmitted bacterium Borrelia burgdorferi (Bb). Previously, we used scRNA-Seq to characterize the cutaneous immune response in the EM lesion, focusing on B cells. Here, with an expanded sample size, we profiled T cell responses in EM lesions compared to autologous uninvolved skin. In addition to CD4+ T cell subsets known to be abundant in the EM lesion, we identified clonally expanded CD8+GZMK+IFNG+ T cells that comprised cells with high or intermediate IFNG expression. These cells e

Biosensor for Lyme Disease: Real-time, Specific, and Label-Free Complementary Metal-Oxide-Silicon Transistor Sensing of Borrelia burgdorferi s.l. Outer Surface Protein A in Unprocessed Blood.

The pressing need for rapid, direct diagnostic tests for infectious diseases is exemplified by Lyme disease. Human cases of this tick-vectored bacterial infection continue to rise globally, yet the conventional laboratory diagnostic is inadequate in capturing early disease, treatment outcome, and reinfection. Direct detection of a pathogen marker from human biological fluid is a coveted strategy that has, until now, presented immense challenges due to the low circulating burden of the Lyme Borrelia pathogen. The current study demonstrates specific, label-free, quantitative, real-time sensing o

From Infection to Anxiety: A Sequential Model Linking Latent Toxoplasmosis to Psychological Distress via Health and Stress.

Identifying the drivers of chronic stress is crucial for understanding its impact on mental health. Latent toxoplasmosis, a widespread parasitic infection, has been linked to various psychological changes. The Stress-Coping Hypothesis proposes that at least some of these changes are consequences of chronic stress arising from the infection's negative impact on physical health, rather than direct parasitic manipulation. To test this mediational pathway, we surveyed 1768 individuals previously tested for toxoplasmosis or borreliosis, using the Perceived Stress Scale and the State-Trait Anxiety I

Prevalence and Risk Factors of Borrelia burgdorferi Sensu Lato IgG Antibodies Among Blood Donors in Western Romania.

Borrelia burgdorferi sensu lato is a complex of spirochetes that includes the main pathogenic species B. burgdorferi sensu stricto, B. afzelii, and B. garinii, the causative agents of Lyme disease. Our aim was to determine the seroprevalence of anti-Borrelia IgG antibodies and assess associated risk factors among blood donors from Western Romania. We conducted a cross-sectional study of 1347 consecutive donors at the Regional Blood Transfusion Center in Timisoara, Western Romania, between November and December 2018. Participants completed an epidemiological questionnaire and serum samples were

The lp17 regulatory elements in Borrelia burgdorferi: a novel small RNA impacts gene expression and mammalian infection.

UNLABELLED: The segmented genome of Borrelia burgdorferi, the tick-borne agent of Lyme disease, encodes numerous chromosomal and plasmid-borne proteins and small RNAs (sRNAs) of unknown function that are critical for infectivity. Two recent examples are the linear plasmid (lp)17-encoded protein BBD18 and sRNA SR0736 (also termed as ittA), which promote spirochete survival in ticks and mammals, respectively. Using targeted mutagenesis of the bbd18 locus, complementation, and phenotypic analysis of isogenic mutants, we herein confirm and extend the regulatory roles of BBD18 and SR0736 (ittA). A

Evaluation of platelet surface-associated immunoglobulin positivity and its association with hematologic findings and vector-borne pathogens in thrombocytopenic dogs.

BACKGROUND: Platelet surface-associated immunoglobulin (PSAIG) occurs in thrombocytopenic dogs with vector-borne diseases and immune thrombocytopenia (ITP) and may be associated with thrombocytopenia severity and inflammatory markers, including neutrophil-to-lymphocyte and platelet-to-lymphocyte ratios (NLR, PLR). HYPOTHESIS/OBJECTIVES: Assess associations between PSAIG positivity, hematologic parameters, thrombocytopenia severity, and vector-borne status in thrombocytopenic dogs. ANIMALS: Sixty-nine client-owned thrombocytopenic dogs (<200 × 103/μL) were enrolled between June 2022 and June

Demographic Predictors of Lyme Disease Knowledge Among Parents in a Highly Endemic Area.

INTRODUCTION: Lyme disease is a growing public health concern in the United States. Despite higher rates of infection among children and disseminated disease among racial minorities, few studies have investigated the role of health care inequities on tick-borne disease knowledge among parents. METHODS: We conducted a single-center, cross-sectional, anonymous survey of tick-borne disease recognition and prevention strategies among English- and Spanish-speaking parents/caregivers in a region of high Lyme disease incidence (Long Island, New York). Survey questions consisted of demographics, know

The Borreliella burgdorferi BosR-associated small non-coding RNA BasA regulates virulence.

Post-transcriptional regulation is an adaptive response used by living systems to alter protein levels independently of transcription initiation. In bacteria, one mechanism of post-transcriptional regulation involves the binding of small, non-coding RNAs (sRNAs) to target mRNAs to regulate their translation into protein. This regulation generally includes an RNA-binding protein that facilitates the sRNA::mRNA interaction. In the etiological agent of Lyme disease, Borreliella burgdorferi, over 400 temperature-dependent sRNAs have been identified. Here, we characterized the sRNA SR0735, which is

Evaluating the detection capabilities of nanopore sequencing for Borrelia burgdorferi detection in blacklegged ticks.

Ticks pose substantial threats to public health. Blacklegged ticks (Ixodes scapularis) are responsible for most tick-borne diseases in the US, transmitting seven human pathogens, including the etiological agent of Lyme disease, Borrelia burgdorferi. Molecular surveillance for tick-borne pathogens has been outpaced by their emergence, revealing a critical need to develop agnostic strategies that characterize expanding and reemerging pathogens. Oxford Nanopore Technology’s nanopore adaptive sampling (NAS), an approach that selectively enriches or depletes for target genomes or genetic loci, prov

Efficacy of Simparica and Simparica TRIO for the prevention of Borrelia burgdorferi by Ixodes scapularis.

BACKGROUND: Both Simparica® and Simparica TRIO® chewable tablets are efficacious within 12 h against existing Ixodes scapularis infestations and within 24 h against re-infestations for 1 month. It is therefore expected that treatment with either product prevents Lyme infections due to their efficacy against I. scapularis ticks before the anticipated transmission of Borrelia burgdorferi by infected ticks. METHODS: In total, four laboratory studies were conducted in which dogs were randomly allocated to two treatment groups of 10 dogs each. On day 0, dogs were either administered a placebo trea

An Fc-silent OspA monoclonal antibody passively protects mice from tick and intradermal Borrelia burgdorferi challenge.

The monoclonal antibody, LA-2, has played a pivotal role in the development of Outer surface protein A (OspA)-based vaccines for Lyme disease, a multisystem illness caused by the tick-borne spirochete, Borrelia burgdorferi sensu lato. Of particular significance was the demonstration more than three decades ago that LA-2 equivalent antibody titers, defined by a competitive-inhibition ELISA, serve as a reliable correlate of vaccine-induced protection across different species, including humans. In vitro characterization of LA-2 has identified both complement-dependent and -independent activities,

Evaluation of immunoreactive epitopes in the sera and cerebrospinal fluid of patients with post-treatment Lyme disease syndrome.

While most patients fully recover after treatment for Lyme disease with recommended antibiotic regimens, some report non-specific symptoms after treatment. When these symptoms are unexplained by other conditions and persist for ≥ 6 months, this condition is called post-treatment Lyme disease symptoms or syndrome (PTLDS). The pathogenesis of PTLDS is unknown and no specific diagnostic biomarkers have been identified. In this study, we used a high-density peptide array to examine antibody responses to > 60 primary antigens of B. burgdorferi from a cohort of patients diagnosed with PTLDS and reco

Natural genetic variation impacts complement inhibitory activity of PFam54 orthologs of Asian Borrelia bavariensis.

European and Asian populations of Borrelia (B.) bavariensis, a causative agent of Lyme borreliosis, substantially differ in their infection dynamics. This is argued to be a byproduct of the unique demographic history of B. bavariensis in relation to colonizing Europe from a highly diverse, ancestral Asian population. Whether genetic factors related to human disease could be unique traits associated with the demographic history of the European population though remains largely unclear. European B. bavariensis possesses at least two anti-complement determinants, BGA66 and BGA71 encoding by genes

A nationwide seroprevalence study of Francisella tularensis in Denmark.

BACKGROUND: Francisella tularensis is the causative agent of tularaemia. The incidence of tularaemia has increased in Europe in recent years. Due to the non-specific symptoms and diverse clinical manifestations, potential underreporting may exist. Additionally, the recent expansion in tick-borne pathogens could have increased the risk of F. tularensis transmission. We therefore aimed to estimate the seroprevalence of F. tularensis in Danish blood donors and tick-exposed individuals. METHODS: Plasma samples from healthy blood donors collected in 2022 (n=999) from all five administrative region

Antibodies against tick-borne pathogens in domestic dogs in Norway: Borrelia burgdorferi sensu lato, tick-borne encephalitis virus, and Anaplasma phagocytophilum.

BACKGROUND: Ticks are important vectors of zoonotic pathogens, posing a growing threat to animals and humans in Europe. In Norway, the tick species Ixodes ricinus is expanding its range, increasing the risk of tick-borne infections. This study aimed to estimate the seroprevalence of IgG antibodies against Borrelia burgdorferi sensu lato (s. l.), tick-borne encephalitis virus (TBEV), and Anaplasma phagocytophilum in domestic dogs residing in Norway, and to assess their potential role as sentinel species for tick-borne pathogen surveillance at the northernmost limit of I. ricinus' geographical r
