Senolytic treatment alleviates cochlear senescence and delays age-related hearing loss in C57BL/6J mice.

BACKGROUND: Age-related hearing loss (ARHL) is a prevalent sensory deficit that significantly affects quality of life in older individuals. Cellular senescence contributes to various age-related degenerative disorders. However, its effect on ARHL remains unclear. PURPOSE: The aim of this study was to explore the therapeutic potential of senolytics in attenuating cochlear senescence and delaying the progression of ARHL. METHODS: The senolytic drugs dasatinib and quercetin (D + Q) were used to target senescent cells at different stages of ARHL in C57BL/6J mice. The impact of D + Q treatment on

Genetic and Environmental Contributions to Age-Related Hearing Loss: Results from a Longitudinal Twin Study.

Over 430 million people worldwide experience disabling hearing loss, a condition that becomes more prevalent with age. Although the genetic component to hearing loss has been well established, there has been less data available regarding changes in the genetic contributions to hearing loss over time. We report the pure tone hearing thresholds across 500, 1,000, 2,000, 4,000, and 8,000 Hz from over 1,000 male twins comprising monozygotic (MZ) and dizygotic (DZ) pairs sampled from the United States-based Vietnam Era Twin Study of Aging (VETSA). Twins were tested during three waves, at an average

Obesity affects the association between Serum Klotho and age-related hearing loss: A cross-sectional study.

OBJECTIVE: The influence of obesity on the interplay between Klotho and age-related hearing loss (ARHL) requires further exploration. This study investigates the impact of obesity on the correlation between Serum Klotho levels and ARHL prevalence in a large cohort. METHODS: The present study analyzed data from the National Health and Nutrition Examination Survey (NHANES) involving 3355 participants. Participants were stratified based on body mass index (BMI) into non-obese (BMI < 30 kg/m²) and obese (BMI ≥ 30 kg/m²) groups. We then assessed the interaction between serum Klotho levels and the

Can Mismatch Negativity Be Used as an Indicator to Predict Central Auditory Deficits in Individuals with Normal Hearing?

Background/Objectives: In the early stage of presbycusis, patients experience reduced speech perception in noisy environments despite normal audiometry, normally known as hidden hearing loss. Diagnostic indicators like the reduced amplitude of ABR wave I, elevated extended high-frequency threshold (EHT), and decreased middle ear muscle reflex (MEMR) amplitude aim to identify biomarkers of peripheral auditory pathology but remain inconsistent. Mismatch negativity (MMN) is a cortical auditory evoked potential generated when the brain detects sound changes. This study aimed to assess MMN as a dia

Anodal direct current stimulation of the auditory cortex at the onset of presbycusis delays cortical aging.

Presbycusis or age-related hearing loss (ARHL) affects millions of people worldwide, increasing their risk of cognitive decline and poor quality of life. However, ARHL remains an irreversible condition due to our inability to induce inner-ear hair cell regeneration. Nevertheless, multisession epidural stimulation of the auditory cortex (AC) at the onset of ARHL prevents hearing threshold elevation in naturally aging Wistar rats. Accordingly, we hypothesized that anodal direct current (DC) stimulation of the AC may also compensate for age-related maladaptive, activity-dependent changes. Here, w

Generation and characterization of a humanized GJB2 p.V37I knock-in mouse model for studying age-related hearing loss.

Age-related hearing loss (ARHL) has been closely linked to genetic factors, with studies identifying the p.V37I mutation in the GJB2 gene as a potential contributor to ARHL. To investigate this, we generated a humanized p.V37I mutant mouse model and performed auditory brainstem response (ABR) testing, cochlear morphology assessments, and transcriptional sequence of mutant and wild-type (WT) mice at different ages. Our results indicated that this kind of GJB2 mutation does not lead to cochlear developmental abnormalities, and aging mutant mice exhibit only mild hearing loss compared to WT mice,

Machine Learning Models Can Predict Tinnitus and Noise-Induced Hearing Loss.

OBJECTIVES: Despite the extensive use of machine learning (ML) models in health sciences for outcome prediction and condition classification, their application in differentiating various types of auditory disorders remains limited. This study aimed to address this gap by evaluating the efficacy of five ML models in distinguishing (a) individuals with tinnitus from those without tinnitus and (b) noise-induced hearing loss (NIHL) from age-related hearing loss (ARHL). DESIGN: We used data from a cross-sectional study of the Canadian population, which included audiologic and demographic informati

Conditional Overexpression of Neuritin in Supporting Cell Protects Cochlear Hair Cell and Delays Age-Related Hearing Loss by Enhancing Autophagy.

Age-related hearing loss (ARHL) is a highly prevalent, burdensome sensorineural hearing loss closely associated with impaired autophagic influx. Our previous studies revealed that neuritin, a neurotrophic factor primarily expressed in the central nervous system, could alleviate drug-induced damages in hair cells (HCs) and spiral ganglion neurons. However, its effects on ARHL and whether these effects are closely related to autophagy remain unclear. Using the Nrn1 knock-in mice and cultured cochlear basilar membrane (CBM) of the neonatal mouse, we show that neuritin could restore aging-associat

Presbycusis: Pathology, Signal Pathways, and Therapeutic Strategy.

Presbycusis, also known as age-related hearing loss (ARHL), is a progressive auditory impairment and ranks among the most prevalent sensory disorders in the elderly population. It is primarily caused by damage to hair cells, degeneration of spiral ganglion neurons, and atrophy of the stria vascularis, which are integral components of the cochlea. While extensive research has been devoted to hair cells and spiral ganglion neurons, this review focuses on the critical role of the stria vascularis in ARHL. The primary function of the stria vascularis is to maintain the endocochlear potential, whic

Cognitive benefits of hearing intervention vary by risk of cognitive decline: A secondary analysis of the ACHIEVE trial.

INTRODUCTION: Results from the Aging and Cognitive Health Evaluation in Elders (ACHIEVE) trial suggest hearing intervention may not reduce 3-year cognitive decline in all older adults with hearing loss but may be beneficial in certain groups. This secondary analysis investigated if participants with multiple risk factors for cognitive decline received greater benefits. METHODS: We used a sample of dementia-free participants (N = 2692) from the Atherosclerosis Risk in Communities (ARIC) cohort to develop a predictive model for cognitive decline. The model was applied to baseline measures of AC

mRNA metabolism regulator human antigen R (HuR) regulates age-related hearing loss in aged mice.

Age-related hearing loss (ARHL) is among the most prevalent and complex disorders in older adults. However, the pathogenesis of ARHL remains poorly understood. Using a single-cell transcriptomic landscape of mouse cochlea at five time points (1, 2, 5, 12 and 15 months), we found that the levels of human antigen R (HuR)-a classical RNA-binding protein-increase with age. Here we show that HuR is specifically transported from the nucleus to the cytoplasm in hair cells in both aging mice and nonhuman primates. HuR overexpression in cochlea could successfully alleviate ARHL in aged mice. Meanwhile,

Genome-Wide Association Study of Age-Related Hearing Loss in CFW Mice Identifies Multiple Genes and Loci, Including Prkag2.

PURPOSE: Age-related hearing loss (ARHL) is one of the most prevalent conditions affecting the elderly. ARHL is influenced by a combination of environmental and genetic factors; the identification of the genes that confer risk will aid in the prevention and treatment of ARHL. The mouse and human inner ears are functionally and genetically homologous. We used Carworth Farms White (CFW) mice to study the genetic basis of ARHL because they are genetically diverse and exhibit variability in the age of onset and severity of ARHL. METHODS: Hearing at a range of frequencies was measured using audito

Loss of microstructural integrity in left hemispheric white matter tracts is associated with poorer digits in noise understanding.

Age-related hearing loss (ARHL) is a prevalent condition among older adults and is regarded as a potentially modifiable risk factor for dementia. Although multiple studies have investigated pure-tone thresholds as a measure for ARHL and its relationship to dementia, the potential role of the central auditory system has received little attention. To address this gap in the literature, this study investigates the relationship between central auditory functioning, assessed using the speech-reception-threshold (SRT) of the digits-in-noise (DIN) test, and the microstructural integrity of white matt

Whole-Genome DNA Methylation Analysis in Age-Related Hearing Loss.

BACKGROUND: Presbycusis, also known as age-related hearing loss (ARHL), is the most frequent sensory disability affecting elderly adults worldwide. ARHL is characterized by bilateral, progressive, sensorineural hearing loss that is more pronounced at a high frequency. Conventional factors associated with ARHL include diabetes, hypertension, and a family history of hearing loss. The severity of hearing impairment varies between individuals. The defined causative molecular pathogenesis for ARHL is unknown, thus the identification of underlying pathogenic mechanisms involved in ARHL is imperative

Trends and driving factors of age-related hearing loss and severity over 30 years: a cross-sectional study.

BACKGROUND: Age-related hearing loss (AHL) is a modifiable risk factor for chronic disability and cognitive decline in adults over 60 years globally. Despite its preventable nature, long-term trends (1992-2021) in AHL burden and its demographic, socioeconomic, occupational noise exposures, and geographic drivers remain underexplored, limiting targeted intervention strategies. METHODS: This observational study analyzed age-standardized prevalence and years lived with disability (YLDs) for AHL among adults aged ≥60 years using the Global Burden of Disease Study 2021 (1992-2021). Data were strat

Age-Related Hearing Loss: Evidence-Based Strategies for Early Detection and Management.

Age-related hearing loss (ARHL) is common in older adults and has been linked to significant health and social challenges. These challenges include cognitive decline, depression, falls, and overall decreased quality of life. Despite its high prevalence, ARHL remains underdiagnosed and undertreated, partly due to its gradual onset, stigma, and lack of standardized screening and management protocols. This article provides a comprehensive overview of evidence-based strategies for the early detection and management of ARHL, with an emphasis on recent clinical practice guidelines where nurses are i

Identifying a gene signature for age-related hearing loss through machine learning and revealing the effect of the CTSS on the mice cochlea.

Age-related hearing loss (ARHL) is one of the most common health conditions among the elderly population. This study used machine learning to screen for a gene signature to predicts ARHL. Four ARHL mice cochlear transcriptome datasets and the mRNA sequencing of C57BL/6J mice were used for analysis. Machine learning was used to screen for gene signatures closely related to ARHL and validate them. Via qPCR, immunohistochemistry, and immunofluorescence confocal microscopy were used to assess the effect of key gene on the cochlea. The gene signature consisting of 38 genes constructed via Stepglm [

Age-related differences in GABAergic synapses across the central inferior colliculus in the Fischer Brown Norway rat.

Presbycusis, one of the most widespread disorders, is in part associated with the loss of temporal precision within the central auditory system. A contributor to the dysfunctional temporal precision during aging is the substantial downregulation of GABA in the central inferior colliculus (ICc), the hub of ascending and descending inputs of the auditory midbrain. However, how GABAergic inputs across the tonotopic axis of the ICc change with age has not been well explored. We sought to determine age-related changes to GABAergic synapses in the lemniscal ICc, and if changes are uniform across the
