Associations between physiological indicators of cochlear deafferentation and listening effort in military Veterans with normal audiograms.

Cochlear synaptopathy, a type of cochlear deafferentation that impacts the synapses between the inner hair cells and the afferent auditory nerve fibers, is predicted to result in speech perception in noise difficulty. However, this has been difficult to confirm due to mixed findings in human studies of the relationship between speech perception in noise and physiological indicators of deafferentation (auditory brainstem response [ABR] wave I amplitude, envelope following response [EFR] magnitude, and middle ear muscle reflex [MEMR] magnitude). One possible explanation for the mixed findings is

Slight and hidden hearing loss differentially affect short- and long-term memory in young rats.

Mild forms of hearing loss (HL) have been linked to cognitive impairments in children, yet the neurobiological mechanisms underlying this connection remain unclear. Existing research using animal models mostly focuses on more severe levels of HL or investigates only limited aspects of cognition. To gain a broader understanding of how slight/hidden HL affects cognitive behaviors, we induced HL in 4-week-old Wistar rats through noise exposure. Auditory brainstem response measurements confirmed slight and hidden HL, but this auditory impairment did not alter the density of inner hair cells or the

Age-Related Decline in Neural Phase-Locking to Envelope and Temporal Fine Structure Revealed by Frequency Following Responses: A Potential Signature of Cochlear Synaptopathy Impairing Speech Intelligibility.

PURPOSE: Assessing the contribution of cochlear synaptopathy (CS) to the variability in speech-in-noise intelligibility among individuals remains a challenge. While several studies have proposed biomarkers for CS based on neural phase-locking to the temporal envelope (ENV), fewer have investigated how CS affects the coding of temporal fine structure (TFS), despite its crucial role in speech-in-noise perception. In this study, we specifically examined whether TFS-based markers of CS could be derived from electrophysiological responses and psychophysical detection thresholds of spectral modulati

[The damaging effects of two exposures to 100 dB white noise on myelination of the auditory nerve in mouse cochleae mechanism of repeated noise exposure on myelin sheath of cochlear auditory nerve in mice].

Objective:This study aims to investigate the mechanism and potential effects of two exposures to 100 dB sound pressure level(SPL) broadband white noise, with a 14-days interval, on the myelin sheath of the cochlear auditory nerve in mice. The research provides experimental evidence for understanding the pathophysiological processes of noise-induced hearing loss and hidden hearing loss. Methods:Fifteen 6-week-old male C57BL/6J mice with normal hearing thresholds were randomly divided into three groups: a control group(no noise exposure), a single noise exposure group, and a double noise exposur

Clonidine mitigates noise-induced hearing loss by regulating TRPC6-mediated calcium influx in cochlear hair cells.

Noise-induced hearing loss (NIHL) is a common auditory disorder driven by calcium overload, oxidative stress, and apoptosis in cochlear sensory hair cells. The transient receptor potential canonical 6 (TRPC6), a nonselective cation channel that can be activated by norepinephrine, is implicated in calcium influx and associated cellular damage. This study explores the protective effects of clonidine, an FDA-approved α2-adrenergic receptor agonist that reduces sympathetic nervous system activity and norepinephrine release, on NIHL in mice. Clonidine treatment significantly preserved hearing thres

Insulin-like growth factor 1 protects cochlear outer hair cells against cisplatin.

Cisplatin, an effective anti-neoplastic drug widely used in oncology protocols, has an adverse effect such as ototoxicity, for which no current treatment exists. Histological lesions in the cochlea after cisplatin administration are most prominent in outer hair cells in the organ of Corti. We have previously reported that insulin-like growth factor 1 (IGF1) protects cochlear hair cells (HCs) against several types of damage to the cochlea, including noise exposure, ischemia, surgical trauma, and aminoglycoside, resulting in hearing recovery. In the present study, we investigated the efficacy of

Galvanic vestibular stimulation for Mal de Debarquement syndrome: a pilot study on therapeutic potential.

Mal de Debarquement Syndrome (MdDS) is a debilitating neuro-otological disorder causing a persistent sensation of self-motion, often triggered by passive motion like being on a boat (MT-MdDS) or non-motion triggered (non-MT MdDS). Due to the unknown pathophysiological mechanism, available treatment options for managing symptoms are limited. Within the framework of a pilot study, our objective was to investigate the effect, safety, and feasibility of galvanic vestibular stimulation (GVS) as a potential alternative treatment option for MdDS. The trial included 12 MdDS patients and 12 controls. I

Is Noise Exposure Associated With Impaired Extended High Frequency Hearing Despite a Normal Audiogram? A Systematic Review and Meta-Analysis.

Understanding the initial signature of noise-induced auditory damage remains a significant priority. Animal models suggest the cochlear base is particularly vulnerable to noise, raising the possibility that early-stage noise exposure could be linked to basal cochlear dysfunction, even when thresholds at 0.25-8 kHz are normal. To investigate this in humans, we conducted a meta-analysis following a systematic review, examining the association between noise exposure and hearing in frequencies from 9 to 20 kHz as a marker for basal cochlear dysfunction. Systematic review and meta-analysis followed

Echolocation frequency alteration, and hearing loss induced by an ototoxic drug in the echolocating bat Hipposideros pratti.

Noise exposure increases the level of reactive oxygen species (ROS) in the outer hair cells (OHCs) of the cochlea, which subsequently damages OHCs and causes noise-induced hearing loss. However, increasing evidences have shown that echolocating bats can maintain their auditory sensitivity after intense noise exposure, indicating that they have a strong capacity to clear ROS, or that noise exposure does not increase ROS levels in the cochlea of the animals. To differentiate between these possibilities, the constant frequency-frequency modulation bats, Hipposideros pratti, were intraperitoneally

Cochlear neural contributions to triple network changes in tinnitus, hyperacusis & misophonia? A perspective review.

What do tinnitus, the perception of sounds without an internal or external source of noise, hyperacusis, the pathological hypersensitivity to noise, or misophonia, an intolerance to certain everyday noises, have in common, and what differentiates them? A large number of excellent studies focused in the last few decades on identifying the neural correlates of tinnitus, hyperacusis, or misophonia on the basis of central triple-network changes. In this perspective review we explicitly examine, possible differential and causal involvement of peripheral components as a presumptive trigger that may

Development of audiometric parameters throughout the lifespan. I: Auditory data.

Age-related hearing loss (presbycusis) is a nonsyndromic sensorineural condition that affects the inner ear, auditory pathway, and related cortical regions to varying degrees, typically beginning around age 65. However, hearing can start to deteriorate as early as age 18 and can be negatively influenced by several external factors throughout the lifespan. Depending on the location and severity of the pathology, different aspects of hearing may be affected, resulting in various phenotypes of hearing loss. The aim of this study is to determine average data for a complex battery of auditory tests

Impact of Different Occupational Noises on Static and Dynamic Postural Stability in Healthy Young Adults.

BACKGROUND: Sounds that cause disturbances and perturbations to the vestibular (inner ear organ responses) and visual (acute oculomotor responses) systems can impact postural stability. The purpose of this study was to assess the impact of different types of sounds and noises on both static and dynamic PS. METHODS: A total of 20 participants (12 females and 8 males; age: 21.35 ± 1.79 years; height: 170.7 ± 9.3 cm; mass: 66.725 ± 14.1 kg) were tested using the limits of stability (LOS) test on the BTrackS™ balance plate and a Timed Up and Go (TUG) test, when exposed to four different sounds an

Prevention and treatment of noise-induced hearing loss and cochlear synapse degeneration by potassium channel blockers in vivo.

Noise can induce hearing loss. In particular, noise can induce cochlear synapse degeneration leading to hidden hearing loss, which is the most common type of hearing disorders in the clinic. Currently, there is no pharmacological treatment, especially no post-exposure (i.e., therapeutic) treatment available in the clinic. Here, we report that systemic administration of K+ channel blockers by intraperitoneal injection before or after noise exposure significantly ameliorated noise-induced hearing loss (NIHL) and cochlear synapse degeneration. After systemic administration of a general K+-channel

A Single Dose of AC102 Reverts Tinnitus by Restoring Ribbon Synapses in Noise-Exposed Mongolian Gerbils.

A single intratympanic application of the small-molecule drug AC102 was previously shown to promote significant recovery of hearing thresholds in a noise-induced hearing loss model in guinea pigs. Here, we report the effects of AC102 to revert synaptopathy of inner hair cells (IHCs) and behavioral signs of tinnitus in Mongolian gerbils following mild noise trauma. This experimental protocol led to minor hearing threshold shifts with no loss of auditory hair cells (HCs) but induced synaptopathy and a sustained and significant tinnitus percept. Treatment by intratympanic application of AC102 was

Elevated lead levels as a risk factor for pediatric sensorineural hearing loss.

INTRODUCTION: Despite legislation banning the use of lead paint in the United States in 1978, pediatric lead exposure still occurs. In 2021, the CDC reduced the reference value for blood lead levels to 3.5ug/dl noting even low levels of lead can be harmful. The neurologic consequences of pediatric lead exposure are well documented. Few studies have linked lead exposure to hearing loss in children. OBJECTIVES: To identify a relationship between pediatric lead exposure and sensorineural hearing loss. METHODS: The TriNetx Analytics Network, a federated health research network that aggregates th

Electrophysiological Effects of Intratympanic Retinoic Acid Application Following Acoustic Trauma in Rats.

BACKGROUND: Retinoic acid (RA), the active metabolite of vitamin A, is essential for the embryonic development of the inner ear, involving processes such as cell proliferation, differentiation, and morphogenesis. It also plays a crucial role in the protection and maintenance of the inner ear during the postnatal period. This study investigated the electrophysiological effects of early intratympanic RA administration following acoustic trauma. METHODS: Two doses of intratympanic RA injections were administered to 21 Sprague-Dawley rats after acoustic trauma, and the distortion product otoacous

Exploring AAV-Mediated Gene Therapy for Inner Ear Diseases: from Preclinical Success to Clinical Potential.

Hearing loss imposes a significant global health burden and often results from genetic factors and various external influences, such as noise exposure and the use of ototoxic drugs. Recent advancements in adeno-associated virus (AAV)-mediated gene therapy offer promising new avenues for the treatment of inner ear diseases. Clinical trials of AAV-mediated gene therapies show impressive preliminary results, although further research is needed to evaluate the safety and long-term effects of these therapies. Preclinical AAV-mediated gene therapy is notable for its high transduction efficiency and

NOX2 Contributes to High-Frequency Outer Hair Cell Vulnerability in the Cochlea.

Most patients with sensorineural hearing loss (SNHL) may initially experience high-frequency hearing loss linked to greater vulnerability of outer hair cells (OHCs) in the cochlear basal turn. However, the molecular mechanism underlying this susceptibility in the high-frequency region remains unclear. Here, NOX2 is first identified as a differentially expressed gene related to oxidative damage in the apical and basal turns through single-cell RNA sequencing. In mouse cochleae damaged by neomycin and noise, Nox2-positive OHCs increase progressively from the apex to base, indicating that NOX2 ex

BDNF Alleviates Noise-Induced Cochlear Synaptopathy Through Inhibition of Autophagy.

Brain-derived neurotrophic factor (BDNF) can protect against metabolic stress-induced synaptic damage via inhibiting autophagy in the brain. It was further proposed to prevent cochlear synaptopathy against the stress of noise exposure (NE). However, it remains elusive whether BDNF's protective role in cochlear synaptopathy is associated with the inhibition of autophagy in the cochlea. In this study, we found significantly increased expression of C-terminal-Binding Protein 2 (CtBP2) and Glutamate Receptor 2 (GluR2), accompanied by greatly reduced expression of microtubule-associated Protein 1 L
