Targeting Phospholipid Metabolism as an Effective Hearing Protection Strategy.

Cochlear hair cell (HC) damage is a primary cause of sensorineural hearing loss. In this study, we performed metabolomic profiling of cochlear sensory epithelium following neomycin-induced HC injury and identified elevated arginine metabolism as a key metabolic characteristic of damaged HCs. Using a highly sensitive and specific biosensor, we confirmed that injury induced an increase in arginine levels within cochlear HCs. By manipulating the levels of arginine and its downstream metabolites, we discovered that unmetabolized arginine exerts a strong protective effect on cochlear HCs, independe

Role of Extracellular Signal-Regulated Kinase 2 in the Etiology of Tinnitus Caused by Moderate Noise Overexposure in Mice.

BACKGROUND: Acoustic trauma is a common cause of acute sensorineural hearing loss associated with tinnitus; however, the underlying molecular mechanisms remain unclear. Extracellular signal-regulated kinase 2 (ERK2), a member of the mitogen-activated protein kinase family, is crucial in cellular signaling, especially in the nervous system, where it helps regulate neuroprotection, neurogenesis, and neuronal plasticity. ERK2 is activated in the cochlea by acoustic stimuli and plays a protective role in cochlear hair cells (HCs), which are the primary sensory receptors for hearing. However, the s

Nanomodulation of blood-labyrinth barrier enhances neuroprotection and antioxidant intervention for noise-induced hearing loss.

The treatment of noise-induced hearing loss (NIHL) is challenged by the blood-labyrinth barrier (BLB), which impedes effective drug delivery to the cochlea. NIHL serves as a model for sensorineural hearing loss (SNHL), highlighting the urgent need for advanced therapeutic strategies that can traverse BLB and target cochlear pathology. Here, we report the development of tFNA-RA@G-Ang2, a multi-stage, biomimetic nanocomposite designed for targeted inner ear therapy. This platform integrates tetrahedral framework nucleic acids (tFNA) to enhance cellular internalization, gelatin nanoparticles (GNP

Effect of stimulus level on speech masking by multitalker noise in a cochlear model.

Previous research shows that intelligibility of speech in noise (SIN) with a constant speech-to-noise ratio (SNR) changes in dependence on overall stimulus level. It has been hypothesized that these changes were associated with nonlinear responses of the cochlear basilar membrane, however, the exact role of auditory periphery in this behavior is not clear. We explore this phenomenon using a nonlinear cochlear model with adjustable gain to examine how multi-talker (babble) noise affects the speech signal at the output of the auditory periphery. The amount of distortion at various intensities ca

TRPA1 deficiency affects cochlear potentials days after noise exposure.

TRPA1 channels are master sensors of tissue damage. We recently showed that, after acoustic trauma, TRPA1 activation in cochlear supporting cells regulates hearing sensitivity and is a component of the temporary threshold shift. In the neonate organ of Corti, TRPA1 activation in the Hensen's cells leads to prolonged calcium responses that propagate across the organ of Corti and cause long-lasting tissue displacements. In the adult cochlea, such tissue shape changes would be expected to affect the geometry and/or stiffness of the cochlear partition and, consequently, cochlear amplification. How

Effects of Contralateral Noise on Cortical Auditory Evoked Potential Latencies and Amplitudes.

PURPOSE: There is evidence from past animal work that the neural signal-to-noise ratio (SNR) is modulated through the action of the medial olivocochlear reflex (MOCR). This is commonly referred to as unmasking. However, evidence of unmasking in humans is limited, perhaps due to the traditional approach of measuring the MOCR using otoacoustic emissions-a preneural metric. The amplitudes and latencies of the late latency response (LLR) are sensitive to changes in SNR and may provide a means to noninvasively evaluate MOCR unmasking at the neural level. The purpose of this study was to investigate

Machine learning identification of tinnitus-related features in auditory peripheral spontaneous activity in a guinea pig noise-induced tinnitus model.

OBJECTIVES: Tinnitus affects millions globally, yet its clinical assessment relies on subjective reports, limiting diagnostic accuracy and treatment development. This study aimed to identify objective, tinnitus-related features within ensemble spontaneous activity (ESA) recorded from the cochlear surface in a guinea pig model and to evaluate their reversibility using extracochlear electrical stimulation (EES) and machine learning. DESIGN: ESA was recorded from four groups: normal controls, noise-exposed animals with tinnitus, noise-exposed animals without tinnitus, and tinnitus animals after

Development and Evaluation of a Novel Transcanal Catheter for Delivery of Hypothermia to the Inner Ear.

Mild therapeutic hypothermia (MTH) has demonstrated neuroprotective effects in the cochlea, particularly against noise-induced and electrode insertion trauma, by reducing inflammation and oxidative stress. Prior cadaveric studies have shown that localized cochlear cooling can be achieved using a probe placed on the promontory or a surface cooling device placed on the mastoid. While the effects of MTH on the vestibular system remain unstudied, its proximity and physiological similarity to the cochlea suggest potential benefits. We aimed to develop the first noninvasive, localized MTH device for

Prevention of aminoglycoside-induced outer hair cell loss by silencing CaMKKβ in an acute mouse model.

The use of ototoxic drugs remains a common cause of acquired hearing loss worldwide. Among them, aminoglycoside antibiotics are still widely used in clinical practice. Although the pathological mechanisms underlying aminoglycoside-induced hearing loss have been extensively studied, no clinically effective pharmacological treatment is currently available to prevent such ototoxicity. Our previous study demonstrated that calcium overload and activation of the Ca²⁺/calmodulin-dependent protein kinase kinase (CaMKK) pathway play a critical role in noise-induced hearing loss. In this study, we asses

Optical coherence tomography imaging demonstrates endolymphatic hydrops in the lateral and posterior semicircular canals in noise-exposed mice.

Optical Coherence Tomography (OCT) has been used to characterize cochlear endolymphatic hydrops (ELH) with distention of Reissner's membrane in mice after noise exposure. Noise exposure has been correlated with vestibular dysfunction, so we hypothesized that noise exposure can lead to ELH in the vestibular system. Little work has been performed using OCT to image the membranous labyrinth in the lateral and posterior semicircular canals (SCCs). We show that OCT with 12.5 µm axial resolution and 13.2 µm lateral resolution can image the SCCs and delineate the membranous labyrinth in anesthetized

Insights into early cochlear damage induced by potassium channel deficiency.

Hearing loss (HL) is the most common sensory disorder, caused by genetic mutations and acquired factors like presbycusis and noise exposure. A critical factor in HL development is the dysfunction of potassium (K+) channels, essential for sensory cell function in the organ of Corti (OC). Inner and outer hair cells (IHCs and OHCs) convert sound into electrical signals, while supporting cells (SCs) maintain ionic and structural balance. KCNQ4 channels, located in the basal membrane of OHCs, regulate K+ efflux. Mutations in KCNQ4 are linked to progressive HL (DFNA2), noise-induced hearing loss, an

Notched noise reveals differential improvement in the neural representation of the sound envelope.

Precise temporal coding in auditory nerve fibers is crucial for sound localization and listening in noise. However, at higher sound levels, typical of everyday listening situations, temporal coding to the stimulus envelope is poor in fibers of the on-frequency channel, i.e., those tuned to the carrier stimulus. We predict that changes in cochlear gain improve temporal coding of the stimulus envelope differentially across frequency channels. Both auditory nerve fiber recordings (in Mongolian gerbils of either sex) and human psychophysics confirm that weak temporal coding at higher levels is imp

Pathological noise exposure results in elevated levels of extracellular vesicles enriched with Hsp70 in the plasma.

Noise-induced hearing loss (NIHL) significantly impacts the quality of life for patients. Extracellular vesicles (EVs) play crucial roles in cellular communication, but it is unclear if the inner ear releases EVs into peripheral circulation under NIHL conditions as well as specific constituents. The NIHL mouse model was established by exposing mice to continuous 120 dB SPL white noise for 2 h, and hearing loss was assessed with auditory brainstem response (ABR). EVs were isolated by size exclusion chromatography (SEC) and characterized. Protein profiling in plasma EVs from NIHL mice was assess

Genetic polymorphisms in human CX3CR1-mediated macrophage dysregulation are associated with the worsening of hearing loss and cochlear degeneration after noise trauma: a study in a humanized mouse model.

Sensorineural hearing loss (SNHL) is characterized by cochlear inflammation, macrophage activation, and degeneration of hair cells, synapses, and neurons. Macrophage-mediated inflammation in the damaged cochlea is regulated via CX3CR1-CX3CL1 signaling, where the fractalkine ligand CX3CL1 serves as a chemotactic and calming signal for macrophage activation. Furthermore, disrupted CX3CR1-CX3CL1 signaling in CX3CR1-KO and CX3CL1-KO mice leads to reduced macrophage numbers, exacerbated inflammation, and loss of hair cells, ribbon synapses, and neurons in the damaged cochlea. Notably, ~ 25% of the

Strengthening Medial Olivocochlear Feedback Reduces the Developmental Impact of Early Noise Exposure.

The early onset of peripheral deafness significantly alters the proper development of the auditory system. Likewise, exposure to loud noise during early development produces a similar disruptive effect. Before hearing onset in altricial mammals, cochlear inner hair cells (IHCs) exhibit spontaneous electrical activity that drives auditory circuit development. This activity is modulated by medial olivocochlear (MOC) efferent feedback through α9α10 nicotinic cholinergic receptors in IHCs. In adults, these receptors are restricted to outer hair cells, where they mediate MOC feedback to regulate co

Analysis of stress-induced hearing loss in repeated cold stress-exposed mice.

Sudden hearing loss is an acute sensorineural hearing loss of unknown etiology. Owing to the significant increase in the number of patients and low efficacy of treatment, elucidation of its pathogenesis is urgently required. An animal model for hearing loss is needed; however, mainstream models, such as noise exposure and drug administration, cause irreversible damage to the inner ear tissue and therefore cannot be said to reflect the pathology of sudden hearing loss, which can sometimes be cured spontaneously. In this study, we prepared a mouse model of hearing loss caused by mental or physic

Effect of liraglutide treatment on mitigation of hearing damage induced by multiday repeated high-intensity blasts.

Exposures to blasts impair hearing, despite the protection of earplugs. Liraglutide showed therapeutic effects against hearing damage induced by single-day blasts. However, the effect of liraglutide on auditory damage caused by multiday repeated blasts remains unknown. Chinchillas protected by earplugs were exposed to three blasts at 15-25 psi on day 1, followed by an identical exposure on day 4. Animals were assigned to pre-blast treatment, post-blast treatment, or blast control groups based on the timing of liraglutide administration relative to the day 1 blasts. Auditory brainstem response

Investigation of the protective and therapeutic effects of coenzyme Q10 on acoustic trauma-induced hearing loss in rats.

BACKGROUND: Acoustic trauma is a process in which the sound energy coming into the ear is so high that it causes hearing loss. Although various treatments have entered clinical practice today, studies on the prevention and treatment of acoustic trauma continue. In our study, we investigated the protective and therapeutic effects of CoQ10 in rats with hearing loss due to acoustic trauma. MATERIALS AND METHOD: 35 Wistar-type Albino rats were used in the study. The rats were randomly divided into five equal groups. The groups are the acoustic trauma group (group 1), the ethanol (solvent of CoQ10
